Amphezonol A (1), a novel polyhydroxyl linear carbon-chain metabolite, has been isolated from the cultured marine dinoflagellate Amphidinium sp., which was isolated from an Okinawan marine acoel flatworm Amphiscolops sp. The structure of 1 was elucidated by detailed analyses of 2D NMR spectra. Amphezonol A (1) possesses one tetrahydrofuran ring, two tetrahydropyran rings, and twenty-one hydroxyl groups on C60-linear aliphatic chain with one exo-methylene and one methyl branch. Amphezonol A (1) exhibited a modest inhibitory activity against DNA polymerase α.  相似文献

A biphenyl-based axially chiral amino acid (S)-2 has been designed and synthesized. The new amino acid (S)-2 has been found to be a more efficient catalyst than (S)-1 in the direct asymmetric aldol reaction of acetone with aldehydes. For instance, the use of only 0.1 mol % of (S)-2 was sufficient to complete the reaction between acetone and 4-nitrobenzaldehyde, giving the corresponding aldol adduct in good yield with an excellent enantioselectivity.  相似文献

[reaction: see text] Catalytic asymmetric allylation of 3,4-dihydro-6,7-dimethoxyisoquinoline was carried out using allyltrimethoxysilane in the presence of Cu(I) and tol-BINAP. A titanium BINOLate catalyzed asymmetric 1,3-dipolar cycloaddition reaction between alpha-substituted acroleins and alkyl diazoacetates has been developed. With this methodology in hand, chiral 2-pyrazolines containing a quaternary stereogenic center were obtained in high to excellent enantioselectivities (up to 95% ee). The synthetic utility of the optically enriched 2-pyrazoline thus obtained was demonstrated in the short synthesis of manzacidin A.  相似文献

The effect of lactose carrier surface property on the inhalation profile of dry powder inhaler (DPI) was evaluated using a micronized drug (Compound A) by inverse gas chromatography (IGC). Mechanofusion with magnesium stearate (Mg-St) or sucrose stearate increased the fine particle fraction (FPF), considered to be due to decrease in the interaction between Compound A and the lactose carrier. The effect of Compound A concentration on FPF was smaller in mechanofusion-processed lactose compared to intact lactose, especially when processed with Mg-St. The relationship between the IGC parameters of the lactose and FPF was also investigated. FPF increased as both the dispersive component of the surface energy and acidity similarity between the lactose carriers and Compound A increased. Although further investigation is necessary, it could be suggested that acidity similarity decreases the interaction between Compound A and lactose, thus contributing to the increase in the FPF. In conclusion, (1) mechanofusion with Mg-St or sucrose stearate could be an effective method to improve FPF of a DPI drug formulation; (2) IGC would be a valuable method to investigate the interaction between a drug and the DPI carrier; and (3) a relationship between surface acidity and inhalation profile was suggested.  相似文献

We demonstrated that the two-photon excitation efficiency of perylene in chloroform solution as well as that of crystalline perylene was dramatically increased by optimizing the shape of intense femtosecond laser pulses of a regenerative amplifier output. The efficiency was three times higher than for an unshaped single femtosecond pulse with the pulse width of shorter than 50 fs. The pulse shape optimized for the solution sample was a pulse train with a repetition frequency of about 340 cm(-1), and the pulse shape optimized for crystalline perylene was very similar. These results supported our previous findings on alpha-perylene crystals using weak femtosecond pulses from a mode-locked laser oscillator [T. Okada et al. J. Chem. Phys. 121, 6385 (2004)]. Furthermore, it was confirmed that the shaped pulse optimized for the liquid sample could also increase the two-photon excitation efficiency of alpha-perylene crystals and vice versa. We concluded that the mechanism for the increase in excitation efficiency of perylene in chloroform was almost the same as that for alpha-perylene crystal, and that the efficiency increased mainly through intramolecular dynamical processes. Processes involving intermolecular interactions and/or energy states delocalized over the crystal cannot play the major role.  相似文献
