Identification of specific packing interactions within in the hydrophobic core of proteins is important for understanding the integrity of protein structure. Finding such interactions is challenging because few tools allow monitoring of a specific interaction in the presence of several non‐specific forces that hold proteins together. It is important to understand how and when such interactions develop during protein folding. In this study, we have used the intrinsic tryptophan residue, Trp53, as an ultraviolet resonance Raman probe to elucidate the packing interactions in the hydrophobic core of the protein barstar. Barstar is extensively studied for its folding, unfolding and aggregation properties. The Trp53 residue is known to be completely buried in the hydrophobic core of the protein and is used extensively as an intrinsic probe to monitor the folding and unfolding reactions of barstar. A comparison of the resonance Raman cross sections of some bands of Trp53 with those observed for N‐acetyl‐tryptophanoamide in water suggests that Trp53 in barstar is indeed isolated from water. Intensity ratio of the Fermi doublet suggests that Trp53 is surrounded by several aliphatic amino acid residues in corroboration with the crystal structure of barstar. Importantly, we show that the side chain of Trp53 is involved in a unique CH–π interaction with CH groups of Phe56 as well as a steric interaction with the methyl group of Ile5. Calcium 5-aminoisophthalate {[Ca(AIP)(OH2)4](H2O)} proportional, variant (1), synthesized from Ca(OH)2 and H2AIP, exists as a double helix. Hydrogen-bonding interactions between metal-bound water molecules, carboxylate oxygen, and the -NH2 group result in the formation of a supramolecular assembly, where the double helices are connected to each other to form hexagonal channels filled with water molecules.  相似文献

Hole transporting material (HTM) is a significant component to achieve the high performance perovskite solar cells (PSCs). Over the years, inorganic, organic and hybrid (organic‐inorganic) material based HTMs have been developed and investigated successfully. Today, perovskite solar cells achieved the efficiency of 22.1 % with with 2,2’,7,7’‐tetrakis(N,N‐di‐p‐methoxyphenyl‐amine) 9,9‐spirobifluorene (spiro‐OMeTAD) as HTM. Nevertheless, synthesis and cost of organic HTMs is a major challenging issue and therefore alternative materials are required. From the past few years, inorganic HTMs showed large improvement in power conversion efficiency (PCE) and stability. Recently CuOx reached the PCE of 19.0% with better stability. These developments affirms that inorganic HTMs are better alternativesto the organic HTMs for next generation PSCs. In this report, we mainly focussed on the recent advances of inorganic and hybrid HTMs for PSCs and highlighted the efficiency and stability of PSCs improved by changing metal oxides as HTMs. Consequently, we expect that energy levels of these inorganic HTMs matches very well with the valence band of perovskites and improved efficiency helps in future practical deployment of low cost PSCs.  相似文献

Plasmodium falciparum (Pf) hypoxanthine‐guanine phosphoribosyltransferase (HGPRT) is a potential therapeutic target. Compared to structurally homologous human enzymes, it has expanded substrate specificity. In this study, 9‐deazapurines are used as in situ probes of the active sites of human and Pf HGPRTs. Through the use of these probes it is found that non‐covalent interactions stabilise the pre‐transition state of the HGPRT‐catalysed reaction. Vibrational spectra reveal that the bound substrates are extensively distorted, the carbonyl bond of nucleobase moiety is weakened and the substrate is destabilised along the reaction coordinate. Raman shifts of the human and Pf enzymes are used to quantify the differing degrees of hydrogen bonding in the homologues. A decreased Raman cross‐section in enzyme‐bound 9‐deazaguanine (9DAG) shows that the phenylalanine residue (Phe186 in human and Phe197 in Pf) of HGPRT stacks with the nucleobase. Differential loss of the Raman cross‐section suggests that the active site is more compact in human HGPRT as compared to the Pf enzyme, and is more so in the phosphoribosyl pyrophosphate (PRPP) complex 9DAG–PRPP–HGPRT than in 9‐deazahypoxanthine (9DAH)–PRPP–HGPRT.  相似文献

Adsorption of Fe, Co and Ni atoms on a hybrid hexagonal sheet of graphene and boron nitride is studied using density functional methods. Most favorable adsorption sites for these adatoms are identified for different widths of the graphene and boron nitride regions. Electronic structure and magnetic properties of the TM-adsorbed sheets are then studied in detail. The TM atoms change the electronic structure of the sheet significantly, and the resulting system can be a magnetic semiconductor, semi-metal, or a non-magnetic semiconductor depending on the TM chosen. This gives tunability of properties which can be useful in novel electronics applications. Finally, barriers for diffusion of the adatoms on the sheet are calculated, and their tendency to agglomerate on the sheet is estimated.  相似文献

The use of cisplatin as a chemotherapeutic drug is impeded by the development of drug resistance. Combination therapies of a chemosensitizer for cisplatin have been studied, but with little success, and the search for an effective combination therapy is continuing. Our earlier reports have shown that Zanthoxylum armatum DC. extract enhances the apoptotic effect of cisplatin in cancer cell lines. In this study, we purified and identified the bioactive phytocompound through bio-assay-guided purification, using column chromatography and HPLC. Chemical characterization using NMR and mass spectrometry revealed the compound as planispine A, with molecular structure C25H30O6 and molecular weight, 426.16 g/mol. Planispine A was found to inhibit cancer cell proliferation in a dose-dependent manner and to sensitize the cancer cells to cisplatin-augmented apoptotic cell death, in a caspase-dependent manner. A combination of planispine A and cisplatin induced S-phase cell cycle arrest, and reduced the expression of survival proteins such as cyclin D1. Interestingly, planispine A inhibits the Fanconi anemia pathway, as shown by reduced FANCD2 foci formation and FANCD2 monoubiquitination, which revealed the molecular mechanism of chemo-sensitization of cancer cells to cisplatin. Evaluation of this combination therapy in cisplatin-resistant tumors may lead to more efficient cisplatin treatment.  相似文献

Microremediation of harmful radioactive waste such as uranium has been carried out by the endophytic actinomycetes strains isolated from the unnoticed fallen leaves of commonly available medicinal plant Azadirachta indica, which are considered as unique source. Among six actinobacteria isolates, one microbe (A5) effectively removed uranium in 12 h at temperature 30 °C and pH 8–9. Molecular characterization and phylogenetic analysis support the classification of the isolate A5 as a new strain which was named as Streptomyces sp.
