Poly(lactic-co-glycolic) acid (PLGA) is an important copolymer used in drug delivery platforms where controlled release is required. In this work we investigated the in vitro degradation of four PLGA copolymers with L/G molar compositions of 50/50, 65/35, 75/25 and 95/5. ATR-IR and Raman spectroscopies were used to differentiate and quantify the degradation rates of glycolic and lactic units. Both techniques were used to determine the polymer composition as a function of degradation time and the degradation rate constants for the hydrolysis of glycolic and lactic units were calculated using a 1st order kinetics approach. Our results revealed a two stage process for the degradation of PLGA cast films in PBS in agreement with our previous work. The degradation rate constant for glycolic unit was found to be 1.3 times higher than for lactic units. In addition the degradation rate constants for L and G units were shown to decrease proportionally with increasing initial lactic content of the copolymer used to prepare the films.  相似文献

The system constants of the solvation parameter model are used to prepare system maps for the retention of small neutral compounds on phenylhexylsiloxane- and pentafluorophenylpropylsiloxane-bonded superficially porous silica stationary phases (Kinetex Phenyl-Hexyl and Kinetex F5) for aqueous mobile phases containing 10–70% (v/v) methanol or acetonitrile. Electrostatic interactions (cation exchange) are important for the retention of weak bases for acetonitrile–water mobile phases, but virtually absent for the same compounds for methanol–water mobile phases. The selectivity of the Kinetex Phenyl-Hexyl stationary phase for small neutral compounds is similar to an octadecylsiloxane-bonded silica stationary phase with similar morphology Kinetex C-18 for both methanol–water and acetonitrile–water mobile phase compositions. The Kinetex Phenyl-Hexyl and XBridge Phenyl stationary phases with the same topology but different morphology are selectivity equivalent, confirming that solvation of the interphase region can be effective at dampening selectivity differences for modern stationary phases. Small selectivity differences observed for XTerra Phenyl (different morphology and topology) confirm previous reports that the length and type of space arm for phenylalkylsiloxane-bonded silica stationary phases can result in small changes in selectivity. The pentafluorophenylpropylsiloxane-bonded silica stationary phase (Kinetex F5) has similar separation properties to the phenylhexylsiloxane-bonded silica stationary phases, but is not selectivity equivalent. However, for method development purposes, the scope to vary separations from an octadecylsiloxane-bonded silica stationary phase (Kinetex C-18) to “phenyl phase” of the types studied here is limited for small neutral compounds. In addition, selectivity differences for the above stationary phases are enhanced by methanol–water and largely suppressed by acetonitrile–water mobile phases. For bases, larger selectivity differences are possible for the above stationary phases if electrostatic interactions are exploited, especially for acetonitrile-containing mobile phases.  相似文献

Compared with an octadecylsiloxane-bonded silica stationary phase (Kinetex C18) retention is reduced due to a less favorable phase ratio for both the octylsiloxane-bonded and diisobutyloctadecylsiloxane-bonded silica stationary phases while selectivity differences are small and solvent dependent. Selectivity differences for neutral compounds are larger for methanol–water but significantly suppressed for acetonitrile–water mobile phases. The selectivity differences arise from small changes in all system constants with solute size and hydrogen-bond basicity being the most important due to their dominant contribution to the retention mechanism. Exchanging the octadecylsiloxane-bonded silica column for either the octylsiloxane-bonded or diisobutyloctadecylsiloxane-bonded silica column affords little scope for extending the selectivity space and is restricted to fine tuning of separations, and in some cases, to obtain faster separations due to a more favorable phase ratio. For weak bases larger differences in relative retention are expected with acetonitrile–water mobile phases on account of the additional cation exchange interactions possible that are absent for the octadecylsiloxane-bonded silica stationary phase.  相似文献

Stimuli-responsive soft materials enable controlled release of loaded drug molecules and biomolecules. Controlled release of potent chemotherapeutic or immunotherapeutic agents is crucial to reduce unwanted side effects. In an effort to develop controlled release strategies that can be triggered by using Cerenkov luminescence, we have developed polymer hydrogels that can release bovine serum albumin and immunoglobulin G by using light (254 nm–375 nm) as a trigger. We describe the synthesis and photochemical characterization of two light sensitive phenacyl bis-azide crosslinkers that are used to prepare transparent self-supporting hydrogel patches. One crosslinker was designed to optimize the overlap with the Cerenkov luminescence emission window, bearing an π-extended phenacyl core, resulting in a high quantum yield (14 %) of photocleavage when irradiated with 375 nm light. We used the extended phenacyl crosslinker for the preparation of protein-loaded dextran hydrogel patches, which showed efficient and selective dosed release of bovine serum albumin or immunoglobulin G after irradiation with 375 nm light. Cerenkov-triggered release is as yet inconclusive due to unexpected side-reactivity. Based on the high quantum yield, efficient release and large overlap with the Cerenkov window, we envision application of these photosensitive soft materials in radiation targeted drug release.  相似文献

