Reaction of C70 with ten equivalents of silver(I) trifluoroacetate at 320-340 degrees C followed by fractional sublimation at 420-540 degrees C and HPLC processing led to the isolation of a single abundant isomer of C70(CF3)n for n = 2, 4, 6, and 10, and two abundant isomers of C70(CF3)8. These six compounds were characterized by using matrix-assisted laser desorption ionization (MALDI) mass spectrometry, 2D-COSY and/or 1D 19F NMR spectroscopy, and quantum-chemical calculations at the density functional theory (DFT) level. Some were also characterized by Raman spectroscopy. The addition patterns for the isolated compounds were unambiguously found to be C1-7,24-C70(CF3)2, C1-7,24,44,47-C70(CF3)4, C2-1,4,11,19,31,41-C70(CF3)6, Cs-1,4,11,19,31,41,51,64-C70(CF3)8, C2-1,4,11,19,31,41,51,60-C70(CF3)8, and C1-1,4,10,19,25,41,49,60,66,69-C70(CF3)10 (IUPAC numbering). Except for the last compound, which is identical to the recently reported, crystallographically characterized C70(CF3)10 derivative prepared by a different synthetic route, these compounds have not previously been shown to have the indicated addition patterns. The largest relative yield under an optimized set of reaction conditions was for the Cs isomer of C70(CF3)8 (ca. 30 mol % of the sublimed mixture of products based on HPLC integration). The results demonstrate that thermally stable C70(CF3)n isomers tend to have their CF3 groups arranged on isolated para-C6(CF3)2 hexagons and/or on a ribbon of edge-sharing meta- and/or para-C6(CF3)2 hexagons. For Cs- and C2-C70(CF3)8 and for C2-C70(CF3)6, the ribbons straddle the C70 equatorial belt; for C1-C70(CF3)4, the para-meta-para ribbon includes three polar hexagons; for C1-7,24-C70(CF3)2, the para-C6(CF3)2 hexagon includes one of the carbon atoms on a C70 polar pentagon. The 10.3-16.2 Hz 7JF,F NMR coupling constants for the end-of-ribbon CF3 groups, which are always para to their nearest-neighbor CF3 group, are consistent with through-space Fermi-contact interactions between the fluorine atoms of proximate, rapidly rotating CF3 groups.  相似文献

Complexes of cerium(III), lanthanum(III) and neodymium(III) with coumarin‐3‐carboxylic acid (HCCA) were synthesized by mixing of equimolar amounts of the respective metal nitrates and coumarin‐3‐carboxylic acid in ethanol. The complexes were characterized and identified by elemental analysis, IR and Raman spectroscopy. DTA and TGA were applied to study the compositions of the compounds. The vibrational study showed bidentate coordination of CCA? to Ln(III) ions through the carbonyl oxygen and the carboxylic oxygen atoms. The newly synthesized compounds were assayed for cytotoxicity against SKW‐3, HL‐60 and Reh cells. The complexes of cerium(III) and lanthanum(III) showed marginal cytotoxic activity against SKW‐3 and Reh cells as compared with the inorganic salts at concentration 200 µM . The complex of neodymium(III) induced approximately 50% reduction of the survival HL‐60 and SKW‐3 cells at concentration 200 µM . Copyright © 2007 John Wiley & Sons, Ltd.  相似文献

Elucidation of physicochemical mechanisms of enzymatic processes is one of the main tasks of modern biology. High efficiency and selectivity of enzymatic catalysis are mostly ensured by conformational dynamics of enzymes and substrates. Here, we applied a stopped-flow kinetic analysis based on fluorescent spectroscopy to investigate mechanisms of conformational transformations during the removal of alkylated bases from DNA by ALKBH2, a human homolog of Escherichia coli AlkB dioxygenase. This enzyme protects genomic DNA against various alkyl lesions through a sophisticated catalytic mechanism supported by a cofactor (Fe(II)), a cosubstrate (2-oxoglutarate), and O2. We present here a comparative study of conformational dynamics in complexes of the ALKBH2 protein with double-stranded DNA substrates containing N1-methyladenine, N3-methylcytosine, or 1,N6-ethenoadenine. By means of fluorescent labels of different types, simultaneous detection of conformational transitions in the protein globule and DNA substrate molecule was performed. Fitting of the kinetic curves by a nonlinear-regression method yielded a molecular mechanism and rate constants of its individual steps. The results shed light on overall conformational dynamics of ALKBH2 and damaged DNA during the catalytic cycle.  相似文献

We consider the problems of the authorship of literary texts in the framework of the quantitative study of literature. This article proposes a methodology for authorship attribution of literary texts based on the use of data compressors. Unlike other methods, the suggested one gives a possibility to make statistically verified results. This method is used to solve two problems of attribution in Russian literature.  相似文献

