Studies on antihypertensive agents with antithrombotic activity. 2. Syntheses and pharmacological evaluation of pyrrolo[2,3-c]azepine derivatives

As an extension of our previous investigation, a series of 7-aminoalkylpyrrolo[2,3-c]azepine derivatives was synthesized and evaluated as alpha1-adrenergic- and serotonin 2 (5-HT2)-receptor antagonists, with the aim of finding a novel potent antihypertensive agent with both activities. Among the compounds obtained in this study, (E)-1-ethyl-7-[3-[4-(4-fluorophenyl)piperazin-1-yl]propyl]-4-hy droxyimino-1,4,5,6,7,8-hexahydropyrrolo[2,3-c]azepin-8-on e (16d) displayed potent alpha1-adrenoceptor blocking activity (pA2=7.83+/-0.20) and 5-HT2-receptor blocking activity (pA2=9.47+/-0.17) in isolated guinea pig arteries. At 3 mg/kg oral administration, compound 16d exhibited antihypertensive activity more potent than that of doxazosin in deoxycorticosterone acetate (DOCA)-salt hypertensive dogs. Furthermore, this compound reduced the rate of mouse acute pulmonary thromboembolytic death induced by collagen and serotonin at oral doses of 0.3 mg/kg or more, and its effect lasted for at least 6 h at 3 mg/kg.     

Bartholdi zeta functions of digraphs

We give a determinant expression for the Bartholdi zeta function of a digraph which is not symmetric. This is a generalization of Bartholdi’s result on the Bartholdi zeta function of a graph or a symmetric digraph.     

Determination of traces of iron(II) in the presence of iron(III) by the bathophenanthroline method

Traces of iron(II) (1-30 ppM) in the presence of iron(III) were determined (error <10%) by the bathophenanthroline method. Interference of iron(III) was eliminated by masking with sodium pyrophosphate (2.5-60 mg). The iron(II) complex was extracted with n-butanol, at pH 4.2-4.7.     

(1R,2R,3S,4S)‐2‐[(2R,3S)‐2,3‐Di­methyl­oxiran‐2‐yl]‐3,4‐bis(4‐methoxy­benzyl­oxy)­cyclo­hexanol

The relative configuration was determined for the title com­pound, C₂₆H₃₄O₆, which was prepared in a synthetic study on immunosuppressant FR­65­814. There is an intra­mol­ecular hydrogen bond between the hydroxy and epoxy groups.     

Site-selective and stereoselective intramolecular (2pi + 2pi) photocycloaddition of arylalkenes to pyrene and its photocycloreversion

Irradiation of a benzene solution containing methyl p-(1-pyrenylmethoxymethyl)cinnamate (1a) with a high-pressure Hg lamp through Pyrex filter stereoselectively gave an intramolecular (2pi + 2pi) photocycloadduct (2a) in an 83% yield in a site-selective manner at the 4,5-position of the pyrene ring. Similar irradiation of an ortho-substituted derivative (3) afforded the corresponding (2pi + 2pi) cycloadduct (4) as a sole product at the 9,10-position of pyrene. The site-selective photocycloaddition can be reasonably explained by the intramolecular sandwich-type singlet exciplexes between the pyrene and phenyl rings.     

Isolation of polyhydroxyalkanoate-producing bacteria from a polluted soil and characterization of the isolated strain Bacillus cereus YB-4

We describe the characterization of polyhydroxyalkanoate (PHA)-producing bacteria isolated from an ammunition-polluted soil in Kitakyushu City, Japan. Over 270 strains were evaluated for PHA accumulation based on a colony staining method using Nile red. Of these, nine strains were selected based on the intensity of Nile red fluorescence and the cells were quantitatively analyzed for PHA by gas chromatography. PHA accumulation was observed in five strains, all of which are inferred to be close to the Bacillus cereus group according to 16S rDNA sequence analysis. Interestingly, these strains produced a PHA copolymer, poly(3-hydroxybutyrae-co-3-hydroxyvalerate) [P(3HB-co-3HV)], with a 3HV fraction up to 2 mol% with glucose as a carbon source. Further characterization was performed on one isolate, B. cereus YB-4. Gel permeation chromatography analysis revealed that the number of average molecular weights of PHA accumulated in B. cereus YB-4 drastically changed from 722,000 to 85,000 over a 72-h cultivation period. Furthermore, the PHA synthase genes were cloned and the deduced amino acid sequences were determined. This study provides new insights into PHA biosynthesis by members of the B. cereus group.     

A new entry to Amaryllidaceae alkaloids from carbohydrates: total synthesis of (+)-vittatine

The stereoselective and chiral synthesis of the Amaryllidaceae alkaloid, (+)-vittatine 1, is described; the quaternary carbon in 1 was generated by Claisen rearrangement of a cyclohexenol derived from D-glucose by way of a Ferrier's carbocyclisation reaction and a hexahydroindole skeleton was effectively constructed by intramolecular aminomercuration-demercuration, followed by Chugaev reaction.     

Ground state of the Si(0 0 1) surface revisited—is seeing believing?

The stable structure of clean Si(0 0 1) surface around 100 K is the c(4 × 2) arrangement constructed by buckled dimers. This structure was widely accepted as the ground state in 1990’s. The view was challenged at the beginning of 2000’s by the observations of a p(2 × 1) structure below 20 K with scanning tunneling microscopy (STM). Recent experimental studies confirm that the dimer is buckled below 30 K. Large tip–surface interaction, and/or tunneling current induced dynamical effect are now experimentally evident in the STM images at low temperatures. Moreover, a current induced structure transformation is discovered below 40 K even in the study by low energy electron diffraction. Dynamical electronic and vibrational effects are theoretically studied for accounting the observation of a p(2 × 1) structure below 20 K.