With recent advances in the computer-aided synthesis planning (CASP) powered by data science and machine learning, modern CASP programs can rapidly identify thousands of potential pathways for a given target molecule. However, the lack of a holistic pathway evaluation mechanism makes it challenging to systematically prioritize strategic pathways except for using some simple heuristics. Herein, we introduce a data-driven approach to evaluate the relative strategic levels of retrosynthesis pathways using a dynamic tree-structured long short-term memory (tree-LSTM) model. We first curated a retrosynthesis pathway database, containing 238k patent-extracted pathways along with ∼55 M artificial pathways generated from an open-source CASP program, ASKCOS. The tree-LSTM model was trained to differentiate patent-extracted and artificial pathways with the same target molecule in order to learn the strategic relationship among single-step reactions within the patent-extracted pathways. The model achieved a top-1 ranking accuracy of 79.1% to recognize patent-extracted pathways. In addition, the trained tree-LSTM model learned to encode pathway-level information into a representative latent vector, which can facilitate clustering similar pathways to help illustrate strategically diverse pathways generated from CASP programs.

Although biological imaging is mostly performed in aqueous media, it is hardly ever considered that water acts as a classic fluorescence quencher for organic fluorophores. By investigating the fluorescence properties of 42 common organic fluorophores recommended for biological labelling, we demonstrate that H2O reduces their fluorescence quantum yield and lifetime by up to threefold and uncover the underlying fluorescence quenching mechanism. We show that the quenching efficiency is significantly larger for red-emitting probes and follows an energy gap law. The fluorescence quenching finds its origin in high-energy vibrations of the solvent (OH groups), as methanol and other linear alcohols are also found to quench the emission, whereas it is restored in deuterated solvents. Our observations are consistent with a mechanism by which the electronic excitation of the fluorophore is resonantly transferred to overtones and combination transitions of high-frequency vibrational stretching modes of the solvent through space and not through hydrogen bonds. Insight into this solvent-assisted quenching mechanism opens the door to the rational design of brighter fluorescent probes by offering a justification for protecting organic fluorophores from the solvent via encapsulation.

The reaction of [PtCl2(COD)] (COD=1,5-cyclooctadiene) with diisopropyl-2-(3-methyl)indolylphosphine (iPr2P(C9H8N)) led to the formation of the platinum(ii ) chlorido complexes, cis-[PtCl2{iPr2P(C9H8N)}2] ( 1 ) and trans-[PtCl2{iPr2P(C9H8N)}2] ( 2 ). The cis-complex 1 reacted with NEt3 yielding the complex cis-[PtCl{κ2-(P,N)-iPr2P(C9H7N)}{iPr2P(C9H8N)}] ( 3 ) bearing a cyclometalated κ2-(P,N)-phosphine ligand, while the isomer 2 with a trans-configuration did not show any reactivity towards NEt3. Treatment of 1 or 3 with (CH3)4NF (TMAF) resulted in the formation of the twofold cyclometalated complex cis-[Pt{κ2-(P,N)-iPr2P(C9H7N)}2] ( 4 ). The molecular structures of the complexes 1–4 were determined by single-crystal X-ray diffraction. The fluorido complex cis-[PtF{κ2-(P,N)-iPr2P(C9H7N)}{iPr2P(C9H8N)}] ⋅ (HF)4 ( 5 ⋅ (HF)4) was formed when complex 4 was treated with different hydrogen fluoride sources. The Pt(ii ) fluorido complex 5 ⋅ (HF)4 exhibits intramolecular hydrogen bonding in its outer coordination sphere between the fluorido ligand and the NH group of the 3-methylindolyl moiety. In contrast to its chlorido analogue 3 , complex 5 ⋅ (HF)4 reacted with CO or the ynamide 1-(2-phenylethynyl)-2-pyrrolidinone to yield the complexes trans-[Pt(CO){κ2-(P,C)-iPr2P(C9H7NCO)}{iPr2P(C9H8N)}][F(HF)4] ( 7 ) and a complex, which we suggest to be cis-[Pt{C=C(Ph)OCN(C3H6)}{κ2-(P,N)-iPr2P(C9H7N)}{iPr2P(C9H8N)}][F(HF)4] ( 9 ), respectively. The structure of 9 was assigned on the basis of DFT calculations as well as NMR and IR data. Hydrogen bonding of HF and NH to fluoride was proven to be crucial for the existence of 7 and 9 .  相似文献
