Ciprofloxacin is a widely used fluoroquinolone drug with broad spectrum antibacterial activities. Clinical experience has shown incidences of adverse effects related to skin, hepatic, central nervous system, gastrointestinal and phototoxicity. India is a tropical country and sunlight is abundant throughout the day. In this scenario exposure to ambient levels of ultraviolet radiation (UV-R) in sunlight may lead to harmful effects in ciprofloxacin users. Phototoxicity assessment of ciprofloxacin was studied by two mouse fibroblast cell lines L-929 and NIH-3T3. Generation of reactive oxygen species (ROS) like singlet oxygen (1O2), superoxide anion radical (O2*-) and hydroxyl radical (*OH) was studied under the exposure of ambient intensities of UV-A (1.14, 1.6 and 2.2 mW cm(-2)), UV-B (0.6, 0.9 and 1.2 mW cm(-2)) and sunlight (60 min). The drug was generating 1O2, O2*- and *OH in a concentration and dose-dependent manner. Sodium azide (NaN3) and 1,4-diazabicyclo 2-2-2-octane (DABCO) inhibited the generation of 1O2. Superoxide dismutase (SOD) inhibited 90-95% O2*- generation. The drug (5-40 microg mL(-1)) was responsible for linoleic acid peroxidation. Quenching study of linoleic acid peroxidation with SOD (25 and 50 U mL(-1)) confirms the involvement of ROS in drug-induced lipid peroxidation. The generation of *OH radical was further confirmed by using specific quenchers of *OH such as mannitol (0.5 M) and sodium benzoate (0.5 M). 2'-deoxyguanosine (2'-dGuO) assay and linoleic acid peroxidation showed that ROS were mainly responsible for ciprofloxacin-sensitized photo-degradation of guanine base. L-929 cell line showed 29%, 34% and 54% reduced cell viability at higher drug concentration (300 microg mL(-1)) under UV-A, UV-B and sunlight, respectively. 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay in NIH-3T3 cell line at higher drug concentration (300 microg mL(-1)) showed a decrease in cell viability by 54%, 56% and 59% under UV-A, UV-B and sunlight, respectively. Results of neutral red uptake assay (NRU) in L-929 cell line were in accordance with MTT assay. The NIH-3T3 cell line showed a higher photosensitizing potential than L-929. The phototoxicity end point shows a time- and concentration-dependent statistically significant (P<0.001) damage. Ciprofloxacin produced ROS by Type I and Type II photodynamic reactions, interacted with nucleic acid moiety and inhibited cell viability. Further, UV-induced photo-peroxidation of linoleic acid accorded the involvement of ROS in the manifestation of drug phototoxicity. Appearance of ciprofloxacin-induced phototoxicity at the ambient level of sunlight is a real risk for the people of India and for those of other tropical countries. We suggest that sunlight exposure should be avoided (especially peak hours) during ciprofloxacin treatment.  相似文献

Halogen Dance (HD) reactions are a useful tool for synthetic chemists as they enable access to positions in aromatic and heteroaromatic systems for subsequent functionalization which are often difficult to address by other methods, hence, allowing entry to versatile scaffolds. While the method can be extremely useful, this transformation is often neglected upon designing synthetic sequences. This may be largely attributed to the lack of comprehensive reference works covering the general principles and outlining the versatility and limitations of the technique. The following review tries to present HD reactions in a clear and concise manner in order to convince more chemists of its advantages. It covers the field of HD reactions from their first observation in 1951 until the present. The important contributions leading to the elucidation of the mechanism are briefly outlined followed by a detailed mechanistic section and a discussion of factors which influence HD reactions. Finally, an overview of HD reactions on various carbocyclic and heterocyclic ring systems and its applications in the synthesis of complex compounds is given.  相似文献

