Determination of proteinases--enzymes that catalyze the hydrolysis of peptide bonds--is often difficult due to the presence of interferences in complex biological media and limited sample size. Capillary electrophoresis (CE), with laser-induced fluorescence (LIF) can serve as a useful tool for such determinations. LIF detection offers the advantages of increased sensitivity and increased selectivity. However, direct LIF detection requires the proteinase analyte to be fluorescently derivatized prior to analysis. A viable alternative is offered by the present work, in which protein substrates are first labeled with BODIPY dye, a relatively pH-insensitive, high-fluorescence quantum yield dye. Upon binding of some 4-10 molecules of dye to a single protein, the dye is effectively fluorescence-quenched. Digestion of the BODIPY--labeled and quenched protein by an unlabeled enzyme yields smaller peptide fragments in which the fluorescence of associated BODIPY tags is restored. We will present how the fragmentation pattern of BODIPY-labeled casein changes as a function of incubation time with trypsin, as well as the effect of varying concentrations of trypsin on the BODIPY-casein digest.  相似文献

Reaction of [Ti(NR)Cl2(py)3] (R=tBu or 2,6-iPr2C6H3) with K(2)[COT] (COT=C8H8) or Li2[COT'] (COT'=1,4-C8H6(SiMe3)2) gave the monomeric complexes [Ti(NR)(eta8-COT)] or [Ti(NR)(eta8-COT')], respectively. The pseudo-two coordinate, "pogo stick" geometry for these complexes is unique in both early transition-metal and cyclooctatetraenyl ligand chemistry. In contrast, reaction of [Ti(N-2,6-Me2C6H3)Cl2(py)3] with K2[COT] gave the mu-imido-bridged dimer [Ti2(mu-N-2,6-Me2C6H3)2(eta8-COT)2]. It appears that as the steric bulk of the imido and C8 ring substituents are decreased, dimerisation becomes more favourable. Aryl imido COT complexes were also prepared by imido ligand exchange reactions between anilines and [Ti(NtBu)(eta(8)-COT)] or [Ti(NtBu)(eta(8)-COT')]. The complexes [Ti(NtBu)(eta(8)-COT)], [Ti(N-2,6-iPr2C6H3)2(eta8-COT)] and [Ti2(mu-N-2,6-Me2C6H3)2(eta8-COT)2] have been crystallographically characterised. The electronic structures of both the monomeric and dimeric complexes have been investigated by using density functional theory (DFT) calculations and gas-phase photoelectron spectroscopy. The most striking aspect of the bonding is that binding to the imido nitrogen atom is primarily through sigma and pi interactions, whereas that to the COT or COT' ring is almost exclusively through delta symmetry orbitals. A DFT-based comparison between the bonding in [Ti(NtBu)(eta8-COT)] and the bonding in the previously reported late transition-metal "pogo stick"complexes [Os(NtBu)(eta6-C6Me6)], [Ir(NtBu)(eta5-C5Me5)] and [Ni(NO)(eta5-C5H5)] has also been undertaken.  相似文献

The naturally occurring 1,4-benzoquinones 2-methoxy-6-propyl-1,4-benzoquinone (1), 2-methoxy-6-pentyl-1,4-benzoquinone (primin 2), 2-methoxy-6-pentadecyl-1,4-benzoquinone (3), 2-methoxy-6-heptadecyl-1,4-benzoquinone (dihydroirisquinone, pallasone B; 4) were synthesized by a simple protocol involving microwave accelerated Claisen rearrangement of allyl ethers 10, followed by hydrogenation of the side chain alkene, and oxidation to the quinone. The Claisen-based methodology was extended to the first synthesis of the marine benzoquinones verapliquinones A and B (5 and 6), and panicein A (7). Isoarnebifuranone (9) was also synthesized by a similar strategy.  相似文献

The syntheses of a series of chiral ureas containing the redox-active ferrocene group are described. Each of these bind chiral carboxylates in organic solvents through hydrogen-bonding interactions, as evidenced by spectroscopic and cyclic voltammetry measurements, the latter allowing these guests to be electrochemically sensed in solution. The enantioselectivity in the complexation of the protected amino acid N-benzenesulfonylproline by a ferrocenylbenzyl host is high enough to allow opposite enantiomers to be distinguished by electrochemical means.  相似文献

The spread of antibiotic resistance is a major challenge for the treatment of Mycobacterium tuberculosis infections. In addition, the efficacy of drugs is often limited by the restricted permeability of the mycomembrane. Frontline antibiotics inhibit mycomembrane biosynthesis, leading to rapid cell death. Inspired by this mechanism, we exploited β‐lactones as putative mycolic acid mimics to block serine hydrolases involved in their biosynthesis. Among a collection of β‐lactones, we found one hit with potent anti‐mycobacterial and bactericidal activity. Chemical proteomics using an alkynylated probe identified Pks13 and Ag85 serine hydrolases as major targets. Validation through enzyme assays and customized 13C metabolite profiling showed that both targets are functionally impaired by the β‐lactone. Co‐administration with front‐line antibiotics enhanced the potency against M. tuberculosis by more than 100‐fold, thus demonstrating the therapeutic potential of targeting mycomembrane biosynthesis serine hydrolases.  相似文献

Diazophosphonates, readily prepared from α-ketophosphonates by oxidation of the corresponding hydrazones in batch or in flow, are useful partners in 1,3-dipolar cycloaddition reactions to alkynes to give N-H pyrazoles, including the first intramolecular examples of such a process. The phosphoryl group imbues a number of desirable properties into the diazo 1,3-dipole. The electron-withdrawing nature of the phosphoryl stabilizes the diazo compound making it easier to handle, whilst the ability of the phosphoryl group to migrate readily in a [1,5]-sigmatropic rearrangement enables its transfer from C to N to aromatize the initial cycloadduct, and hence its facile removal from the final pyrazole product. Overall, the diazophosphonate acts as a surrogate for the much less stable diazoalkane in cycloadditions, with the phosphoryl group playing a vital, but traceless, role. The cycloaddition proceeds more readily with alkynes bearing electron-withdrawing groups, and is regiospecific with asymmetrical alkynes. The potential of diazophosphonates for use in bioorthogonal cycloadditions is demonstrated by their facile addition to strained alkynes.  相似文献
