A synthesis of (−)-crobarbatic acid is reported along with the first use of a vinyl-substituted ketene dithioacetal as a coupling partner for an anodic cyclization reaction. The use of the vinyl-substituted ketene dithioacetal enables the construction of a cyclic product having a tetrasubstituted carbon with a relative stereochemistry opposite to that originally obtained from the cyclization.  相似文献

YM-254890 and FR900359 are potent and selective inhibitors of the Gq/11-signaling pathway. As such, they have been attractive targets for both synthesis and biological studies. Yet in spite of this effort, a versatile synthetic approach to the molecules that allows for the rapid construction of a variety of non-natural and labelled analogs and an increase in the amount of those analogs available remains elusive. The biological responses of four freshwater daphniid species, Daphnia middendorffiana, D. pulicaria, D. pulex and D. parvula, to a single acute dose of ultraviolet B radiation (UVB) were compared. In addition to survival, we compared the induction of DNA damage (i.e. cyclobutane pyrimidine dimers) between species as well as the ability to repair this damage in the presence or absence of photoreactivating light. All four species showed high levels of shielding against DNA damage when compared to damage induced in purified DNA dosimeters at the same time and dose. Significant variation in survival was observed between species depending on temperature and light conditions. Contrary to our expectations, all species showed significantly higher survival and light-dependent DNA damage removal rates at 10 degrees C compared to 20 degrees C, suggesting that the enhanced rate of photoenzymatic repair (PER) at the lower temperature contributed significantly to the recovery of these organisms from UVB. PER was highly effective in promoting survival of three of the four species at 10 degrees C, but at 20 degrees C it was only partially effective in two species, and ineffective in two others. None of the species showed significant dark repair at 20 degrees C and only D. pulicaria showed a significant capacity at 10 degrees C. Two species, D. middendorffiana and D. pulex, showed some short-term survival at 10 degrees C in absence of PER despite their inability to repair any appreciable amount of DNA damage in the dark. All species died rapidly at 20 degrees C in absence of PER, as predicted from complete or near-absence of nucleotide excision repair (NER). Overall, the protective effects of tissue structure and pigmentation were similar in all Daphnia species tested and greatly mitigated the absorption of UVB by DNA and its damaging effects. Surprisingly, the visibly melanotic D. middendorffiana was not better shielded from DNA damage than the three non-melanotic species, and in fact suffered the highest damage rates. Melanin content in this species was not temperature dependent under the experimental growth conditions, and so did not contribute to temperature-dependent responses. It is evident that different species within the same genus have developed diverse biological responses to UVB. Our data strongly suggest that DNA damage is lethal to Daphnia and that photoenzymatic repair is the primary mechanism for removing these lesions. In the absence of light, few species are capable of removing any DNA damage. Surprisingly, the single species in which significant excision repair was detected did so only at reduced temperature. This temperature-dependence of excision repair is striking and may reflect adaptations of certain organisms to stress in a complex and changing environment.  相似文献

Time-of-flight secondary ion mass spectrometry (TOF SIMS) has been used in conjunction with a mass spectrometry cleavable linker to determine the percent conversion of reactions that were conducted site-selectively on an addressable microelectrode array. When combined with fluorescence techniques for analysis of the reactions, the TOF SIMS experiment provides a means for optimization of both reaction confinement and reaction efficiency on the microelectrode arrays.  相似文献

What is new in the field of neutrino detection? In addition to new projects probing both the low and high ends of the neutrino energy scale, an inexpensive, effective technique is being developed to allow tagging of antineutrinos in water Cherenkov (WC) detectors via the addition to water of a solute with a large neutron cross-section and energetic γ daughters. Gadolinium is an excellent candidate since in recent years it has become very inexpensive, now less than $8 per kilogram in the form of commercially available gadolinium trichloride. This non-toxic, non-reactive substance is highly soluble in water. Neutron capture on gadolinium yields an 8.0 MeV gamma cascade easily seen in detectors like Super-Kamiokande. The uses of GdCl3 as a possible upgrade for the Super-Kamiokande detector — with a view toward improving its performance as an antineutrino detector for supernova neutrinos and reactor neutrinos — are discussed, as are the ongoing R&;D efforts which aim to make this dream a reality within the next two years.  相似文献

[reaction: see text] The removal of electroauxiliaries from peptide substrates with chemical oxidants has been examined as a method for inserting N-acyliminium ions into the peptides. To this end, it was found that both 4-methoxyphenyldimethylsilyl and 2,4-dimethoxyphenyldimethylsilyl electroauxiliaries were readily cleaved with the use of ceric ammonium nitrate. Of the two groups, the 2,4-dimethoxyphenyldimethylsilyl electroauxiliary was the most labile under the oxidative conditions. The oxidation reactions were shown to be compatible with the use of a solid-phase substrate.  相似文献

[reaction: see text]. Intramolecular coupling reactions between enol ether radical cations and oxygen nucleophiles are primarily governed by stereoelectronics. By taking advantage of this observation, a tetrahydrofuran building block for use in constructing (+)-linalool oxide and rotundisine has been synthesized in four steps from a commercially available starting material.