From the endophytic fungus Phomosis sp., four known phomosines A-D (1-4) and three new phomosines H-J (5-7) have been isolated. The structures of the new compounds were determined on the basis of their spectroscopic data analysis (1H, 13C, 1H-1H COSY, HMQC, and HMBC NMR, as well as mass spectrometry). The structures of phomosine H (5) and J (7) were also confirmed by semisynthesis from phomosine A (1). The remaining four known compounds [phomosines A-D (1-4)] were identified by comparing their spectroscopic data with those reported in the literature. The four known metabolites were biologically active. Of the novel metabolites, only 6 was antifungal and antibacterial.  相似文献

Dynamic and equilibrium interfacial tensions between crude oil fractions and aqueous solutions of various compositions and pH were measured. The basic oil components seemed to determine the interfacial tensions at pH 2, while the non-dissociated and dissociated acidic components governed the interfacial tension at the natural pH and pH 9, respectively. The ionic composition of the aqueous phase influenced the degree of dissociation of the acidic components at pH 9: Na+ ions in the aqueous phase promoted dissociation of the interfacial acidic components (compared to pure water), while Ca2+ ions resulted in complexation with the dissociated acids and most likely formation of stable interfacial films. The amount of Ca2+ determined which of these phenomena that dominated when both ions were present in sea water solutions. Generally, the interfacial tensions of the oil fractions were lower when measured against the high salinity aqueous solutions than against the corresponding low salinity solutions.   相似文献

In our previous articles we gave step by step refinement process towards the development of safety properties of moving block interlocking system (MBRIS). The refinement process started from abstraction to fuzzy based safety properties using Z and then fuzzy multi agent specification language. However, one dimensional control of train passing through a switch and level crossing were not discussed. This paper reduces the existing two dimensional controls along the switch and level crossing to one dimensional for shifting it to a train only. For example, in the existing model the train movement along components switches and level crossings depends on both the train and components control. Whereas, in one dimensional control train is the only authority to control a switch and level crossing required for its desired operation. For this reduction, concurrent and mobile agent concepts are required. Therefore, we integrate mobile agent concepts with Petri nets to develop the mobile Petri net (MPN) a new class of PNs. This supports both mobility and concurrency. Further, we prove that the collection of different MPNs in a connected network is a PN. This proof allowed us to use the properties of PN to verify the system. Finally, we use MPN to model the safety properties of MBRIS along the switch and level crossing. This provides one dimensional control to a train along a switch and level crossing which increases the safety of the railway interlocking system. Moreover, we use reachability graph (RG) to verify the switch and level crossing models.  相似文献

Photo-detachment of a noncollinear triatomic anion is investigated by considering each atom of the molecular anion as a coherent source of detached-electron waves, originating in all possible directions. The waves traveling along three different trajectories result in a quantum interference that displays on a screen placed at a very large distance from the system. To explain this quantum interference, an analytical formula of detached-electron flux is derived using a collinear three-center model recently published. The detached-electron flux versus laser photon energy expression displays molecular geometry-dependent oscillatory structures on an observation plane. This oscillatory behavior in the result can be explained using the semiclassical closed-orbit theory. The outgoing electron waves produced from one coherent center are propagated in the vicinity of the sources at other two coherent centers which cause these oscillations. It is also observed that in a particular case, the noncollinear triatomic system reduces to the collinear three-center system.  相似文献

Novel magnesium oxide (MgO) nanobelt structures were successfully synthesized on a large scale, low cost and catalyst free by a microwave hydrothermal route using the magnesium metal and potassium hydroxide precursors at 200 °C for 10 min. The synthesized MgO nanobelts were systemically characterized by power X-ray diffraction (XRD), scanning electron microscopy, energy dispersion spectroscopy and transmission-high resolution electron microscopy. The XRD results indicate that the MgO nanobelts have the well-crystalline cubic phase. The detailed morphological studies revealed that the synthesized products were nanobelts and were grown in large quantity. The optical energy gap of MgO nanobelts was found to be 3.1 eV. The photoluminescence spectra of the MgO nanobelts show a strong and broad green emission band, a weak ultraviolet emission band, and a weak red emission band. This novel method will open new dimensions for synthesized 1-D metal oxide and can be easily extended to the synthesis and assembly of other inorganic nanocrystals.  相似文献

