SciFinder-guided rational design of fluorescent carbon dots for ratiometric monitoring intracellular pH fluctuations under heat shock

Intracellular pH plays a significant role in various biological processes, including cell proliferation, apoptosis, metabolism, enzyme activity and homeostasis. In this work, a novel design strategy for the preparation of pH responsive carbon dots (CDs-pH) for ratiometric intracellular imaging was reported. By using SciFinder database, fluorescent CDs-pH with the required pK_a value of 6.84 were rationally designed, which is vital important for precise sensing of intracellular pH. As a result, the synthesized CDs-pH demonstrated robust ability to test pH fluctuations within the physiological range of 5.4-7.4. The CDs-pH was further utilized for fluorescent ratiometric imaging of pH in living HeLa cells, effectively avoided the influence of autofluorescence from native cellular species. Moreover, real-time monitoring of intracellular pH fluctuation under heat shock was successfully realized. This SciFinder-guided design strategy is simple and flexible, which has a great potential to be used for the development of other types of CDs for various applications.     

火焰原子吸收光谱法测定天然水中微量铁的形态

研究了用抗坏血酸为还原剂,使二价铁离子与邻菲啰啉(phen)形成螯合物,用硝基苯萃取,火焰原子吸收光谱法测定有机相中的铁。方法检出限为1.7ng·ml~(-1),回收率为98.0％～109.6％,线性范围为0～10.0μg·ml~(-1)(原水相),线性相关系数为0.9994。测定结果与ICP-AES法结果对照相对误差小于9.46％。     

An organic-phase optical phenol biosensor coupling enzymatic oxidation with chemical reduction

This paper presents a recycle amplification optical biosensor to monitor phenol in hydrophobic organic solvents. Tyrosinase was first immobilised by entrapping it in a copolymer membrane of poly(vinyl alcohol)-hydroxyethyl carboxymethylcellulose doped with octadecylsilica particles. The biosensor was then constructed by co-mixing small particles of the immobilised tyrosinase with the adduct of L-ascorbic acid-poly(vinyl alcohol) (AsA-PVA) in conjunction with an optical oxygen transducer. The biosensor was characterised by its amplifying response to phenol, stable biocatalytic activity of entrapped-tyrosinase, free of interference from o-quinone polymerisation, and large water buffer capacity in hydrophobic organic solvents. The working range of the biosensor to phenol was 0.08-40 mmol dm(-3) in the flow mode. The response times (95%) of the biosensor were 4-7 min for phenol. The operational lifetime was more than 40 assays and the shelf lifetime of the biosensor was longer than 3 months. The biosensor has been successfully applied to quantify the phenol contents in some commercial ointment samples.     

5′-鸟苷酸与茜素红S的反应及其在分析化学中的作用

采用UVVis光谱法研究了茜素红S(ARS)与5′鸟苷酸(5′GMP)在pH4.80的弱酸性缓冲溶液中生成络合物的结合反应。与试剂比较,络合物的最大吸收峰红移92nm,测得络合物和表观摩尔吸光系数为ε=1.3×104L·mol-1·cm-1;最大结合数n=10;浓度线性范围0.2～16mg/L;检出限为6.1×10-8mol/L。研究了ARS与5′GMP是分子间作用力的结合反应,并对时间、温度、离子强度对结合反应的影响,以及无机物、生物物质对反应体系的干扰情况进行了初步研究。     

Efficient Debromination of Vicinal α，β—Dibromo Carboxylic Acid Derivatives with the Sm／HOAc System

The α，β vicinal dibrome carboxylic acid and its derivatives were debrominated with Sm/HOAc system to afford the corresponding cinnamic acid and its derivatives in good yields under mild conditions.     

Anomalies in the asymmetric hydroboration of olefins with the 11 adduct of (+)-α-pinene and BH3 · THF

The hydroboration of cis-2-butene and cis-3-hexene with pure diisopinocampheylborane (IPC₂BH) and monoisopinocampheylborane (IPCBH₂), both prepared from (+)-α-pinene, were studied. In contrast to IPC₂BH, which yielded R-(?)-2-butanol and R-(?)-3-hexanol in 98.4 and 94.5% optical purities, IPCBH₂ yielded S-(+)-2-butanol and S-(+)-3-hexanol in 23.6 and 19.7% optical purifies respectively. PMR examination of the methanolyzed products were utilized to establish the species present in the freshly prepared and aged $\text{1}\text{1}$ adduct from (+)-α-pinene and BH₃ · THF. The results confirm the interpretation previously advanced for the major differences realized in asymmetric hydroboration with the fresh and aged $\text{1}\text{1}$ adducts.     

Indium-mediated one-pot reductive conversion of nitroarenes to N-arylacetamides

N-Arylacetamides were prepared in excellent yields from nitroarenes in the presence of acetic anhydride, acetic acid and indium by a one-pot procedure.     

丽春红G-高碘酸钾催化光度法测定痕量锇

以Os(Ⅳ )催化高碘酸钾氧化丽春红G(PG)的褪色反应为基础 ,在 90℃加热 1 2min和 5 0 0nm波长处采用固定时间法测定丽春红G吸收值的降低 ,建立了测定痕量锇 (Ⅳ )的新方法。锇 (Ⅳ )的质量浓度在 0～ 0 48μg/L范围内呈良好的线性关系 ,检出限为 6 2× 1 0 - 9mg/L。对 0 4μg/L锇 (Ⅳ )测定的相对标准偏差为 0 71 % (n =1 1 )。催化反应对Os(Ⅳ )和丽春红G均为一级反应 ,催化反应的表观活化能为 78 80kJ/mol。所提出的方法在蒸馏分离后已应用于某些岩矿及冶金产品中锇的测定     

Single-strand conformation polymorphism-based analysis of mitochondrial cytochrome c oxidase subunit 1 reveals significant substructuring in hookworm populations

Sequence heterogeneity in a portion of the mitochondrial cytochrome c oxidase subunit 1 gene (pcox1) was measured for the hookworms, Ancylostoma caninum from Australia, A. duodenale from China, and Necator americanus from China and Togo using single-strand conformation polymorphism (SSCP) analysis combined with DNA sequencing. The pcox1 sequences were characterised for individual nematodes displaying genetic variation within each of the three species, and those were compared with pcox1 sequences of four other species of hookworm. While intraspecific variation in the pcox1 sequence ranged from 0.5 to 8.6% for A. caninum, 0.3 to 3.3% forA. duodenale, and 0.3 to 4.3% for N. americanus, interspecific differences varied from 4.8 to 12.9%. Sequence data also provided information on nucleotide compositions and substitution patterns. Genetically distinct groups were detected within A. caninum and A. duodenale, indicating significant population substructuring within these species. Also, N. americanus individuals from China all differed from those from Togo at four nucleotide positions, supporting a previous proposal (based on ribosomal DNA sequence data) that N. americanus may represent a species complex. The findings indicated the value of pcox1 sequence data and the mutation scanning approach for studying the genetic structures of hookworm populations, which should have important epidemiological relevance.
