Improving the germination of economically important crops and the condition of young plants is a major challenge currently facing agricultural practice. Pea (Pisum sativum L.) is one of the four most important cultivated legumes, along with groundnut (Arachis hypogaea L.), soybean (Glycine max L.) and beans (Phaseolus vulgaris L.). Due to the high protein content (23–33%), there is an interest in growing this crop as a source of protein for humans and animals. In this study, we focused on the effect of Cold Atmospheric Pressure Plasma (CAPP) on the decontamination and germination of pea seeds, on young seedling growth and production parameters, and on increasing their resistance and mechanical strength. We can state that germination increased by 10 to 25% after plasma treatment, and the most significant decontamination effect was detected when using non-thermal plasma generated in the ambient air (A-variants) and in the nitrogen atmosphere (N-variants). The increased in situ activity of peroxidases (POX) in the cell walls of A-variants and N-variants is also closely related to the increase in the mechanical strength of the cell walls and thus contributes to the higher resistance of these seedlings. This is also illustrated by the differences in lignin deposition among the different variants after CAPP treatment. To our knowledge, this is the first study concerning the influence of CAPP on the lignification of root tissues and on increasing the strength and resistance of plants.

Two analytical procedures based on the generation of volatile tributyltin derivatives, their separation by headspace solid-phase microextraction (HS SPME) and subsequent determination using plasma optical emission spectrometry (OES) have been developed for the selective determination of trace tributyltin (TBT) in the presence of other butyltins and inorganic tin in sediments without the use of chromatography. A microwave-assisted leaching of tin compounds from the sediment using 25%v/v acetic acid is applied for sample pretreatment. The first method takes advantage of TBT chloride releasing from the lecheate after adding 3 M hydrochloric acid, and subsequent separation of the analyte by HS SPME using Carboxen-poly(dimethylsiloxane) (CAR/PDMS). The second method involves the use of masking agents, namely ethylenediaminetetraacetic acid (EDTA) and diphenylcarbazone (DFC), which form stable chelates with monobutyltin (MBT) and dibutyltin (DBT), respectively, followed by the ethylation of tributyltin at pH 5 using sodium tetraethylborate (NaBEt4) solution. The final concentration of NaBEt4 is 0.05%w/v. The parameters affecting the TBT derivatisation and separation by HS SPME have been optimised including the selection of SPME fibre coating (PDMS, CAR/PDMS), the amount of masking agents and NaBEt4 added, sorption time (2–40 min) and sorption temperature (25–60°C). Higher sensitivity and robustness are attained with the method involving ethylation derivatisation, leading to the limit of detection (LOD) of 3 ng L?1. The selective release of TBT is observed from aqueous solutions, where the concentrations of MBT and DBT were in 2–50-fold excess to TBT. The SPME-TD-MIP-OES methods have been validated against several certified reference materials (CRMs), including SOPH-1 marine sediment, PACS-2 marine sediment and BCR 646 freshwater sediment.  相似文献

The influence of meteorological parameters on radon exhalation rate was analyzed. The analyses show a significant influence of precipitation on radon exhalation rate. The highest exhalation rate was found during dry periods and the lowest during the periods of strong precipitation. We also tested two different types of detectors for the measurement of radon exhalation rate—the ionization chamber (AlphaGUARD) and the scintillation chamber of Lucas type. The exhalation rates measured by these detectors were in a good agreement. Likewise, there was an agreement in radon exhalation rates determined from short term and long term increases in radon activity concentration.  相似文献

A simple and convenient thin-layer chromatography (TLC) method combined with image analysis technique was developed to determine thiophanate methyl. The detection of pesticide was based on iodine–azide reaction. Digital images of TLC plate chromatograms were analysed using TLSee software, and quantitative analysis was conducted. The influence of the implicit solvent model on transition state structures of two S N2 reactions of biochemical importance is presented. In the considered methyl transfer reaction, we show experimentally that the rate constant in blood serum is about 60% slower than in the aqueous solution and that the implicit solvent model with slightly modified parameters for water captures correctly the energetics of this reaction. With the example of the reaction between 4-methyl-1,2,4-triazol-3-thione and ethyl bromoacetate, we show that relative stabilities of the conformationally different transition states depend upon the solvent inclusion strategy.  相似文献

This paper presents a study of the permeation of poly(ethylene oxide) (PEO) chains through the nanoporous wall of hollow polymeric capsules prepared by self-assembly of polyelectrolytes. We employ the method of pulsed field gradient (PFG) NMR diffusion to distinguish chains in different sites, i.e., in the capsule interior and free chains in the dispersion, by their respective diffusion coefficient. From a variation of the observation time, the time scale of the molecular exchange between both sites and thus the permeation rate constant is extracted from a two-site exchange model. Permeation rate constants show two different regimes with a different dependence on chain length. This suggests a transition between two different mechanisms of permeation as the molecular weight is increased. In either regime, the permeation time can be described by a scaling law tau approximately N (b) , with b = (4)/ 3 for short chains and b = (1)/ 3 for long chains. We discuss these exponents, which clearly differ from the theoretical predictions for chain translocation.  相似文献

Bone-targeting N-(2-hydroxypropyl)methacrylamide (HPMA) copolymer-PGE(1) conjugates, containing cathepsin K sensitive spacers, were incubated with induced osteoclasts and osteoblasts, their precursors, and control non-skeletal cells. The release of PGE(1) was monitored by an HPLC assay. In both murine and human cell lines, osteoclasts appeared to be the most active cells in the cleavage (PGE(1) release). Incubation with osteoblasts also resulted in fast PGE(1) release, whereas precursor and control cells released PGE(1) with a substantially slower rate than bone cells (apparently through ester bond cleavage). Experiments in the presence of inhibitors revealed that other enzymes, in addition to cathepsin K, were participating in the cleavage of the conjugate. Confocal fluorescence studies exposed internalization of the conjugate by endocytosis with ultimate localization in the lysosomal/endosomal compartment.  相似文献
