Collagen model peptide (Pro-Pro-Gly)10 has a triple helical structure and undergoes a thermal transition to a single random coil structure. The transition temperature of the analogous model peptides depends largely on amino acid substitution. Substitution of Pro by 4-hydroxyproline (Hyp) or 4-fluoroproline (fPro) has especially attracted attention because the position of substitution and chirality of the hydroxyl group or fluorine atom affect the transition temperatures. Here, we demonstrated that (4(S)-fPro-Pro-Gly)10 takes a triple helical structure, but (4(R)-fPro-Pro-Gly)10 exists in a single chain structure. This is not consistent with the case of Hyp substitution in our previous report where both (4(S)-Hyp-Pro-Gly)10 and (4(R)-Hyp-Pro-Gly)10 are in a single random coil state.  相似文献

A novel flow-coulometric detector integrating an immobilized uricase reactor and an electrolytic cell was fabricated and used for the determination of uric acid in human urine. The procedure is based on the measurement of the total charge with and without passing the sample through an enzyme reactor which allows the complete conversion of the electro-active uric acid in electro-inactive products. The amount of uric acid is linearly related to the difference between the two total charges. The current efficiencies for 1 × 10?4-1 × 10?3 M uric acid were found to be nearly 100% (r.s.d. < 1%).  相似文献

The electrolytic conductances of NaClO4, NaB(C6H5)4, (n-C4H9)4NClO4 and (n-C4H9)4NPF6 have been measured in methyl acetate at 25°C. A dominating feature of these electrolyte solutions is, as expected, strong ion pairing. In addition it is found that the formation of triple ions makes an important contribution to the overall molar conductivities. The data suggest that there are strong ion-solvent interactions leading to structure-enhanced (after Diamond) ion association and triple ion formation which has an exact analogy in the phenomena of salting-in. The effects of increasing solution permittivity and viscosity are discussed, particularly in regard to comparing two models of treating conductivity data. The first model includes ion pairs and triple ions, and the second model ignores triple ion formation ascribing the anomalous increase in molar conductivities to a decrease in the ion association equilibrium constant caused by increasing solution permittivity.  相似文献

The structural and thermodynamic behavior of the complex formation of eIF4E with either or both mRNA cap analogue (m7GTP, m7GpppA, or m7GpppG) and 4EBP1 has been investigated by spectroscopic measurements. Although the circular dichroism (CD) spectrum of eIF4E was little affected by the association with any cap analogue, the association constant of eIF4E with m7GpppA/G, estimated from the fluorescence quenching, was about 10 times larger than that with m7GTP. The van't Hoff analyses showed that the m7GpppA/G binding is enthalpy-driven with a large negative deltaH(o), and this is in contrast with the entropy-driven binding of m7GTP, where the positive deltaS(o) is large enough to overcome an increase of deltaH(o). This different behavior obviously originates in the interaction of the second nucleotide in m7GpppA with eIF4E, suggesting the importance of the nucleotide sequence linked to the m7Gppp terminal moiety, in addition to the specific interaction with the m7G base, for the recognition of mRNA cap structure by eIF4E. On the other hand, the CD spectra indicated that the binding of 4EBP1, an endogenous eIF4E-regulatory protein without having any defined secondary structure, shifted the m7GTP- or m7GpppA/G-bound eIF4E to an irregular structure, although such a structural change was not observed for eIF4E alone. The association constant of 4EBP1 with m7GTP- or m7GpppA/G-bound eIF4E was by two orders of magnitude larger than that with eIF4E alone. These results suggest the close interrelation in the supramolecular formation of 4EBP-eIF4E-mRNA cap structure.  相似文献

Di-isopropyl fluorophosphatase is contained in the giant axon of squid nerve tissue. This tissue can be coupled with a fluoride ion-selective electrode to produce an electrode selective to di-isopropyl fluorophosphate. The semilogarithmic calibration curve at pH 7.1 is linear over the range 2 × 10?6?7 × 10?3 M (RSD < 3% ) with a slope of about 44 mV/decade. The detection limit is 8 × 10?6 M. The electrode lifetime (with constant response characteristics) is about 16 days.  相似文献

