In this work we demonstrated a facile method for the fabrication of C18 coordination polymer gel in a capillary, called stage-frit, which was efficiently applied to pack sub-2 μm C18 beads into the capillary by a high pressure bomb for the online separation of proteolytic peptides. The back pressure of the column with 10 cm × 75 μm i.d. is regularly lower than 170 bar at a flow rate of 300 nl/min, which could be operated on a common nanoLC system instead of nanoUPLC system due to the good permeability, low back pressure and high mechanical stress of the frit that will totally reduce the cost for the purchase of instrument. The stage-frit allows long-term continuous flow of the solvent and no significant beads loss or pressure instability was observed during the period. The repeatability of retention time for fifteen BSA tryptic peaks was found to be less than 1.08% (RSD) in six time nanoLC-ESI-MS/MS experiments. The average full width at half maximum (FWHM) of peptide peaks is 5.87 s. The sub-2 μm stage-frit nanoLC column showed better sensitivity than the commercial available for large scale proteomic analysis of total tissue proteins from human spleen. The number of identified peptides is approximately 0.4-fold and 0.2-fold higher than that obtained by utilizing commercial columns packed with 3 μm and 1.8 μm C18 materials, respectively. In the field of analytical chemistry, particularly the use of nanoLC systems, stage-frit nanoLC column offers a great potential for the separation of complex mixtures.  相似文献

The damping coefficient of capillary waves due to the evaporation-condensation process at the interface of the two phases of a fluid is evaluated. To highlight the mechanism of the effect of heat and mass transfer across the interface between regions of liquid and vapor, potential flow of incompressible fluids are assumed. Thus other mechanisms of damping are neglected. To fascilitate the analysis, the method of multiple-scale is employed in the analysis, even though the problem is linear.  相似文献

Aloe‐emodin, a natural polyphenolic anthraquinone, has shown various beneficial bioactivities in vitro. The aim of this study was to investigate the pharmacokinetics and metabolism of aloe‐emodin. Aloe‐emodin was intravenously and orally administered to rats. The concentrations of aloe‐emodin and rhein, a metabolite of aloe‐emodin, were determined by HPLC method prior to and after hydrolysis with β‐glucuronidase and sulfatase/β‐glucuronidase. The results showed that the systemic exposures of aloe‐emodin and its metabolites were ranked as aloe‐emodin glucuronides (G) > rhein sulfates (S) > aloe‐emodin > rhein and rhein G when aloe‐emodin was given intravenously. In contrast, when aloe‐emodin was administered orally, the parent form of aloe‐emodin was not absorbed per se, and the systemic exposures of its metabolites were ranked as aloe‐emodin G > rhein G > rhein. Tamm-Dancoff calculations including the complete 1p and 2s-1d shells are presented for non-normal parity states of nuclei with mass 15, 16 and 17. The basis is confined to ?ω excitations. Spurious states have been eliminated. The particle-particle and hole-hole interactions were taken from previous work. The particle-hole interaction was represented by 80 matrix elements from Kuo, eight of which were varied to fit 30 selected levels with a rms deviation of 310 keV. Spectroscopic factors and electromagnetic transition rates generally agree quite well with the experimental data. It is shown that the inclusion of the 1p32 and 1d32. shells is essential even at low excitation energies in these nuclei.  相似文献

Utilizing a sum rule in a spin-resolved photoelectron spectroscopic experiment with circularly polarized light, we show that the orbital moment in LaTiO3 is strongly reduced from its ionic value, both below and above the Ne el temperature. Using Ti L2,3 x-ray absorption spectroscopy as a local probe, we found that the crystal-field splitting in the t2g subshell is about 0.12-0.30 eV. This large splitting does not facilitate the formation of an orbital liquid.  相似文献

We present experimental demonstrations of spectral diversity filters with spherical beam volume holograms for multimodal multiplex spectroscopy. Major properties of filters under diffuse-light illumination are discussed. The comparisons of spectral diversity between the transmission geometry holograms and the reflection geometry holograms are also studied. The results show that there is a trade-off between the degree of the spatial coherence of the source and the spectral diversity of the filter. We also conclude that the reflection geometry holograms have better spectral diversity and less sensitivity to the spatial coherence of the source.  相似文献
