The changes in the surface chemistry of polystyrene films exposed to natural weathering in Saudi Arabia have been followed with ESCA. Extensive incorporation of oxygen is observed, involving reaction of the main backbone chain and the pendant phenyl groups. The rate of surface photo-oxidation is strongly dependent on the photon flux during the time of exposure.  相似文献

We present our investigations of 2-ethyl-3-methyl-(10H)-dipyrrin-1-one, its self-association, and anion binding properties. This receptor is easily accessible in a facile single step synthesis with a straightforward workup. An examination of the concentration dependence of the dipyrrinone NH chemical shifts in CDCl3 and (CDCl2)2 over the temperature range from −20 °C to 100 °C determined the self-association constant to be 3850 M−1. Molecular recognition studies have shown that it has a preference for guests with an OH moiety, such as hydrogen sulfate (HSO4) and carboxylic acids (RCO2H).  相似文献

A new kind of chiral dienophiles, cyclic vinyl-p-tolylsulfilimines (2a and 2b), were obtained from the corresponding (Z)-sulfinylacrylonitriles with HBF(4) and methanol. The asymmetric Diels-Alder reaction of optically pure 2a with cyclopentadiene under mild thermal or catalyzed conditions afforded only the endo-4a adduct with complete endo and pi-facial selectivities. The ability of the sulfilimine moiety to enhance the dienophilic reactivity of the double bond is similar to that of the sulfinyl group.  相似文献

Inclusion of a polymer cushion between a lipid bilayer membrane and a solid surface has been suggested as a means to provide a soft, deformable layer that will allow for transmembrane protein insertion and mobility. In this study, mobile, tethered lipid bilayers were formed on a poly(ethylene glycol) (PEG) support via a two-step adsorption process. The PEG films were prepared by coadsorbing a heterofunctional, telechelic PEG lipopolymer (1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-poly(ethylene glycol)-2000-N-[3-(2-(pyridyldithio)propionate]) (DSPE-PEG-PDP) and a nonlipid functionalized PEG-PDP from an ethanol/water mixture, as described in a previous paper (Munro, J. C.; Frank, C. W. Langmuir 2004, 20, 3339-3349). Then a two-step lipid adsorption strategy was used. First, lipids were adsorbed onto the PEG support from a hexane solution. Second, vesicles were adsorbed and fused on the surface to create a bilayer in an aqueous environment. Fluorescence recovery after photobleaching experiments show that this process results in mobile bilayers with diffusion coefficients on the order of 2 microm2/s. The mobility of the bilayers is decreased slightly by increasing the density of tethered lipids. The formation of bilayers, and not multilayer structures, is also confirmed by surface plasmon resonance, which was used to determine in situ film thickness, and by fluorimetry, which was used to determine quantitatively the fluorescence intensity for each 18 by 18 mm sample. Unfortunately, fluorescence microscopy also shows that there are large defects on the samples, which limits the utility of this system.  相似文献

To examine how azole inhibitors interact with the heme active site of the cytochrome P450 enzymes, we have performed a series of density functional theory studies on azole binding. These are the first density functional studies on azole interactions with a heme center and give fundamental insight into how azoles inhibit the catalytic function of P450 enzymes. Since azoles come in many varieties, we tested three typical azole motifs representing a broad range of azole and azole-type inhibitors: methylimidazolate, methyltriazolate, and pyridine. These structural motifs represent typical azoles, such as econazole, fluconazole, and metyrapone. The calculations show that azole binding is a stepwise mechanism whereby first the water molecule from the resting state of P450 is released from the sixth binding site of the heme to create a pentacoordinated active site followed by coordination of the azole nitrogen to the heme iron. This process leads to the breaking of a hydrogen bond between the resting state water molecule and the approaching inhibitor molecule. Although, formally, the water molecule is released in the first step of the reaction mechanism and a pentacoordinated heme is created, this does not lead to an observed spin state crossing. Thus, we show that release of a water molecule from the resting state of P450 enzymes to create a pentacoordinated heme will lead to a doublet to quartet spin state crossing at an Fe-OH(2) distance of approximately 3.0 A, while the azole substitution process takes place at shorter distances. Azoles bind heme with significantly stronger binding energies than a water molecule, so that these inhibitors block the catalytic cycle of the enzyme and prevent oxygen binding and the catalysis of substrate oxidation. Perturbations within the active site (e.g., a polarized environment) have little effect on the relative energies of azole binding. Studies with an extra hydrogen-bonded ethanol molecule in the model, mimicking the active site of the CYP121 P450, show that the resting state and azole binding structures are close in energy, which may lead to chemical equilibrium between the two structures, as indeed observed with recent protein structural studies that have demonstrated two distinct azole binding mechanisms to P450 heme.  相似文献

Absorption and luminescence excitation spectra of solid benzene and benzene isolated in rare-gas matrices have been studied. The absolute absorption cross sections of various electronic band systems including the Rydberg region of C6H6 are obtained using transmission spectra of C6H6-doped rare-gas films of different thickness. An interpretation of the structure in the Rydberg region is given in terms of two Rydberg series using the quantum defect method. A comparative study has been made of luminescence excitation spectra of matrix-isolated and of pure C6H6. The luminescence excitation spectra of matrix-isolated benzene gives information on the internal conversion and medium-induced vibrational relaxation.  相似文献

Noncentrality of large impurity atoms—Pb and Sn atoms—substituting for Ge atoms in a GeTe lattice has been discovered by means of EXAFS investigations in Ge0.9Pb0.1Te and Ge0.85Sn0.15Te samples. The transition of impurity atoms into a noncentral position under conditions of a strong local stress is explained by the participation of an unshared electron pair from the impurity atoms in the formation of the chemical bond. We demonstrate that photoexcitation of NAD(P)H at 355 nm using a Nd:YAG laser leads to rapid reduction of the heme domain of the Bacillus megaterium fatty acid hydroxylase flavocytochrome P450 BM3. An aqueous electron derived from photoexcited NAD(P)H is rapidly transferred to the heme domain, enabling the formation of a carbon monoxy complex of the ferrous P450 (FeII-CO) on the microsecond time scale. Using this approach we have determined the limiting rate constant (1770 s-1 for substrate-free heme domain) for formation of the FeII-CO complex. We find no dependence of the observed rate of FeII-CO complex formation on NAD(P)H concentration but demonstrate a hyperbolic dependence on carbon monoxide concentration. The apparent dissociation constant for the complex of carbon monoxide bound noncovalently to the ferric form of the BM3 heme domain (and with NADH as reductant) is 323 microM. Binding of a P450 substrate (N-palmitoylglycine) weakened the complex between carbon monoxide and the ferric BM3 heme domain (Kd increased to 1404 microM) but enhanced the rate of formation of the FeII-CO complex (3036 s-1 for substrate-free heme domain). This study demonstrates the applicability of NAD(P)H photoexcitation as a method for rapid electron delivery to P450 enzymes and provides a new route to probing the P450 catalytic cycle and its transient intermediates.  相似文献

This paper pursues the current debate about emancipation as one of the principal tenets of critical OR. It highlights some conceptual ambiguities involved in the notion of emancipation and so attempts to multiply the possibilities for an emancipatory approach to OR. The paper will explore three different emancipatory themes, one which understands the concept of emancipation in terms of a democratic discourse, a second which views it in terms of the cultivation of different virtues and values, and a third which questions the very essence of ethics and emancipation. Central to this analysis is a concern for the methodological implications of these three themes, and the current neglect of emancipatory methodologies for OR practice.  相似文献
