A new multiple-layer matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) sample-spotting technique is described. This fast and easy technique was evaluated with poly(ethylene glycol) (PEG) standards and optimized conditions for these synthetic polymers were obtained. PEGs up to 35 kDa were detectable with this approach and single monomer resolution was observed up to 20 kDa. The spotting was performed using a multiple-layer approach, which offers the capability of complex sample preparation without the requirement of premixing the different matrix, analyte and doping salt solutions. The technique reduces the time required for sample preparation and offers high flexibility with respect to sample composition and solvents utilized for the crystallization of the compounds. The technique is thus perfectly suited for applications in combinatorial material research.  相似文献

1-(5-Nitro-2-thiazolyl)-2-imidazolidinone is synthesized from 2-amino-5-nitro-thiazole and 2-chloroethyl-isocyanate. The structure of this new potent schistosomicide is established by acid and alkaline degradation as well as by synthesis starting from 2-imidazolidinone. Motivated by pump-probe experiments of I(2) in a room-temperature sample, the detection of fractional revivals is investigated using full-dimensional quantum wave packet calculations. It is shown that the structures observed in the pump-probe signal depend sensitively on the probe parameters employed and that the observed signal reflects a particular phase effect between fractional revivals.  相似文献

Total Synthesis of (+)-D-Homoestrone 3-methyl ether A novel total synthesis of (+)-D-homoestrone 3-methyl ether ( 21 ) is described starting from (S)-8a-methyl-3,4,8,8a-tetrahydro-2H, 7H-naphthalene-1,6-dione ( 1 ) as a chiral synthon for the rings C and D. The key step involves alkylation of the derived 3 with m-methoxyphenacyl bromide ( 4 ) as an AB-building block to give the dioxo-secosteroid 5 . Hydrogenation of 5 affords the trans-decalone 11 . As by-products the epimeric cis-decalones 12 and 13 were characterized. Cyclization of 11 leads under kinetic control predominantly to the Δ9(11)-tetraene 14 . Catalytic hydrogenation of 14 and subsequent modification in ring D give the title compound 21 . It was found that 14 and also the derived Δ8-isomer 15a add hydrogen from the α-face of the molecule to an extent of about 80%. The 8α-D-homoestrone derivatives 20a and 23 as well as the 9β-isomers 19a and 22 were characterized.  相似文献

Enantioselective Reactions on Porphine Type Nickel Complexes The thermodynamically controlled addition of alcohols to (+)-(1R)-[1-methyl-8H-HDP]nickelperchlorate ( 1 ; e.e. 92%) yields exclusively the corresponding cis-1,11-disubstituted porphinoids. Chemical transformation of functional groups in the alkoxy side-chain of the chiral addition product followed by acid catalyzed elimination yields the derived alcohols and 1 . By this procedure, the following enantioselective transformations were studied: methylation of meso-2,3-butandiol ( 5 ) to (+)-(2R,3S)-3-methoxy-2-butanol ( 8a ; e.e. 87%), diimide reduction of 2-ethylallyl alcohol ( 9 ) to (+)-(2R)-2-methyl-1-butanol ( 12a ; e.e. 15%), and hydride reduction of 4-hydroxy-2-butanone ( 13 ) to (+)-(3S)-1,3-butandiol ( 16a ; e.e. 20%). Addition of 2,2-dimethyl-1,3-propandiol ( 17 ) to 4 , followed by esterification of the free hydroxy group with trifluoromethanesulfonic anhydride and solvolysis of the sulfonate 19 yielded a bridged complex with unrearranged alkyl chain for which structure 20 is proposed.  相似文献

Translational tracer diffusion of spherical macromolecules in crowded suspensions of rodlike colloids is investigated. Experiments are done using several kinds of spherical tracers in fd-virus suspensions. A wide range of size ratios L/2a of the length L of the rods and the diameter 2a of the tracer sphere is covered by combining several experimental methods: fluorescence correlation spectroscopy for small tracer spheres, dynamic light scattering for intermediate sized spheres, and video microscopy for large spheres. Fluorescence correlation spectroscopy is shown to measure long-time diffusion only for relatively small tracer spheres. Scaling of diffusion coefficients with a/xi, predicted for static networks, is not found for our dynamical network of rods (with xi the mesh size of the network). Self-diffusion of tracer spheres in the dynamical network of freely suspended rods is thus fundamentally different as compared to cross-linked networks. A theory is developed for the rod-concentration dependence of the translational diffusion coefficient at low rod concentrations for freely suspended rods. The proposed theory is based on a variational solution of the appropriate Smoluchowski equation without hydrodynamic interactions. The theory can, in principle, be further developed to describe diffusion through dynamical networks at higher rod concentrations with the inclusion of hydrodynamic interactions. Quantitative agreement with the experiments is found for large tracer spheres, and qualitative agreement for smaller spheres. This is probably due to the increasing importance of hydrodynamic interactions as compared to direct interactions as the size of the tracer sphere decreases.  相似文献
