Abstract —The effects of ultraviolet irradiation of double-stranded synthetic polynucleotides containing BrU and A have been investigated. Homopolymer pairs and alternating copolymers composed of either ribo- or deoxyribo-nucleotides were prepared and were irradiated with either 313 nm or ˜ 285 nm light. Strand separation and a modest amount of strand breakage followed irradiation of the homopolymer pairs. Changes in the ultraviolet absorption spectra of the polymers during irradiation reflected the sum of hyperchromic increases caused by progressive strand separation and loss of absorbance caused by photoproduct formation. Extensive debromination occurred. An RNase digest of irradiated poly(rA)–poly14C(rBrU), analysed by column chromatography, showed components similar to those found previously upon irradiation of single-stranded poly(rBrU). Little photoproduct was released by RNase digestion as mononucleotides. The major photoproduct was in the dinucleotide fraction, and may be 5,5'-diuracil. Base sequence had a profound effect on the sensitivity of the polynucleotides. Irradiation of alternating copolymers with doses of light comparable to those that produced major photochemical changes in the homopolymer pairs brought about little if any change in the copolymers of alternating base sequence.  相似文献

The magnetic susceptibility (κRT) and saturation magnetization (MS) of microbially synthesized magnetites were systematically examined. Transition metal (Cr, Mn, Co, Ni and Zn)- and lanthanide (Nd, Gd, Tb, Ho and Er)-substituted magnetites were microbially synthesized by the incubation of transition metal (TM)- and lanthanide (L)-mixed magnetite precursors with either thermophilic (TOR-39) or psychrotolerant (PV-4) metal-reducing bacteria (MRB). Zinc incorporated congruently into both the precursor and substituted magnetite, while Ni and Er predominantly did not. Microbially synthesized Mn- and Zn-substituted magnetites had higher κRT than pure biomagnetite depending on bacterial species and they exhibited a maximum κRT at 0.2 cationic mole fraction (CMF). Other TMs’ substitution linearly decreased the κRT with increasing substitution amount. Based on the MS values of TM- and L-substituted magnetite at 0.1 and 0.02 CMF, respectively, Zn (90.7 emu/g for TOR-39 and 93.2 emu/g for PV-4)- and Mn (88.3 emu/g by PV-4)-substituted magnetite exhibited higher MS than standard chemical magnetite (84.7 emu/g) or pure biomagnetite without metal substitution (76.6 emu/g for TOR-39 and 80.3 emu/g for PV-4). Lanthanides tended to decrease MS, with Gd- and Ho-substituted magnetites having the highest magnetization. The higher magnetization of microbially synthesized TM-substituted magnetites by the psychrotroph, PV-4 may be explained by the magnetite formation taking place at low temperatures slowing mechanics, which may alter the magnetic properties compared to the thermophile, through suppression of the random distribution of substituted cations.  相似文献

This study explores the hypothesis that clear speech is produced with greater "articulatory effort" than normal speech. Kinematic and acoustic data were gathered from seven subjects as they pronounced multiple repetitions of utterances in different speaking conditions, including normal, fast, clear, and slow. Data were analyzed within a framework based on a dynamical model of single-axis frictionless movements, in which peak movement speed is used as a relative measure of articulatory effort (Nelson, 1983). There were differences in peak movement speed, distance and duration among the conditions and among the speakers. Three speakers produced the "clear" condition utterances with movements that had larger distances and durations than those for "normal" utterances. Analyses of the data within a peak speed, distance, duration "performance space" indicated increased effort (reflected in greater peak speed) in the clear condition for the three speakers, in support of the hypothesis. The remaining four speakers used other combinations of parameters to produce the clear condition. The validity of the simple dynamical model for analyzing these complex movements was considered by examining several additional parameters. Some movement characteristics differed from those required for the model-based analysis, presumably because the articulators are complicated structurally and interact with one another mechanically. More refined tests of control strategies for different speaking styles will depend on future analyses of more complicated movements with more realistic models.  相似文献

Electron-electron coincidence spectra of Ar-Kr clusters after photoionization have been measured. An electron with the kinetic energy range from 0 to approximately 1 eV is found in coincidence with the Ar 3s cluster photoelectron. The low kinetic energy electron can be attributed to an Ar + Kr+ + Kr+ final state which forms after electron transfer mediated decay. This autoionization mechanism results from a concerted transition involving three different atoms in a van der Waals cluster; it was predicted theoretically, but hitherto not observed.  相似文献

