Determination of Vecuronium Bromide in Pharmaceuticals: Development, Validation and Comparative Study of HPLC and CZE Analytical Methods

Vecuronium bromide is a neuromuscular blocking agent used for anesthesia to induce skeletal muscle relaxation. HPLC and CZE analytical methods were developed and validated for the quantitative determination of vecuronium bromide. The HPLC method was achieved on an amino column (Luna 150 × 4.6 mm, 5 μm) using UV detection at 205 nm. The mobile phase was composed of acetonitrile:water containing 25.0 mmol L^?1 of sodium phosphate monobasic (50:50 v/v), pH 4.6 and flow rate of 1.0 mL min^?1. The CZE method was achieved on an uncoated fused-silica capillary (40.0 cm total length, 31.5 cm effective length and 50 μm i.d.) using indirect UV detection at 230 nm. The electrolyte comprised 1.0 mmol L^?1 of quinine sulfate dihydrate at pH 3.3 and 8.0% of acetonitrile. The results were used to compare both techniques. No significant differences were observed (p > 0.05).     

Modeling of Selenoprotein Nitrozation: Synthetic Approaches

Model reactions have been carried out to evaluate the question of what kind of links may exist between the biological cycles of nitrogen oxide (NO) and those of selenoproteins, especially the amino acid selenocysteine (Sec). To collect information about the properties of the as-yet unknown selenonitrites (RSeNO) in comparison with the well-known thionitrites, (RSNO), the interaction of nitrosating reagents with a choice of molecular thiols and related selenoles as model compounds have been studied. Selenol nitrosation is clearly preferred in vitro to thiol nitrosation, but selenonitrites are thermally significantly less stable than the related thionitrites, suggesting that selenonitrites may be important, but yet-undetected intermediates in selenoprotein chemistry. Chemical trapping of RSeNO was achieved for the first time by its 1,4-addition to dimethylbutadiene leading to a stable unsaturated oxime.

The reaction of 2-(3-methylbiphenyl-2-yl)pyridine with IrCl(3).nH(2)O in the presence of water incorporates a tightly bonded CO at the iridium center; in the subsequent reaction with picolinic acid a complex revealing an unusual CH activation of a methyl group is formed.     

A diversity oriented, microwave assisted synthesis of N-substituted 2-hydro-4-amino-pyrido[2,3-d]pyrimidin-7(8H)-ones

A protocol for the synthesis of N-substituted 2-hydro-4-amino-pyrido[2,3-d]pyrimidin-7(8H)-ones (11) is described. Thus, the formylation of a 2-aminopyridone 12 in 85% formic acid/Ac₂O, proceeding via in situ cyclization to the intermediate formamide 13, affords the corresponding 2-hydro-4-oxo-pyridopyrimidine 14, which is converted to a 4-chloro-pyridopyrimidine 15 upon treatment with POCl₃. The subsequent transformation to the title compounds is carried by treatment with the corresponding amine in MeOH under microwave irradiation conditions.
