Benzoic acid priming of the enterocin and actinorhodin type II polyketide synthase complexes was accomplished in vitro via an unprecedented type II nonribosomal peptide synthetase-like mechanism involving the benzoate:acyl carrier protein (ACP) ligase EncN and the ACP EncC. The transfer of the aryl acid to the ACP is ATP-dependent, yet coenzyme A-independent, as characterized with radiolabeled substrates and protein mass spectrometry. Subsequent transport of the ACP-bound aryl group to the native enterocin and the aberrant actinorhodin ketosynthase chain length factor heterodimers was further demonstrated, thereby demonstrating the potential of this biocatalyst for engineering diverse aryl-primed aromatic polyketide agents.  相似文献

Theranostic probes provide both therapeutic and diagnostic imaging capabilities in one molecule and show significant promise for use in magnetic resonance imaging (MRI) examinations. The present study describes for the first time the synthesis and utility of nitroxide‐based contrast agents exhibiting a nonsteroidal anti‐inflammatory drug effect. The target theranostic probes were prepared by connecting the carboxyl group of ibuprofen or ketoprofen to the hydroxyl group of 3‐hydroxymethyl‐2,2,5,5‐tetramethylprrolidine‐1‐oxyl by a condensation reaction in the presence of dicyclohexylcarbodiimide and 4‐dimethylaminopyridine in dichloromethane. MRI of mouse heads after administration of either synthesized theranostic probe indicated that the probes enter the brain by passing through the blood–brain barrier (BBB), resulting in T1 contrast enhancement in mouse brain. This enhancement persisted for the duration of the half‐life of about 40 min, which is longer than that obtained by most of pyrrolidine nitroxide molecules. The therapeutic capacities of these theranostic probes were examined using a lipopolysaccharide (LPS)‐induced brain inflammation model. The production of nitric oxide, an inflammation marker in septic mouse brain induced by LPS, was remarkably inhibited by the addition of either synthesized probe, indicating that they also act as anti‐inflammatory drugs. The present results indicate that nitroxide‐based theranostic probes act as both BBB‐permeable redox‐sensitive contrast agents and as an anti‐inflammatory drug in septic mouse brain. To develop monodisperse amphiphilic polymer particles on which a large amount of lipase could be immobilized, we performed seed polymerizations of glycidyl methacrylate and allyl methacrylate in the presence of nonpolar inert and polar inert solvents. The amphiphilic porous polymer particles, which had both hydrophilic guanidino groups and hydrophobic stearoyl groups, were synthesized in the presence of n‐decane and had a large amount of macropores with diameters of 50–1000 nm. The amount of lipase immobilized on the amphiphilic particles synthesized in the presence of n‐decane was 3.85 times that of the lipase immobilized on the amphiphilic particles synthesized in the absence of a solvent. The immobilized lipase prepared with the amphiphilic particles synthesized in the presence of n‐decane exhibited a high transesterification activity in n‐hexane and could be used repeatedly without a considerable activity loss. © 2002 Wiley Periodicals, Inc. The salting-out effect has been characterized on the basis of the relative peak intensity of silica species, observed by FAB-MS (fast atom-bombardment mass spectrometry) in solutions of sodium chloride, sodium nitrate, sodium sulfate, calcium chloride, lithium chloride and magnesium chloride. A critical change in the peak intensity ratios of the linear and cyclic tetramers of silica against the sodium ion (Na+) concentrations was observed at Na+ concentration between 0.1 and 1 mol⋅dm−3. The degrees of the changes of these peak intensity ratios increased in the order NaNO3 < Na2SO4 < NaCl. In CaCl2 solutions, these peak intensity ratios changed significantly at Ca2+ concentrations between 0.05 and 0.5 mol⋅dm−3. The salting-out effect observed is the total change in the concentration of silica brought about by complex factors, such as the changing solubility of silicate complexes, the increases in the concentrations of different kinds of soluble silicate complexes induced by changes in the hydrophobicity and hydrophilicity of the solution, and the contribution of hydrolysis.  相似文献

