Biomimetic tandem protein/lipid damage : The reactions of reductive reactive species, produced by γ irradiation of water, with Zn and Cd complexes of a metallothionein from a plant afford diffusible sulfur‐centred radicals generated from methionine residues and sulfide ligands. These species migrate from the aqueous phase to the lipid bilayer and transform the cis double bond of the oleate moiety to trans isomer (see scheme).

We consider aggregation of products with similar characteristics in a two-level hierarchical production planning model. A robust aggregate plan at the upper level is such that, at the lower level, a disaggregation policy exists even when detailed demands may vary within some given bounds. We provide necessary and sufficient conditions for the robustness of an aggregate plan and obtain a closed form expression of these conditions. The synthesis and characterization of a series of picoplatin-based (picoplatin = [PtCl(2)(mpy)(NH(3))], mpy = 2-methylpyridine), Pt(iv) complexes with axial carboxylato ligands of increasing length are reported. The synthesis is based on the oxidation with hydrogen peroxide of picoplatin to give the cis,cis,trans-[PtCl(2)(mpy)(NH(3))(OH)(2)] intermediate and then its transformation into the dicarboxylato complexes cis,cis,trans-[PtCl(2)(mpy)(NH(3))(RCOO)(2)] (R = CH(3)(CH(2))(n), n = 0-4) with the corresponding anhydride. Pt(iv) complexes with n = 0-2 were selected to be tested on four malignant pleural mesothelioma (MPM) cell lines, on human mesothelial cells (HMC), and on the cisplatin-sensitive ovarian A2780 cell line along with cisplatin as a metallo-drug reference. In general, the longer the axial chain, the more cytotoxic and selective the Pt(IV) complex is. Pt(IV) analogs show good activity on the MPM cell lines, approaching or in some case bypassing that of cisplatin and represent quite promising drug candidates for the treatment of tumors whose chemoresistance is mainly based on glutathione overexpression, such as MPM.  相似文献

The mechanism of magnetic interactions in the bulk ferromagnet para-(methylthio)phenyl nitronyl nitroxide crystal (YUJNEW) has been theoretically reinvestigated, using only data from ab initio calculations and avoiding any a priori assumptions. We first calculate the microscopic magnetic interactions (JAB exchange couplings) between all unique radical pairs in the crystal, and then generate the macroscopic magnetic properties from the energy levels of the corresponding Heisenberg Hamiltonian. We thus propose a first principles, bottom-up (i.e. micro-to-macro) approach that brings theory and experiment together. We have applied this strategy to study the magnetism of YUJNEW using data from the previously reported 298 and 114 K crystal structures, and also data from a 10 K neutron diffraction structure fully reported in this work. The magnetic topology at 298 K is two-dimensional: noninteracting planes, with three different in-plane JAB pair interactions (+0.24, +0.09, and -0.11 cm(-1)) and one numerically negligible (+0.02 cm(-1)) inter-plane JAB interaction. In contrast, the magnetic topology at 114 and 10 K is three-dimensional, with two non-negligible in-plane JAB constants (+0.11 and +0.07 cm(-1) at 114 K; +0.22 and +0.07 cm(-1) at 10 K) and one inter-plane pair interaction (+0.07 cm(-1) at 114 K; +0.08 cm(-1) at 10 K). Although this three-dimensional magnetic topology is consistent with YUJNEW being a bulk ferromagnet, there is only a qualitative agreement between computed and experimental magnetic susceptibility chiT(T) data at 114 K. However, the experimental chiT(T) curve is quantitatively reproduced at 10 K. The heat capacity curve presents a peak at around 0.12 K, close to the estimated experimental peak (0.20 K).  相似文献

Following the theoretical predictions [1], the observation of two-photon processes by interaction of vacuum ultraviolet (VUV) radiation with inner-shell levels of atoms requires focused intensities in the 1013-1014 W/cm2 range. Our aim is to reach this regime in order to study non-linear optics at these wavelengths. We first optimized the high harmonic conversion efficiency in argon by studying the best experimental conditions for phase-matching, concentrating on focus geometry related to laser energy, cell length and position relative to the focus. We then studied the resulting harmonic beam focusability by a toroidal mirror (f=10 cm) and made an image of the harmonic focus. We present in this Letter a type of quadriwave lateral shearing interferometer for x-ray phase imaging. This device is based on a phase chessboard, and we take advantage of the large spectrum of the source to produce interferograms with a propagation-invariant contrast. Such a grating has been created for hard x-ray interferometry and experimentally tested on a synchrotron beamline at Soleil.  相似文献

