Polymeric carriers are extensively used in photodynamic therapy (PDT) for increase of efficacy of photosensitizers. Here, we report the influence of nine Pluronic copolymers on phototoxicity of chlorin e6 (Ce6), in particular 5‐ to 7‐fold rise in the phototoxicity caused by hydrophilic Pluronics F127, F108, F68 and F87 and practically no influence on Ce6 of more hydrophobic polymers. The revealed value of 0.2 mg mL?1 of Pluronic F127 concentration sufficient for half‐of‐maximal increase of Ce6 photodynamic activity proved to be close to 0.16 mg mL?1 inherent in well‐documented carrier poly(N‐vinylpyrrolidone) (PVP). The dissociation constants of Ce6 complexes with Pluronic F127 and PVP that were estimated from UV spectra were 0.252 and 0.036 mg mL?1, respectively, indicating higher stability of Ce6 complex with PVP. According to the results of 1H‐NMR studies of Ce6 complexes, the porphyrin interacts not only with hydrophobic regions but also with hydrophilic sides of both polymers.  相似文献

Treatment of trans-[PtCl(4)(RCN)(2)] (R = Me, Et) with R'OH (R' = Me, Et, n-Pr, i-Pr, n-Bu) at 45 degrees C in all cases allowed the isolation of the trans-[PtCl(4)[(E)-NH=C(R)OR'](2)] imino ester complexes, while the reaction between cis-[PtCl(4)(RCN)(2)] and the least sterically hindered alcohols (methanol and ethanol) results in the formation of cis-[PtCl(4)[(E)-NH=C(R)OR'](2)] (R/R' = Me/Me) or trans-[PtCl(4)[(E)-NH=C(Et)OR'](2)] (R' = Me, Et), the latter being formed via thermal isomerization (ROH, reflux, 3 h) of the initially formed corresponding cis isomers. The reaction between alcohols R'OH and cis-[PtCl(4)(RCN)(2)] (R = Me, R' = Et, n-Pr, i-Pr, n-Bu; R = Et; R' = n-Pr, i-Pr, n-Bu), exhibiting greater R/R' steric congestion, allowed the isolation of cis-[PtCl(4)[(E)-NH=C(R)OR'][(Z)-NH=C(R)OR']] as the major products. The alcoholysis reactions of poorly soluble [PtCl(4)(RCN)(2)] (R = CH(2)Ph, Ph) performed under heterogeneous conditions, directly in the appropriate alcohol and for a prolonged time and, for R = Ph, with heating led to trans-[PtCl(4)[(E)-NH=C(R)OR'](2)] (R = CH(2)Ph, R' = Me, Et, n-Pr, i-Pr; R = Ph, R' = Me) isolated in moderate yields. In all of the cases, in contrast to platinum(II) systems, addition of R'OH to the organonitrile platinum(IV) complexes occurs under mild conditions and does not require a base as a catalyst. The formed isomerically pure (imino ester)Pt(IV) complexes can be reduced selectively, by Ph(3)P=CHCO(2)Me, to the corresponding isomers of (imino ester)Pt(II) species, exhibiting antitumor activity, without change in configuration of the imino ester ligands. Furthemore, the imino esters NH=C(R)OR' can be liberated from both platinum(IV) and platinum(II) complexes [PtCl(n)[H=C(R)OR'](2)] (n = 2, 4) by reaction with 1,2-bis(diphenylphosphino)ethane and pyridine, respectively. All of the prepared compounds were characterized by elemental analyses (C, H, N), FAB mass spectrometry, IR, and (1)H, (13)C[(1)H], and (195)Pt (metal complexes) NMR spectroscopies; the E and Z configurations of the imino ester ligands in solution were determined by observation of the nuclear Overhauser effect. X-ray structure determinations were performed for trans-[PtCl(4)[(E)-NH=C(Me)OEt](2)] (2), trans-[PtCl(4)[(E)-NH=C(Et)OEt](2)] (10), trans-[PtCl(4)[(E)-NH=C(Et)OPr-i](2)] (11), trans-[PtCl(4)[(E)-NH=C(Et)OPr-n](2)] (12), and cis-[PtCl(4)[(E)-NH=C(Et)OMe](2)] (14). Ab initio calculations have shown that the EE isomers are the most stable ones for both platinum(II) and platinum(IV) complexes, whereas the most stable configurations for the ZZ isomers are less stable than the respective EZ isomers, indicating an increase of the stability on moving from the ZZ to the EE configurations which is more pronounced for the Pt(IV) complexes than for the Pt(II) species.  相似文献

There is a great demand for simple, fast and accurate methods for quantification of volatile organic contaminants in soil samples. Solid-phase microextraction (SPME) has a huge potential for this purpose, but its application is limited by insufficient accuracy caused by a matrix effect. The aim of this research was to develop the method for BTEX quantification in soil using combined standard addition (SA) and internal standard (IS) calibration. Deuterated benzene (benzene-d6) was used as the internal standard for all analytes. The optimized method includes spiking replicate samples with different concentrations of BTEX standards and the same concentration of benzene-d6, equilibration of soil samples at 40 °C during 2 h, and SPME–GC–MS analysis. Precision and accuracy of IS and SA methods were compared on different soil matrices. Combined SA + IS method provided more precise calibration plots compared to the conventional SA calibration. The SA + IS calibration provided more precise and accurate results compared with a reference method based on solvent extraction followed by GC–MS when applied to BTEX quantification in real soil samples (spiked with diesel fuel and aged). Recoveries of BTEX from soil samples spiked with known concentrations of analytes using the developed method were in the range of 73–130% with RSD values less than 15% for all BTEX. The proposed simultaneous standard addition and internal standard approach can be advantageous and adopted for improved quantification of other toxic VOCs in soil.  相似文献