To characterize the photocatalytic activity of TiO2 via solid-state reaction, the relationship between the physicochemical properties and photocatalytic activity of TiO2 was investigated and estimated from the results of photodegradation of nisoldipine. The photodegradation of nisoldipine was significantly enhanced by addition of TiO2. Two degradation products, nitroso-phenylpyridine derivative and nitro-phenylpyridine derivative, were formed. The degree of photocatalytic activity of TiO2 was quite different between the various types of TiO2 investigated, even when the crystalline phase was the same. As a result of the investigations into the relationship between the photocatalytic activity and physicochemical properties of TiO2, it was found that for the rutile form the photocatalytic activity has good correlation with specific surface area of TiO2, but poor correlation with water loss on drying of TiO2. However, for the anatase form, the photocatalytic activity has good correlation with water loss on drying of TiO2, but poor correlation with specific surface area. Moreover, it was found that the crystallinity of TiO2 has a moderate correlation with the photocatalytic activity of both crystal forms of TiO2. These results suggest that a degree of photocatalytic activity of TiO2 depends on the various physicochemical properties of each type of TiO2 investigated.  相似文献

A flow injection-catalytic spectrophotometric method using a serial flow cell was proposed for the successive determination of trace amounts of copper and iron. This method is based on the oxidation coupling of p-anisidine with N,N-dimethylaniline in the presence of hydrogen peroxide to form a dye, which has an absorption maximum at 740 nm. In this indicator reaction, ligands such as 1,10-phenanthroline (phen) and diphosphate were achieved to improve the sensitivity and selectivity. Under the optimal experimental conditions, the determinable ranges were 0.05-5 ppb for copper and 0.5 - 100 ppb for iron, respectively. The RSDs (n = 10) were 0.78% for 0.5 ppb copper(II) and 0.5% for 200 ppb iron(III). The sample throughput was 30 h(-1). The present flow-injection method was applied to the determination of copper and iron in standard river water, tap water, and other natural water samples, and also to the analysis of labile and inert complexes in synthesized samples containing humic acid with copper(II) or iron(III).  相似文献

When the sodium ion (Na+) concentration is increased above 0.5 mol-dm−3 (M), the concentrations of dissolved silica in aqueous sodium chloride (NaCl) and sodium nitrate (NaNO3) solutions decrease because of the salting out effect. On the other hand, the concentration of the dissolved silica in aqueous sodium sulfate (Na2SO4) solutions increases monotonously as the concentration of Na+ is increased above 0.5 M. The purpose of this study is to determine the reasons why the salting-out effect is not observed in Na2SO4 solutions. FAB-MS (Fast Atom Bombardment Mass Spectrometry) was used to sample directly the silica species dissolved in aqueous Na2SO4, NaCl, and NaNO3 solutions. In the FAB-MS spectra of these solutions, the peak intensity ratios of the linear tetramer to the cyclic tetramer largely increased for Na+ concentrations between (0.1 and 1) M. This shows that some characteristics of the Na2SO4 solutions are similar to those of the NaCl and NaNO3 solutions. In Na2SO4 solutions, however, when the concentration of Na+ is higher than 1 M, the peak intensity of the dimer is much higher than those of the other silicate complexes. In Na2SO4 solutions, the SO42− ion undergoes partial hydrolysis to form HSO4 and OH is produced. In particular, in the range where the concentration of SO42− is high, the pH of the solution increases slightly. This higher pH yields more dimers from the hydrolysis of silicate complexes. This increase in dimer production agrees with the observation that silica dissolves in sodium hydroxide (NaOH) solutions mainly as a dimer when the concentration of NaOH is less than 0.1 M. In Na2SO4 solutions at high concentrations, a salting-out effect is not observed for silica. This is due to the increase in the concentration of OH, which accelerates the hydrolysis of silica and results in dimer formation.  相似文献
