A substoichiometric method for the determination of heavy rare earths (holmium, thulium) has been developed. After the addition of a substoichiometric amount of EDTA to the test sample of rare earth labelled with its radioactive isotopes, the negatively charged complex formed was separated by passage through a column of Dowex 50 x 8. Interfering metals can be removed by preliminary cupferron and diethylammonium diethyldithiocarbamate extraction. Concentrations of rare earth down to 4 x 10(-7)g 5 ml have been determined. 相似文献

The application of d.c. and square-wave polarographic measurements for the indication of the end-point in titrations with triethylenetetraminehexa-acetic acid (TTHA) has been studied. TTHA has ten co-ordinating groups and forms complexes with metal to ligand ratios of either 1:1 or 2:1, depending on the metal and experimental conditions. The shape of the titration curves, determined by the dependence of the limiting current (peak current of the square-wave polarographic curve) or of the half-wave potential (peak potential) on the amount of TTHA added indicates the composition of the complex formed in the titration. The composition of this complex as determined from the studied titration curves agrees with that predicted by theory. Titrations with polarographic indication of the end-point were also applied for direct titrations of binary mixtures of metal ions. The resulting titration curves indicated the existence of mixed dinuclear complexes and also the kinetic factors involved in the reactions between two different metal ions and TTHA. 相似文献

Using two-dimensional electrophoresis we investigated the effect of 5-aminolevulinic acid (ALA)-based photodynamic therapy (PDT; induction with 1 mM ALA for 4 h followed by blue light dose of 18 J/cm2) on the protein expression in HL60 leukemia cells. ALA-PDT resulted in extensive qualitative and quantitative changes in the protein pattern of HL60 cell lysates. Of more than 1350 protein spots recognized on the protein maps of ALA-induced cells, seven proteins were enhanced and 17 suppressed following irradiation. Three of these, calreticulin precursor, p58 microsomal protein (ERp57) and protein disulfide isomerase (p55) have been identified by matrix-assisted laser desorption and ionization-mass spectrometry and the pI/molecular weight parameters of the affected proteins were estimated by computer analysis. The findings suggest participation of endoplasmic reticulum Ca(2+)-binding chaperones and/or Ca2+ signaling in ALA-PDT mediated cytotoxicity. 相似文献

A simple and rapid method for the substoichiometric determination of traces of manganese in various materials by neutron-activation analysis has been developed. After dissolution of the irradiated test sample, manganese(II) carrier is added and subsequently oxidised by peroxodisulphate to the heptavalent state. The permanganate thus formed is finally extracted into chloroform as tetraphenylarsonium permanganate using a substoichiometric amount of tetraphenylarsonium chloride. This single separation step isolates radiochemically pure manganese-56 in the analysis of a relatively simple material in which interfering elements (gold, rhenium, etc.) are absent. When this is not true, a preliminary separation of manganese from the irradiated sample, based on the extraction of manganese diethyldithio-carbamate into chloroform and followed by stripping of the manganese with dilute sulphuric acid, must be used. A simultaneously irradiated standard containing manganese must be treated in exactly the same way as a test sample. In the materials analysed by the new method 10(-5) to 10(-3)% of manganese has been determined. 相似文献

The presence of excited-state buffer-mediated proton exchange reactions influences the steady-state fluorescence signals from dyes in solution. Since biomolecules in general have some chemical groups that can act as proton acceptors/donors and are usually dissolved in buffer solutions which can also behave as appropriate proton acceptors/donors, the excited-state proton exchange reactions may result in distorted steady-state fluorescence signals. In a previous paper (J. Phys. Chem. A 2005, 109, 734-747), we evaluated kinetic and other pertinent parameters for the excited-state proton reactions of the prototropic forms of 2',7'-difluorofluorescein (Oregon Green 488, OG488), recording a fluorescence decay surface at different pH values and acetate buffer concentrations, analyzed by means of global compartmental analysis. In this article we use the rate constants and the corrected pre-exponential factors from the previously recorded fluorescence decay traces to simulate the decay times and associated pre-exponentials at different acetate buffer concentrations and constant pH and compare these theoretically calculated values with new experimental data. We also calculate the steady-state fluorescence intensity vs pH and vs acetate buffer concentration (at constant pH) and compare these calculated emission values with the experimental data previously published. Isoprenoids are natural compounds essential for a great number of cellular functions. One of them is farnesol (FOH), which can reduce cell proliferation, but its low solubility in aqueous solvents limits its possible clinical use as a pharmacological tool. One alternative is the use of cyclodextrins (CDs) which house hydrophobic molecules forming inclusion complexes. To assess FOH potential application in anticancer treatments, Sulfobutylated β-cyclodextrin Sodium Salt (SBE-β-CD) was selected, due to it has high solubility, approbation by the FDA, and numerous studies that ensure its safety to be administered parenterally or orally without nephrotoxicity associated. The therapeutic action of farnesol and complex were studied in different carcinoma cells, compared with a normal cell line. Farnesol showed selectivity, affecting the viability of colon and liver cancer cells more than in breast cancer cells and fibroblasts. All cells suffered apoptosis after being treated with 150 μM of free FOH, but the complex reduced their cell viability between 50 and 75%. Similar results were obtained for both types of isomers, and the addition of phosphatidylcholine reverses this effect. Finally, cell cycle analysis corroborates the action of FOH as inducer of a G0/G1 phase; when the cells were treated using the complex form, this viability was reduced, reaching 50% in the case of colon and liver, 60% in fibroblasts, and only 75% in breast cancer. 相似文献

Various one- and two-step extraction procedures with and without a short application of ultrasound at the beginning of the extraction were used to examine the effect of sonication on the extractibility of the hemicellulose components of buckwheat hulls. The polysaccharides recovered from the extracts were characterised by yield as well as composition determined by chemical methods and spectroscopic techniques. They comprised a complex of glucuronoxylan and co-extracted amylose-rich starch in various proportions contaminated with other cell wall components (protein, pectic polysaccharides). The hemicellulose fractions obtained by classical and ultrasound-assisted extraction exhibited significant immunomodulatory activities. The increased yield of ultrasonically extracted hemicelluloses, which have preserved their structural and molecular properties as well as immunological activity, confirmed the importance and great potential of ultrasound-assisted extraction of industrially important polysaccharides from different tissues of plant materials. 相似文献