The biological activity of drugs on organisms is associated with the pharmacokinetic properties, such as the ability to penetrate through environments of varying polarity such as cellular organelles. In this area, particular attention is turned to the physicochemical properties that determine the potential of drugs to pass across the blood–brain barrier and thus to act on the central nervous system. In this study, special effort has been devoted to the simulation of passive diffusion of seven drugs (propranolol, ibuprofen, atenolol, promazine, chlorpromazine, imipramine, and desipramine) through the blood–brain barrier by high-performance liquid chromatography (HPLC) using a column with an immobilized artificial membrane. Gradient reverse elution was used to develop a linear correlation model for the capacity factors kIAM and the in vivo logarithmic values of brain-to-blood drug concentration ratios (log BB) with R of 0.9851. Eleven additional pharmaceuticals were determined by the same method to predict their potential to penetrate the blood–brain barrier. The reported analytical method represents an alternative tool for rapid and noninvasive assessment of the absorption properties of chemicals, especially for the development of novel drugs. The retention of the studied compounds on the immobilized artificial membrane column was also compared with three other C18-based stationary phases. Herein, the results of the HPLC determination of drugs using an immobilized artificial membrane are briefly discussed with respect to a general application of the method for evaluating a broader spectrum of pharmaceutical compounds.  相似文献

The nucleophilic addition of 2,4-dithiobiurete, 1- and 1,4-substituted 2,4-dithiobiuretes (2 a-e) with benzoylacetylene (1) has been studied. 2-(Benzoylmethyl)-4-(R1-imino)-6-(R2-imino)dihydro-4H-1,3,5-dithiazinium perchlorates (3 a-e) are obtained in glacial acetic acid (AcOH) in the presence of equimolar quantities of HClO4. The reaction of benzoylacetylene with 1,5-diphenyl-2,4-dithiobiurete in either of MeOH, C6H6, or MeCN solvents proceeds non-selectively to give a mixture of products such as 2-(benzoylmethyl)-4,6-di(phenylimino)dihydro-4H-1,3,5-dithiazine (5), 2-(benzoylmethyl)-4-(β-benzoylvinyl)thio-3-phenyl-6-(phenylimino)-3,6-dihydro-2H-1,3,5-thiadiazine (8), 2-(benzoylmethyl)-1,3-diphenylhexahydro-1,3,5-triazine-4,6-dithione (7) and N-(β-benzoylvinyl)-N-phenylthioureas (6).  相似文献

Post-Hartree-Fock ab initio quantum chemical calculations were performed for 5-fluorouracil in the gas phase and in a three-water cluster. Full geometry optimizations of the 5-fluorouracil-water complexes were carried out at the MP2/6-31+G(d,p) level of theory. MP4/6-31+G(d,p)//MP2/6-31+G(d,p) and MP4/6-31++G(d,p)//MP2/6-31+G(d,p) single-point calculations were performed to obtain more accurate energies. In water solution, 5-fluorouracil exists mainly in the 2,4-dioxo form (A). We propose that the populations of the 2-hydroxy-4-oxo (B) and 4-hydroxy-2-oxo (D) tautomers are 1 x 10(-4)% and 3.9 x 10(-8)%, respectively, on the basis of the relative stabilities of the tautomers calculated at the MP4/6-31++G(d,p)//MP2/6-31+G(d,p) level of theory. A profound difference between isolated and hydrated 5-fluorouracil is noted for the height of the tautomerization barrier. In the absence of water, the process of proton transfer is very slow. The addition of water molecules decreases the barrier by 2.3 times, making the process much faster. The minimum energy path (MP2/6-31+G(d,p)) for water-assisted proton transfer in trihydrated 5-fluorouracil was followed. CNDO/S-CI calculations predict singlet pi-pi(*) electron transitions at 312 nm for B and at 318 nm for D. The fluorescence spectrum of 5-fluorouracil in water confirms the presence of the hydroxy tautomer.  相似文献

A method based on solid-phase microextraction (SPME) followed by gas chromatography with microwave-induced plasma atomic emission detection for determining 16 pesticides of different chemical families (organochlorines, organophosphorus compounds and pyrethrins) in honey is proposed. Parameters affecting the sample enrichment step, such as sample mass, ionic strength, absorption and desorption times and temperatures, were carefully optimized in the direct immersion mode. Element-specific detection and quantification was carried out by monitoring the chlorine (479 nm), bromine (478 nm) and sulphur (181 nm) emission lines, which provided nearly specific chromatograms. The matrix effect was evaluated for samples of different floral origin, it being concluded that standard addition calibration was required for quantification purposes. The detection limits ranged from 0.02 to 10 ng g−1, depending on the compound and the honey sample under analysis. The method is reliable and can be considered useful for routine monitoring. None of the honey samples analyzed contained the studied compounds at concentrations above the corresponding detection limits.  相似文献
