The relationship between free radicals and acute or chronic inflammation has been well established. We have previously reported the significant antioxidant activity of the natural azulene derivatives chamazulene and guaiazulene. Furthermore, some synthetic azulene analogues have been found to possess anti-inflammatory activity. In this investigation we report the synthesis of five 3-alkyl or 3-(hydroxy)alkylazulene-1-carboxylic acids and esters, from tropolone, via the corresponding furanone. The synthesised compounds were tested for their effect on the peroxidation of rat hepatic microsomal membrane lipids, applying the 2-thiobarbituric acid test. Their anti-inflammatory activity was evaluated in vitro by the offered inhibition of soybean lipoxygenase. All the tested molecules were found to inhibit lipid peroxidation by 100% at 1 mM. They were also found to considerably inhibit lipoxygenase activity. The above results are discussed in relation to the structure and physicochemical properties of the examined azulene derivatives.  相似文献

Protein recognition and binding play a fundamental role in living systems but sometimes also result in pathological aggregates. To probe the coupling between folding and binding in a homoheptameric system, we have characterized the time-resolved unfolding/disassembly mechanism of human co-chaperonin protein 10 (cpn10) by a combination of experimental and computational methods. The results from both approaches are in excellent agreement and make obvious that the kinetic process is three-state: an initial polypeptide-unfolding step, resulting in a non-native heptamer, is followed by a slower heptamer-dissociation step. This demonstrates that the barriers on the kinetic free-energy landscape are defined by thermodynamic stability. cpn10 is one of few, and the only heptameric, experimentally characterized system that follows the "fly casting scenario" of molecular recognition.  相似文献

Sample preparation techniques based on non-porous membrane extraction generally offer a high degree of selectivity and enrichment power, together with convenient possibilities for direct and automated connections to chromatographic and other analytical instruments. Neutral oxosteroids have been derivatised with Girard T and P hydrazine reagents to give the corresponding Girard hydrazone quaternary ammonium salts. Both Girard T (GT) and Girard P (GP) hydrazones of oxosteroids give very intense [M](+) ion signals in electrospray (ES) mass spectra and fragment within the ES interface and collision cell to give characteristic fragment ions. GT and GP derivatives give informative high-energy collision-induced dissociation spectra, from which the structure of the precursor oxosteroid can be determined. Both charge-remote and charge-mediated mechanisms are responsible for the formation of the fragment ions at high collision-energy.  相似文献

Three polyamine ligands, ethylenediamine (EDA), diethylenetriamine (DTA), and triethylenetetramine (TETA), were bound to three chloromethylated “popcorn” polystyrene resins (16, 50, and 100% phenyl ring substitution) with the use of pyridine as the reaction medium. The rate of chloride displacement decreased with increasing molecular weight of the amine and higher degree of resin chloromethylation, while the extent of multiple attachments to the polymer matrix increased. The additional crosslinking, a result of multiple attachments, caused the polyamine resins to swell to a lesser extent in pyridine and water. The ability of the insoluble polyamine–polystyrene resins to chelate Cu2+ ions from dilute solutions (200 ppm) was determined at pH 5. With EDA resins the capacity for Cu2+ increased with increasing amount of the bound polyamine, with DTA it remained unchanged, while with TETA it was found to decrease.  相似文献

Mono- or 1,1′-bis-acylation of ferrocene, its mono and 1,1′-dimethyl and phenyl derivatives and of [3]ferrocenophane with o-chlorobenzoyl chloride/AlCl3 affords the corresponding (isomeric) chlorobenzoyl ferrocenes in high yields which can be separated by column, layer or high pressure liquid chromatography and in the case of the monomethylferrocene monoketones also by crystallization. The cleavage of the (o-chlorobenzoyl)ferrocenes by potassium-t-butoxide (and traces of water) yields the corresponding ferrocene carboxylic acids except for the α-phenyl derivatives in almost quantitative yields, thus offering a very convenient access to these acids. In all cases the isomer distribution and thereby the relative site reactivities were determined.  相似文献

When Eu2(SO3)3·3H2O is heated in air it forms after dehydration two sulfite-sulfate phases: Eu2(SO3)2SO2 and Eu2SO3(SO4)2. The latter phase is thermally remarkably stable and decomposes above 700°C via Eu2O2SO4 to Eu2O3. Anhydrous Eu2(SO3)3 and Eu2O2SO4 were also found as intermediates before Eu2O3 when the TG experiments were carried out in nitrogen but in other details the decomposition mechanism differs from that in air.
Zusammenfassung Eu2(SO3)3·3H2O bildet durch Erhitzen in Luft nach Dehydratation zwei Sulfit-Sulfat-Phasen: Eu2(SO3)2SO4 und Eu2SO3(SO4)2·Letztere ist thermisch bedeutend stabiler und zersetzt sich oberhalb 700°C über Eu2O2SO4 zu Eu2O3. Bei Durchführung der TG-Experimente in Stickstoff wurden als Zwischenprodukte vor Eu2O3 auch wasserfreies Eu2(SO3)3 und Eu2O2SO4 gefunden, in anderen Details weicht jedoch der Mechanismus der Zersetzung von dem der Zersetzung in Luft ab.

The azido tetrazolo valence isomerism of twenty 9-methyl-s-triazolo[4,3-c]tetrazolo[1,5-a]pyrimidines with different substituents at position 5 has been studied by means of 1H NMR spectroscopy. All the compounds have been found to be tetrazoles in the solid state and in (CD3)2SO solution; in CF3COOH azido and tetrazolo isomers are in equilibrium. From equilibrium constants K and thermodynamic data determined it is concluded that in this series K depends on both electronic effects and steric requirements of the 5-substituents. A linear relation between K and σ was found for 5-arylderivatives because ΔS° keeps approximately constant and the substituent mainly operates on ΔH°.  相似文献

The acidity functions of aqueous trifluoroacetic and trifluoromethanesulphonic acid mixtures, and aqueous hexafluoropropane-2, 2-diol solutions, have been determined by differential pulse polarography. The apparent shift of the half-wave potential for the ferrocene—ferricinium couple, as the solvent composition is changed, is used to indicate the change in potential of a glass electrode; acidity is measured as the HGF acidity function. The densities of two of these solvent systems as a function of composition are also reported. Trifluoromethanesulphonic acid—water mixtures represent the strongest aqueous acid solvent system so far studied.  相似文献
