A new class of chiral stationary phases (CSP) with peptide chiral selectors was prepared by solid-phase synthesis with a tert-butoxycarbonyl-L-amino acid on silica. The type of amino acid that is favorable for this class of CSP is discussed. Using the CSP with the phenylalanine peptide selector, the effect of peptide length on the enantioselectivity was investigated in normal-phase mode. The applicability of the CSP with a phenylalanine peptide to chiral ligand-exchange chromatography was also examined.

We investigated simultaneous determination of haloperidol (HAL), its three metabolites [reduced HAL (R-HAL), 3-(4-fluorobenzoyl)propionic acid (FBPA) and 4-(4-chlorophenyl)-4-hydroxypiperidine (CPHP)] and two related compounds [spiperone (SPI) and droperidol (DRO)] in phosphate-buffered saline using high-performance liquid chromatography (HPLC) coupled with dual ultraviolet detection (220 and 250 nm). Retention times of HAL, R-HAL, FBPA, CPHP, SPI and DRO were 16.8, 11.8, 10.2, 4.1, 12.6 and 8.3 min, respectively. Their lower limits of detection were 7.5, 14, 4.5, 12, 10 and 20 ng/mL in the same order. The coefficients of variation for their intra- and inter-day assays were less than 7.8 and 9.4%, respectively. Of the other centrally acting drugs, only amoxapine interfered with the peak of DRO. Using our procedure, the binding study of tested compounds to synthetic melanin, human serum albumin and alpha1-acid glycoprotein was performed by determining the unbound concentration to total concentration ratio. These results indicated that simultaneous assay of HAL, R-HAL, FBPA, CPHP, SPI and DRO in phosphate-buffered saline by HPLC equipped with dual ultraviolet detection is simple, sensitive and reproducible. Also, our assay system can be applied to the binding study of these compounds to synthetic melanin, human serum albumin and alpha1-acid glycoprotein.  相似文献

The Tl(NO3)3 (TTN) oxidation of the halogenated bisphenols, 14a, 14b, 24, and 43, effected regio-controlled cyclization to provide the corresponding diaryl ethers, 15a, 15b, 25, and 46. This investigation revealed a structural revision of eurypamide A as to possess (2″S,3″R,4″S)-configuration (47), along with the spectral data of pure 2 and 4, which were previously characterized in a mixture.  相似文献

Flexible dimeric host, 4,4'-azobenzenediamido-bis-{6-(2-dansyl-aminoethyl)6-deoxy--cyclodextrin}(beta; -1), has been synthesized in order to investigate itsfluorescent molecular sensory system transforming photoisomerization. Photoisomerizationof -1 was performed with photoirradiation by a 500W-Xelamp using a Corning 7-37 filter. Host beta; -1 showed puremonomer fluorescence, exhibiting a decrease in fluorescence intensity on accommodationof the guest, and the extent of fluorescence variation with a guest was employed to displaythe sensing ability of -1, in which the sensing parameter( I/I0)was used to describe the sensing ability. Host -1could detect chenodeoxycholic acid, ursodeoxycholic acid and hyodeoxycholic acid withhigh selectively. The behaviors of the appended moieties of -1during host-guest complexation were studied by induced circular dichroism (ICD) andfluorescence spectra and MM2 energy-minimized structure. The ICD and fluorescenceintensities of -1 decreased on the addition of a guest.The guest-induced variations in the ICD and fluorescence spectra, and MM2 energy-minimized structure suggested that the dansyl moieties of -1 worked as a hydrophobic cap when a host-guest complexation occurred.  相似文献

The total synthesis of alpha-C-mannosyltryptophan (C-Man-Trp), a naturally occurring C-glycosylamino acid, was achieved from a commercially available alpha-methyl-D-mannoside in 10 steps including the following key steps: the C-glycosidation of a mannose derivative with a stannylacetylene, Castro indole synthesis, and Sc(ClO4)3-promoted coupling with L-serine-derived aziridine carboxylate. The glucose- and galactose-analogues of C-Man-Trp were also synthesized in a similar manner. Conformational analyses of the synthesized C-glycosyltryptophan and its synthetic intermediate are briefly discussed.  相似文献

At 200°C, temperature-induced transformations of peroxide radicals from ? CF2CF(OO·)CF2? (radical I) to ? CF2CFCF2? (radical 1) and ? CF2CF2OO· (radical II) to ? CF2CF2 (radical 2) in vacuo, and from I to II in oxygen atmosphere were observed. In these thermal transformations, the evolution of CO2 with a small amount of CO and CF2O was observed. Further, measurements of the heat of crystallization of the treated polytetrafluoroethylene suggest that a reduction of polymer molecular weight occurs during the transformations from I to 1 and I to II.  相似文献

The effects of acetylene on the γ-radiation-induced polymerization of ethylene were studied from the viewpoint of the gaseous products and polymer structure. The experiments were carried out under a pressure of 400 kg/cm2; the temperature was 30°C; the does rate was 1.1 × 105 rad/hr; and the acetylene content was 0–20%. The solid polymer was obtained in the polymerization of ethylene containing 2.2% acetylene, while the monomer containing 19.7% acetylene gave a yellowish viscous oil. The polymer yield and molecular weight decreased remarkably with acetylene content. The main gaseous product was hydrogen, and trace amounts of butane, butene-1, butadiene-1,3, and benzene and its derivatives were also observed. The rate of formation of hydrogen was almost independent of acetylene content and there was no difference in acetylene contents before and after the irradiation was found. The infrared spectra of the polymers showed the presence of vinylidene, trans-vinylene, and terminal vinyl unsaturations, 1,4-disubstituted benzene, and carbonyl groups. The contents of trans-vinylene, terminal vinyl, and methyl groups increased with acetylene content, and that of vinylidene was independent of acetylene content. The monomer reactivity ratios of ethylene and acetylene were evaluated as 45.5 and 66.0, respectively. On the basis of the results, the effects of acetylene on the γ-radiation-induced polymerization of ethylene were discussed.  相似文献

The poly(1-trimethylsilyl-1-propyne) (PMSP) has the potential to be an important membrane gas separation material due to the fact that it has the highest gas permeability of all polymeric membranes. One problem with PMSP is a decrease in the gas permeability with age. In order to understand the aging processes, we studied the change in free volume and the molecular motions of the PMSP and its membranes modified with 1-phenyl-1-propyne (PP) structures; that is, a copolymer and a blend of PMSP and PPP. During aging, the unrelaxed volume of the PMSP membrane was relaxed, and the molecular motion of carbons dropped, suggesting that the decrease in the microvoids caused a tighter chain packing. The copolymer and blend membrane had stable permeability compared to the PMSP. In particular, the addition of a small amount of the PP structure provided excellent stability with high gas permeability. A decrease in the unrelaxed volume of modified membranes was hardly observed with age; however, the molecular motion of some carbons slightly changed. This change did not affect the gas permeability. In this case, a larger unrelaxed volume was probably a dominant factor in the gas permeation of the PMSP rich membranes relative to the molecular motion in the T1 measurement. © 1997 John Wiley & Sons, Inc.  相似文献