The oxidation processes undergone by the [Pt2(mu-S)2] core in [Pt2(P[intersection]P)2(mu-S)2](P[intersection]P = Ph2P(CH2)nPPh2, n= 2,3) complexes have been analysed on the basis of electrochemical measurements. The experimental results are indicative of two consecutive monoelectronic oxidations after which the [Pt2(mu-S)2] core evolves into [Pt2(mu-S2)]2+, containing a bridging disulfide ligand. However, the instability of the monoxidised [Pt2(P[intersection]P)2(mu-S)2]+ species formed initially, which converts into [Pt3(P[intersection]P)3(mu-S)2]2+, hampered the synthesis and characterisation of the mono and dioxidised species. These drawbacks have been surpassed by means of DFT calculations which have also allowed the elucidation of the structural features of the species obtained from the oxidation of [Pt2(P[intersection]P)2(mu-S)2] compounds. The calculated redox potentials corresponding to the oxidation processes are consistent with the experimental data obtained. In addition, calculations on the thermodynamics of possible processes following the degradation of [Pt2(P[intersection]P)2(mu-S)2]+ are fully consistent with the concomitant formation of monometallic [Pt(P[intersection]P)S2)] and trimetallic [Pt3(P[intersection]P)3(mu-S)2]2+ compounds. Extension of the theoretical study on the [Pt2Te2] core and comparisons with the results obtained for [Pt2S2] have given a more general picture of the behaviour of [Pt2X2](X = chalcogenide) cores subject to oxidation processes.  相似文献

Some 5-arylmethylene-3,4-dimethyl-3-pyrrolin-2-ones react with both bromine and nitronium tetrafluoroborate (NO2BF4) to give 5-(aryl)nitromethylene-3-pyrrolin-2-ones and 5-(aryl)bromomethylene-3-pyrrolin-2-ones, respectively. The use of bromine in methanol affords 5-(aryl)bromomethyl-3,4-dimethyl-5-methoxy-3-pyrrolin-2-ones. Whereas pyrromethenones react mainly on the pyrrole ring, ethyl 3,4-demethyl-5-[(3,4-dimethyl-5-oxo-3-pyrrolin-2-yl)methylene]-1H-pyrrole-2-carboxylate reacts as the aryl derivatives, however, with bromine in methanol the addition of two methoxy groups at the exocyclic double bond takes place. 3,4-Dimethyl-5-(2-pyridylmethylene)-3-pyrrolin-2-one does not react with bromine or NO2BF4, but reacts as the aryl derivatives with bromine in methanol. The reactivity patterns are in agreement with the theoretical ones obtained from MINDO/3 calculations, using theFukui frontier orbital model. The obtained results are used to explain the reactivity of rubins (biladienes-a,c) and verdins (bilatrienes-a,b,c) in front of electrophiles.Einige 5-Arylmethylen-3,4-dimethyl-3-pyrrolin-2-one reagieren sowohl mit Brom als auch mit Nitroniumtetrafloroborat (NO2BF4). Bei Verwendung einer methanolischen Bromlösung werden 5-(aryl)bromomethyl-3,4-dimethyl-5-methoxy-3-pyrrolin-2-one gebildet. Pyrromethenone reagieren hauptsächlich am Pyrrolring, Ethyl 3,4-dimethyl-5-[(3,4-dimethyl)-5-oxo-3-pyrrolin-2-yl)methylen]-1H-pyrrol-2-carboxylat hingegen verhält sich wie ein Arylderivat, mit methanolischen Bromlösung jedoch erfolgt Eintritt zweier Methoxygruppen an der exocyclischen Doppelbindung.5-(2-Pyridyl)methylen-3,4-dimethyl-3-pyrrolin-2-on reagiert nicht mit Brom oder NO2BF4, wohl aber mit einer methanolischen Bromlösung und verhält sich unter diesen Bedingungen wie ein Arylderivat; 3- und 4-Pyridylderivate verhalten sich analog. Die Reaktivität ist in Übereinstimmung mit theoretischen Werten aus MINDO/3-Rechnungen unter Verwendung des Fukui frontier orbital model. Die Reaktivität von Rubinen (Biladiene-a, c) und Verdinen (Bilatriene-a,b,c) gegenüber Elektrophilen werden im Zusammenhang mit den erhaltenen Resultaten diskutiert.
