High yield protection of alcohols, including tertiary and base sensitive alcohols, as benzhydryl ethers by heating with diphenyldiazomethane in the absence of any other reagent

A protecting group that can be introduced efficiently without the need for any acid or base catalysis and which is not prone to acid or base catalysed migration is a significant advantage for many syntheses. Benzhydryl [diphenylmethyl] ethers of sugar lactones are formed in high yield under neutral conditions when the corresponding alcohol is heated with diphenyldiazomethane in an inert solvent such as acetonitrile or toluene; this allows the easy protection of base sensitive and highly hindered tertiary alcohols in the absence of any other reagents.     

Fractionation‐dependent improvements in proteome resolution in the mouse hippocampus by IEF LC‐MS/MS

An assessment of fractionated mouse hippocampal peptides was conducted. Protein extract from a single mouse hippocampus was enzymatically digested and fractionated by IEF. Aliquots of fractions were pooled into fewer, more complex samples. The unfractionated lysate, fractions, and pooled fractions were subjected to LC‐MS/MS analysis. Samples consisting of many individual fractions had more protein identifications, greater protein sequence coverage, and quantified proteins with more spectral counts than protein extract that was unfractionated or pooled into fewer LC‐MS/MS samples. Additionally, prefractionation reduced the median CV for spectral counts as much as 33%. However, the relative gain in proteome resolution was found to saturate with increasing fractionation extent. This study demonstrates how prefractionation by offline IEF can improve the resolution of proteomic investigations of the mouse hippocampus, and that a data‐driven pooling methodology can reduce excessive and cost‐ineffective fractionation.     

Recovering incomplete data using Statistical Multiple Imputations (SMI): a case study in environmental chemistry

This paper presents a statistical technique that can be applied to environmental chemistry data where missing values and limit of detection levels prevent the application of statistics. A working example is taken from an environmental leaching study that was set up to determine if there were significant differences in levels of leached arsenic (As), chromium (Cr) and copper (Cu) between lysimeters containing preservative treated wood waste and those containing untreated wood. Fourteen lysimeters were setup and left in natural conditions for 21 weeks. The resultant leachate was analysed by ICP-OES to determine the As, Cr and Cu concentrations. However, due to the variation inherent in each lysimeter combined with the limits of detection offered by ICP-OES, the collected quantitative data was somewhat incomplete. Initial data analysis was hampered by the number of ‘missing values’ in the data. To recover the dataset, the statistical tool of Statistical Multiple Imputation (SMI) was applied, and the data was re-analysed successfully. It was demonstrated that using SMI did not affect the variance in the data, but facilitated analysis of the complete dataset.     

Syntheses of aza and fluorine-substituted 3-(piperidin-4-yl)-4,5-dihydro-1H-benzo[d][1,3]diazepin-2(3H)-ones

A practical and expedient synthesis of the title compounds is described. They were prepared by Stille reaction of nitro halopyridines 4 or nitro fluro-halobenzenes 10, followed by Michael addition of tert-butyl 4-aminopiperidine-1-carboxylate to the resulting activated vinyl compounds 5 and 11, hydrogenation (-NO₂→-NH₂), cyclic urea formation, Boc removal, and HCl salt formation. However, N3 and F1 analogs could not be made by this general strategy. Activated vinyl compounds 5a and 5d when reacted with tert-butyl 4-aminopiperidine-1-carboxylate did not stop at the desired Michael addition stage; but proceeded to produce azaindolines 8 and 9. Michael addition did not occur to compound 11d; instead, the fluorine atom was displaced.     

Improved algorithm for analysis of the least-squares treatment of titration data

In least-squares data-processing, previous investigators have reported the evaluation of partial derivatives that show the effect on the parameters, from errors in individual data points. By replacing the finite difference method used earlier, an increase in the speed of the computations by a factor of 40 or greater can be achieved.