A number of novel heteroligand Zn(II) complexes (1–8) of the general type (Ln)Zn(NN) containing O,N,O′-, O,N,S-donor redox-active Schiff bases and neutral N,N′-chelating ligands (NN) were synthesized. The target Schiff bases LnH2 were obtained as a result of the condensation of 3,5-di-tert-butyl-2-hydroxybenzaldehyde with substituted o-aminophenols or o-aminothiophenol. These ligands with combination with 2,2′-bipyridine, 1,10-phenanthroline, and neocuproine are able to form stable complexes upon coordination with zinc(II) ion. The molecular structures of complexes 4∙H2O, 6, and 8 in crystal state were determined by means of single-crystal X-ray analysis. In the prepared complexes, the redox-active Schiff bases are in the form of doubly deprotonated dianions and act as chelating tridentate ligands. Complexes 6 and 8 possess a strongly distorted pentacoordinate geometry while 4∙H2O is hexacoordinate and contains water molecule coordinated to the central zinc atom. The electrochemical properties of zinc(II) complexes were studied by the cyclic voltammetry. For the studied complexes, O,N,O′- or O,N,S-donor Schiff base ligands are predominantly involved in electrochemical transformations in the anodic region, while the N,N′-coordinated neutral nitrogen donor ligands demonstrate the electrochemical activity in the cathode potential range. A feature of complexes 5 and 8 with sterically hindered tert-butyl groups is the possibility of the formation of relatively stable monocation and monoanion forms under electrochemical conditions. The values of the energy gap between the boundary redox orbitals were determined by electrochemical and spectral methods. The parameters obtained in the first case vary from 1.97 to 2.42 eV, while the optical bang gap reaches 2.87 eV.  相似文献

One of the powerful antioxidants used clinically is Edaravone (EDA). We synthesized a series of new EDA analogs, 4-aminopyrazol-5-ol hydrochlorides, including polyfluoroalkyl derivatives, via the reduction of 4-hydroxyiminopyrazol-5-ones. The primary antioxidant activity of the compounds in comparison with EDA was investigated in vitro using ABTS, FRAP, and ORAC tests. In all tests, 4-Amino-3-pyrazol-5-ols were effective. The lead compound, 4-amino-3-methyl-1-phenylpyrazol-5-ol hydrochloride (APH), showed the following activities: ABTS, 0.93 TEAC; FRAP, 0.98 TE; and ORAC, 4.39 TE. APH and its NH-analog were not cytotoxic against cultured normal human fibroblasts even at 100 μM, in contrast to EDA. According to QM calculations, 4-aminopyrazolols were characterized by lower gaps, IP, and η compared to 4-hydroxyiminopyrazol-5-ones, consistent with their higher antioxidant activities in ABTS and FRAP tests, realized by the SET mechanism. The radical-scavenging action evaluated in the ORAC test occurred by the HAT mechanism through OH bond breaking in all compounds, directly dependent on the dissociation energy of the OH bond. All the studied compounds demonstrated the absence of anticholinesterase activity and moderate inhibition of CES by some 4-aminopyrazolols. Thus, the lead compound APH was found to be a good antioxidant with the potential to be developed as a novel therapeutic drug candidate in the treatment of diseases associated with oxidative stress.  相似文献

α-Functionalized α,β-unsaturated aldehydes is an important class of compounds, which are widely used in fine organic synthesis, biology, medicine and pharmacology, chemical industry, and agriculture. Some of the 2-substituted 2-alkenals are found to be the key metabolites in plant and animal cells. Therefore, the development of efficient methods for their synthesis attracts the attention of organic chemists. This review focusses on the recent advances in the synthesis of 2-functionally substituted 2-alkenals. The kinetics of air copper slag oxidation under nonisothermal conditions is studied using simultaneous TG–DTA at a varying heating rate of slag and flow rate of the oxidizing gas flux. The values of the kinetic parameters, activation energy and pre-exponential factor, have been determined based on: data from DTA by the methods of Kissinger and Ozawa; data from TG using an isoconversion method and the computation procedures of Ozawa–Flynn–Wall and Kissinger–Akahira–Sunose. No relationship between the kinetic parameters and the oxidation gas flow rate has been established. The changes of the phase composition with temperature are investigated by X-ray powder diffraction analysis on the basis of data obtained for the products formed at the different stages of the oxidation process. The morphology of the oxidized slag as well as the elements distribution is studied by electron microscopy and EDS analysis.  相似文献

A bioluminescent assay based on a system of coupled enzymatic reactions catalyzed by bacterial luciferase and NADH:FMN‐oxidoreductase was developed to monitor toxicity and antioxidant activity of bioactive compounds. The assay enables studying toxic effects at the level of biomolecules and physicochemical processes, as well as determining the toxicity of general and oxidative types. Toxic and detoxifying effects of bioactive compounds were studied. Fullerenols, perspective pharmaceutical agents, nanosized particles, water‐soluble polyhydroxylated fullerene‐60 derivatives were chosen as bioactive compounds. Two homologous fullerenols with different number and type of substituents, C60O2–4(OH)20–24 and Fe0.5C60(OH) xOy (x + y = 40–42), were used. They suppressed bioluminescent intensity at concentrations >0.01 g L?1 and >0.001 g L?1 for C60O2–4(OH)20‐24 and Fe0.5C60(OH)xOy, respectively; hence, a lower toxicity of C60O2–4(OH)20–24 was demonstrated. Antioxidant activity of fullerenols was studied in model solutions of organic and inorganic oxidizers; changes in toxicities of general and oxidative type were determined; detoxification coefficients were calculated. Fullerenol C60O2–4(OH)20–24 revealed higher antioxidant ability at concentrations 10?17?10?5 g L?1. The difference in the toxicity and antioxidant activity of fullerenols was explained through their electron donor/acceptor properties and different catalytic activity. Principles of bioluminescent enzyme assay application for evaluating the toxic effect and antioxidant activity of bioactive compounds were summarized and the procedure steps were described.  相似文献
