Electrospray ionization mass spectrometry (ESI-MS) is used to probe the binding of small anions to the macrocycle of bambus[6]uril. For the halide ions, the experimental patterns suggest F(-) < Cl(-) < Br(-) < I(-), which is consistent with the order of anion binding found in the condensed phase. Parallel equilibrium studies in the condensed phase establish the association constants of halide anions and bambus[6]uril in mixed solvents. A detailed analysis of the mass spectrometric data is used to shed light on the correlations between the binding constants in the condensed phase and the ion abundances observed using ESI-MS. From the analysis it becomes apparent that ESI-MS can indeed represent the situation in solution to some extent, but the sampling in the gas-phase experiment is not 1:1 compared to that in solution. 相似文献

Vibrational energy flow and conformational transitions following excitation of the OH stretching mode of the most stable conformer of glycine are studied by classical trajectories. "On the fly" simulations with the PM3 semiempirical electronic structure method for the potential surface are used. Initial conditions are selected to correspond to the ν=1 excitation of the OH stretch. The main findings are: (1) An an equilibrium-like ratio is established between the populations of the 3 lowest-lying conformers after about 10 picoseconds. (2) There is a high probability throughout the 150 ps of the simulations for finding the molecule in geometries far from the equilibrium structures of the lowest-energy conformers. (3) Energy from the initial excited OH (ν=1) stretch flows preferentially to 5 other vibrational modes, including the bending motion of the H atom. (4) RRK theory yields conformational transition rates that deviate substantially from the classical trajectory results. Possible implication of these results for vibrational energy flow and conformational transitions in small biological molecules are discussed. 相似文献

The influence of perrhenate concentration, the concentration of the reducing agent and pH of the reaction mixture on the yield of perrhenate reduction were studied to find a possibility to decrease the stannous chloride concentration in the reaction mixture without negative changes on the yield of perrhenate reduction. Britton-Robinson buffer solutions were selected as the background electrolytes because of their buffering capacity in a wide pH interval. The highest degree of perrhenate reduction was obtained at pH 2 at perrhenate concentrations ranging from 10(-4) to 10(-3) mol/L. The stability of reduced rhenium against pH change from 2 to 5.5 and against dilution of rhenium in the reaction mixture to the concentration suitable for the application in radiotherapy were studied as well. The Platinum Group Elements (PGEs) used in automotive catalytic converters are partly emitted into the air during use and can enter the human respiratory system. Due to the increasing use of automotive catalytic converters, the importance of this problem cannot be overlooked.The goal of this investigation was to determine the concentration of Pt in the urine of individuals occupationally exposed to urban air with heavy traffic. Sector field inductively coupled plasma mass spectrometry (SF–ICP–MS) was used for determination of Pt in the urine of tram drivers. 38 and 34 subjects were investigated in Vienna and Budapest, respectively. Samples were taken from the tram drivers both before and after the shift.The results for Pt were compared to those from a previous study performed by our team. The comparison showed that the concentration medians were 4 times higher than the previous ones. Moreover, the values in Budapest were about twice as high as those from Vienna. A partly significant change could be observed between the two sets of data: before, and after the shift. 相似文献

Mass spectrometry (MS) has become a powerful tool for the quantitative analysis of complex protein samples. A high-throughput strategy for the comparative analysis of multiple protein samples with high complexity becomes more and more important. Two strategies, spectral count and peak intensity, for label-free MS analysis of prefractionated complex mixtures have been described recently to be useful for quantitation. Here we compare both strategies for rapid and quantitative 1-D shotgun LC/MS/MS analyses of highly complex protein mixtures using silica-based monolithic columns. First, we validated linearity and sensitivity of these methods by spiking varying amounts of an internal standard protein in a complex plant protein extract. Secondly, quantitative data of proteins of Medicago truncatula nodules were visualized with independent components analysis using data either obtained from spectral count or peak integration performed with commercial software. Spectral count showed apparent advantages over peak integration because several peptides per protein are automatically averaged, the linear dynamic range of quantitation increases in complex matrices and the number of quantified proteins surpasses the number of proteins using peak integration. Thus, for the need of rapid comparative analysis of highly complex protein samples, spectral count enables sample pattern recognition and identification of biomarkers in nongel based proteomic studies. 相似文献