BACKGROUND AND PURPOSE: Systemic lupus erythematosus (SLE) is an autoimmune disease in which almost all the organs are involved. Neuropsychiatric SLE is of one of the major concerns in the clinical evaluation of this disease. Routine magnetic resonance imaging (MRI) findings are often nonspecific or negative. In this study, we explored the use of diffusion tensor imaging in assisting with the diagnosis of SLE. METHODS: Data from 34 SLE patients (age range, 18-73 years) and 29 age-matched volunteers (age range, 29-64 years) were analyzed. MRI was performed on a 1.5-T clinical MR scanner with a quadrature head coil. The average diffusion constant (D(av)) and diffusion anisotropy maps [fractional anisotropy (FA)] were determined on a pixel-by-pixel basis. Regional diffusion measurements were made by region of interest in the genu and splenium of the corpus callosum (CC), anterior and posterior limb of the internal capsule (IC) and frontal lobe and thalamus. The diffusion distribution was fitted to a triple-Gaussian model. The mean of the brain tissue distribution was determined as a mean diffusion constant for the whole brain (BD(av)). Student's t test was used to determine the diffusion difference between SLE patients and control subjects. The SLE patients were separated into two groups according to their MRI results. A P value lower than .05 was considered to be statistically significant. RESULTS: Twenty of the 34 SLE patients with abnormal MRI results showed findings dominated by nonspecific white matter disease. The BD(av) and D(av) values of the frontal lobe, splenium CC and anterior IC were significantly higher in all SLE patients as compared with the control subjects. The SLE patients with normal MRI results also showed higher BD(av) and D(av) values in the frontal lobe, splenium and anterior and posterior limbs of the IC as compared with the control subjects. There was no significant difference in the D(av) values of the thalamus between the SLE patients and the control subjects. The BD(av) value in the SLE patient group was robustly correlated with the D(av) values of the frontal lobe, splenium and thalamus. These correlations were found to be similarly significant for the SLE patients with normal MRI findings. The diffusion anisotropy measurements showed that splenium CC had the highest FA value in both the control subjects and SLE patients. Overall, SLE patients had lower FA values in the genu and splenium CC as compared with the control subjects. In the group of patients with normal MRI findings, the FA values of the genu and splenium CC as well as the anterior IC were also lower than those in the control subjects. Pearson's correlation statistics revealed robust correlations between the measurements of D(av) and FA values in the SLE patient group. CONCLUSION: Quantitative diffusion imaging and diffusion anisotropy showed early changes in the brains of the SLE patients. Increased BD(av) and D(av) values of the frontal lobe as well as decreased anisotropy in the genu CC and anterior IC may represent preclinical signs of central nervous system involvement of SLE even when the routine MRI findings are negative or nonspecific. Quantitative diffusion analysis may prove to be useful in detecting the initial brain involvement of SLE and may enable monitoring of early disease progression and treatment efficacy.  相似文献

A theoretical study on the structure and adsorption mechanism of hydrazine (N2H4) on Ni(1 0 0) are presented. The hydrazine molecule was found to adsorb on the surface through one of its nitrogen atom in its anti-conformation. The charge transfer from hydrazine lone pair orbitals played a key role in the formation of the bonding. The mechanism involved in the bonding was found to reduce the necessity of hyper-conjugation interaction, that reduces the gauche effect found in hydrazine at the gas-phase. Upon adsorption to the surface, the reduced interaction resulted in the promotion of a more favored conformation through its anti-conformation.  相似文献

The adhesion strength of polybutylene terephthalate (PBT) on aluminum was investigated using density functional theory-based total energy calculations. Aluminum atom was connected to a PBT monomer at different orientations and total energies were calculated in order to determine the most stable orientation. The energy differences showed that the Al oriented at 180° with the ester group of the monomer bonded strongly. Using this orientation, the PBT monomer-adhesion on aluminum surface and the aluminum atom adhesion on PBT bulk were also investigated.  相似文献

Photolysis of substituted thiophene‐S‐oxides in solution results in the formation of either the corresponding thiophene or furan, in addition to uncharacterized materials. No good rationalization is available for the choice of which pathway may predominate, but it is demonstrated that the photolysis of 2,5‐bistrimethylsilylthiopene‐ S‐oxide produces O(3P) in the same manner as the well‐established photolysis of dibenzothiophene‐S‐oxide. OBJECTIVE: The objective of this study was to use magnetic resonance imaging (MRI) to detect the time when and the location at which orally delivered mucoadhesive drugs are released. MATERIALS AND METHODS: Drug delivery systems comprising tablets or capsules containing a mucoadhesive polymer were designed to deliver the polymer to the intestine in dry powder form. Dry Gd-DTPA [diethylenetriaminepentaacetic acid gadolinium(III) dihydrogen salt hydrate] powder was added to the mucoadhesive polymer, resulting in a susceptibility artifact that allows tracking of the application forms before their disintegration and that gives a strong positive signal on disintegration. Experiments were performed with rats using T(1)-weighted spin-echo imaging on a standard 1.5-T MRI system. RESULTS: The susceptibility artifact produced by the dry Gd-DTPA powder in tablets or capsules was clearly visible within the stomach of the rats and could be followed during movement towards the intestine. Upon disintegration, a strong positive signal was unambiguously observed. The time between ingestion and observation of a positive signal was significantly different for different application forms. Quantification of the remaining mucoadhesive polymer in the intestine 3 h after observed release showed significant differences in mucoadhesive effectiveness. CONCLUSION: MRI allows detection of the exact time of release of the mucoadhesive polymer in vivo, which is a prerequisite for a reliable quantitative comparison between different application forms.  相似文献

Alzheimer's disease (AD) is a progressive neurodegenerative disorder that is the most common cause of dementia in aging populations. Although senile plaques and neurofibrillary tangles are well-established hallmarks of AD, changes in cerebral white matter correlate with cognitive decline and may increase the risk of the development of dementia. We used the triple transgenic (3xTg)-AD mouse model of AD, previously used to show that white matter changes precede plaque formation, to test the hypothesis that MRI detectable changes occur in the corpus callosum, external capsule and the fornix. T2-weighted and diffusion tensor magnetic resonance imaging and histological stains were employed to assess white matter in older (11–17 months) 3xTg-AD mice and controls. We found no statistically significant changes in white matter between 3xTg-AD mice and controls, despite well-developed neurofibrillary tangles and beta amyloid immunoreactive plaques. Myelin staining was normal in affected mice. These data suggest that the 3xTg-AD mouse model does not develop MRI detectable white matter changes at the ages we examined.  相似文献