A protocol of producing multiple polymeric masters from an original glass master mold has been developed, which enables the production of multiple poly(dimethylsiloxane) (PDMS)-based microfluidic devices in a low-cost and efficient manner. Standard wet-etching techniques were used to fabricate an original glass master with negative features, from which more than 50 polymethylmethacrylate (PMMA) positive replica masters were rapidly created using the thermal printing technique. The time to replicate each PMMA master was as short as 20 min. The PMMA replica masters have excellent structural features and could be used to cast PDMS devices for many times. An integration geometry designed for laser-induced fluorescence (LIF) detection, which contains normal deep microfluidic channels and a much deeper optical fiber channel, was successfully transferred into PDMS devices. The positive relief on seven PMMA replica masters is replicated with regard to the negative original glass master, with a depth average variation of 0.89% for 26-microm deep microfluidic channels and 1.16% for the 90 mum deep fiber channel. The imprinted positive relief in PMMA from master-to-master is reproducible with relative standard deviations (RSDs) of 1.06% for the maximum width and 0.46% for depth in terms of the separation channel. The PDMS devices fabricated from the PMMA replica masters were characterized and applied to the separation of a fluorescein isothiocyanate (FITC)-labeled epinephrine sample.  相似文献

A thermal lens microscope (TLM) detection of capillary electrophoresis (CE) utilizing microchip technology was developed. Fused-silica capillaries with an inner diameter of 50 microm were directly connected to a microchannel in a microchip. The detection limit by TLM was estimated as 2.8 x 10(-7) absorbance by measuring pure water. The detection limit of derivatized amino acids determined by CE-TLM was estimated as 2.4 x 10(-8) M, which was 100 times lower than that of conventional absorbance detection.  相似文献

A regio- and chemoselective direct method for generation of functionalized aromatic aluminum compounds through deprotonative directed ortho-alumination using triisobutyl(tetramethylpiperidino)aluminate (iBu3Al(TMP)Li), prepared by mixing of triisobutylaluminum (iBu3Al) and lithium tetramethylpiperidide (LTMP) in THF, has been developed. Deprotonative alumination of various functionalized benzenes with the use of iBu3Al(TMP)Li proved effective for the direct generation of various ortho-functionalized aluminum aromatic and heteroaromatic derivatives, particularly those with electrophilic functional groups such as cyano, amide, and halogen. Direct alumination, followed by electrophilic trapping (with I2), provided a convenient preparative method for 1,2- or 1,2,3-multisubstituted aromatic compounds. The functionalized aromatic aluminate intermediate also was found to undergo copper- and palladium-catalyzed C-C bond-forming reactions very efficiently and highly regio- and chemoselectively.  相似文献

As in the case of vinylhydroquinone (I), its alkyl-substituted derivative, 2-methyl-5-vinylhydroquinone (II) was found to copolymerize with methyl methacrylate by tri-n-butylborane in cyclohexanone at 30°C. II was prepared from the O,O′-bisether compound, 2-methyl-5-vinyl-O,O′-bis(1′-ethoxyethyl)hydroquinone (III). The monomer reactivity ratios (M2 = II) were determined to be r1 = 0.37 and r2 = 0. No homopolymerization proceeded under the same conditions. Ordinary free-radical initiators, such as azobisisobutyronitrile and benzoyl peroxide, were not effective in the homopolymerization of II. 1:1 Copolymers were obtained from II and maleic anhydride by using tri-n-butylborane as an initiator. The copolymers exhibited no definite melting range and decomposed at 370–375°C endothermally (DSC). The polymerization behavior of III was also investigated. Although tri-n-butylborane did not initiate the homopolymerization of the monomer, azobisisobutyronitrile was capable of initiating the homopolymerization and copolymerization of III. The monomer reactivity ratios (M1 = styrene) were determined to be r1 = 0.83 and r2 = 0.18. The ratios gave the following Q and e values; Q = 0.15 and e = ?2.2.  相似文献
