Macromolecular engineering is presented as a tool to control the degradation rate and release rate of acidic degradation products from biomedical polyester ethers. Three different caprolactone/1,5-dioxepan-2-one (CL/DXO) copolymers were synthesized: DXO/CL/DXO triblock, CL/DXO multiblock, and random cross-linked CL/DXO copolymer. The relation of CL and DXO units in all three copolymers was 60/40 mol %. The polymer discs were immersed in phosphate buffer solution at pH 7.4 and 37 degrees C for up to 364 days. After different time periods degradation products were extracted from the buffer solution and analyzed. In addition mass loss, water absorption, molecular weight changes, and changes in thermal properties were determined. The results show that the release rate of acidic degradation products, a possible cause of acidic microclimates and inflammatory responses, is controllable through macromolecular design, i.e., different distribution of the weak linkages in the copolymers. 相似文献

Nuclear magnetic resonance (NMR) parameters are determined theoretically for the oxygen and hydrogen/deuterium nuclei of differently hydrogen-bonded water molecules in liquid water at 300 K. The parameters are the chemical shift, the shielding anisotropy, the asymmetry parameter of shielding, the nuclear quadrupole coupling constant, and the asymmetry parameter of the nuclear quadrupole coupling. We sample instantaneous configurations from a Car-Parrinello molecular dynamics simulation and feed nuclear coordinates into a quantum chemical program for the calculation of NMR parameters using density-functional theory with the three-parameter hybrid exchange-correlation (B3LYP) functional. In the subsequent analysis, molecules are divided into groups according to the number of hydrogen bonds they possess, and the full average NMR tensors are calculated separately for each group. The classification of the hydrogen-bonding cases is performed using a simple distance-based criterion. The analysis reveals in detail how the NMR tensors evolve as the environment changes gradually from gas to liquid upon increasing the number of hydrogen bonds to the molecule of interest. Liquid-state distributions of the instantaneous values of the NMR properties show a wide range of values for each hydrogen-bonding species with significant overlap between the different cases. Our study shows how local changes in the environment, along with classical thermal averaging, affect the NMR parameters in liquid water. For example, a broken or alternatively extra hydrogen bond induces major changes in the NMR tensors, and the effect is more pronounced for hydrogen or deuterium than for oxygen. Small‐angle and wide‐angle X‐ray scattering and anomalous small‐angle X‐ray scattering were used to investigate proton‐conducting membranes prepared by radiation‐induced styrene grafting and sulfonation of commercial poly(vinylidene fluoride) (PVDF‐g‐PS) films. The membranes retain the lamellar and highly oriented structure of the original PVDF films even through excessive grafting and sulfonation. The sulfonate groups aggregate in the central part of the amorphous layers, where they form a weakly ordered structure that does not show any preferred orientation. This structure is suggested to be lamellar with alternate metal‐sulfonated hydrate and PVDF‐g‐PS layers. The lamellar period is 15.1 Å. © 2000 John Wiley & Sons, Inc. A theoretical analysis on the contact vibration problem of a nip rolling system composed with two paper machine rolls is presented. It is modelled at the contact region by a two-degree-of-freedom (DOF) mass-spring-damper system. The rolling contact force in a case of viscoelastic polymer covered roll is formulated by integration of the contact stress. Regenerative chatter effects to the stability of the vibrations are considered. The perturbation technique is applied through the multi-scale method to calculate the nonlinear normal form of the governing equations to determine the stability behavior of the system. Spin diffusion between 13CH3 groups in solids is studied both theoretically and experimentally. It is shown to be dominated by mutual spin flip–flops of protons belonging to neighbouring methyl groups. Also nonmethyl protons may contribute significantly if present in the sample. The spin–rotational ground state of 13CH3 consists of 16 sublevels. When their populations are used to describe spin diffusion, eight population combinations are shown to be important, two of them corresponding to the 13C–proton and proton–proton intra-methyl magnetic dipolar energies, Dc and Dp, respectively. Spin-diffusion transitions modulate these combinations so that a further reduction to two sets of four combinations is possible, with no coupling between the sets. Coupled differential equations are derived to describe the time dependence of the combinations in each set. They are solved numerically and compared with experimental results on a single crystal of aspirin with 13C-labelled methyl groups at the carbon resonance. The 13C NMR induction signal was observed as a function of time after the preparation either at the carbon resonance (a two-pulse sequence) or at the proton resonance (proton saturation). Usually carbon spectra were computed first and then three of the mentioned population combinations were obtained from the individual spectral components. Some results on the time dependence of Dc were also obtained directly from the amplitude of the out-of-phase induction signal. Theoretical predictions are found to describe semiquantitatively the overall time dependence of these three combinations and especially their variation with different initial conditions, which are discussed in detail. Also the partial transfer of the magnetic dipolar energy between Dc and Dp is nicely explained. Reasons for discrepancies are discussed. 相似文献

Electrospray ionization Fourier transform ion cyclotron resonance mass spectrometry was used to investigate Ca(2+), Mg(2+), and La(3+) binding to bovine bone osteocalcin (OCN). OCN was shown to bind 3 mol Ca(2+) per mol protein. There was also evidence for the presence of four additional metal binding sites. Ca(2+) increased the formation of the OCN dimer. Mg(2+) bound to OCN to the same extent as Ca(2+) but did not induce the dimerization of OCN. La(3+) bound to a lesser extent than either Ca(2+) or Mg(2+) to OCN and, like Mg(2+), did not influence dimerization. Each Gla residue of OCN participates in Ca(2+) binding, whereas Mg(2+) binding may occur preferentially at sites other than Gla residues. This implies that the different natures of Ca(2+)- and Mg(2+)-containing OCN complexes influence the tendency of OCN to form a dimer. 相似文献

The efficient asymmetric synthesis of new chiral γ-chloro-α,β-diamino acid derivatives via highly diastereoselective Mannich-type reactions of N-(diphenylmethylene) glycine esters across a chiral α-chloro-N-p-toluenesulfinylimine was developed. The influence of the base, LDA or LiHMDS, used for the formation of the glycine enolates, was of great importance for the anti-/syn-diastereoselectivity of the Mannich-type reaction. The γ-chloro-α,β-diamino acid derivatives proved to be excellent building blocks for ring closure towards optically pure anti- and syn-β,γ-aziridino-α-amino esters, and subsequent ring transformation into trans-3-aminoazetidine-2-carboxylic acid derivatives and α,β-diamino-γ-butyrolactones. 相似文献