The toxicity of ZnO nanoparticles (NPs) has been widely investigated because of their extensive use in consumer products. The mechanism of the toxicity of ZnO NPs to algae is unclear, however, and it is difficult to differentiate between particle-induced toxicity and the effect of dissolved Zn2+. In the work discussed in this paper we investigated particle-induced toxicity and the effects of dissolved Zn2+ by using the chiral perturbation approach with dichlorprop (DCPP) as chiral perturbation factor. The results indicated that intracellular zinc is important in the toxicity of ZnO NPs, and that ZnO NPs cause oxidative damage. According to dose–response curves for DCPP and the combination of ZnO NPs with (R)-DCPP or (S)-DCPP, the toxicity of DCPP was too low to perturb the toxicity of ZnO NPs, so DCPP was suitable for use as chiral perturbation factor. The different glutathione (GSH) content of algal cells exposed to (R)-DCPP or (S)-DCPP correlated well with different production of reactive oxygen species (ROS) after exposure to the two enantiomers. Treatment of algae with ZnO NPs and (R)-DCPP resulted in reduced levels of GSH and the glutathione/oxidized glutathione (GSH/GSSG) ratio in the cells compared with the control. Treatment of algae with ZnO NPs and (S)-DCPP, however, resulted in no significant changes in GSH and GSH/GSSG. Moreover, trends of variation of GSH and GSH/GSSG were different when algae were treated with ZnSO4·7H2O and the two enantiomers. Overall, the chiral perturbation approach revealed that NPs aggravated generation of ROS and that released Zn2+ and NPs both contribute to the toxicity of ZnO NPs.

The nanoparticles of monovacant lacunary keggin-type polyoxometalate (C19H42N)4H3(PW11O39) has been successfully synthesized by Micelle Directed Method using hexadecyltrimthyl ammonium bromide (CTAB) as a template. Non-ionic copolymer Pluronic F127 was used as a capping agent that helps in formation of nanoparticles by preventing the aggregation during the coating of H3(PW11O39)4- onto the counter ion (C19H42N)+. The synthesized material was characterized by Scanning electron microscope (SEM), TGA, DSC and FT-IR techniques. SEM images indicated that (C19H42N)4H3(PW11O39) nanoparticles possess spherical structure. FTIR results revealed that the structural integrity of monovacant lacunary keggin-type polyoxometalates (POM) was not affected when reducing the size. TGA–DSC results showed that the material was thermally stable (with 37 % weight loss) and POM encapsulates CTAB. The efficiency of synthesized material was checked by applying it as a catalyst for the oxidation of toxic mordant yellow dye i.e., potentially hazardous and non-biodegradable azo dye. The (C19H42N)4H3(PW11O39) nanoparticles gave 96 % dye degradation in 60 min at 60 °C temperature. Moreover, on recycling POM nanoparticles demonstrate 92 % oxidative degradation of mordant yellow dye.  相似文献

The present investigation was carried out to appraise the levels of total phenols and vitamin C as well as antioxidant potential at three different ripening stages (un-ripe, semi-ripe and fully-ripe) of guava (Psidium guajava L.) fruit collected from three different geographical regions of Pakistan (Islamabad, Faisalabad and Bhakkar). The antioxidant potential of guava fruit extracts was assessed by means of different in-vitro antioxidant assays, namely inhibition of peroxidation in linoleic acid system, reducing power and radical scavenging capability. Overall, fruit at the un-ripe stage (G1) exhibited the highest levels of TPC, TFC, reducing power and DPPH radical scavenging activity, followed by the semi-ripe (G2) and fully-ripe (G3) stages. On the other hand, vitamin C content increased as the fruit maturity progressed, with highest value seen at the fully-ripe stage (G3) followed by the semi-ripe (G2) and un-ripe stage (G1). The concentration of vitamin C in fruits varied as: Faisalabad (136.4-247.9 mg 100 g?1), Islamabad (89.7-149.7 mg 100 g?1) and Bhakkar (73.1-129.5 mg 100 g?1). The results showed that different stages of maturation and geographical locations had profound effects on the antioxidant activity and vitamin C contents of guava fruit.  相似文献
