The purification and characterization of intracellular tyrosine phenol lyase from Citrobacter freundii has been carried out. The enzyme was purified 35-fold to homogeneity by ammonium sulphate precipitation and hydrophobic interaction chromatography. Its subunit molecular weight was found to be 52 kDa on sodium dodecyl sulphate polyacrylamide gel electrophoresis. The purified tyrosine phenol lyase showed maximum activity in borate buffer (0.05 M at pH 8.5) at 45 °C after 20 min of incubation. The K m and V max values of purified enzyme were found to be 0.446 mm and 0.342 mM/min/mg. This enzyme exhibits t 1/2 of 10, 52 and 130 min at 55, 45 and 35 °C, respectively. The N-terminal amino acid sequence was determined as MET-ASN-TYR-PRO-ALA-GLU-PRO-PHE-ARG-ILE-TRP-TRP-VAL-GLY. 相似文献

Tracking signals are used to determine whether a sequence of forecasts is unbiased. We analyse a version of the CUSUM tracking signal that is based on the unsmoothed average mean absolute deviation of forecasting errors. We make the common assumption that demand values in different periods are independently, identically, and normally distributed. We show that, when forecast errors in different periods are also normal iid, this version of the tracking signal can indicate that the sequence is not unbiased even when it is unbiased. We suggest a simple modification to the tracking signal that serves to eliminate this flaw. 相似文献

In this paper, the electrical conductivity and relaxation studies in two different temperature regions (T?<?T m and T?>?T m) on plasticized PEO–PMMA blend polymer electrolyte system with AgNO3 salt are reported. The polymer electrolyte system has been prepared by solution cast technique and characterized by X-ray diffraction and differential scanning calorimetry. The conduction and relaxation processes at various temperatures have been investigated in the framework of modulus formalism. The distribution of relaxation times is discussed using Argand plot. Snake venom hyaluronidases known as “spreading factor” are not extensively studied. Recently, it is argued that beyond its role as a spreading factor, venom hyaluronidase (HYL) deserves to be explored as a possible therapeutic target for inhibiting the systemic distribution of venom/toxins and also for minimizing local tissue destruction. In this context, in the present study, a low-molecular weight HYL has been isolated from Bungarus caeruleus (Indian krait) venom by single step chromatography on HPLC system. The apparent molecular weight determined by SDS-PAGE is 14 ± 2 kDa, as confirmed by zymogen study and LC–MS as well. The enzyme had optimal pH 6 and temperature 37°C. The Michaelis–Menten constant (Km) was found to be 8.48 µg/mL at 37°C. The activity of purified enzyme was completely inhibited by Ba2+ metal ion and N-acetyl imidazole group-specific agents. This work yielded a highly active HYL from B. caeruleus the first one to be isolated. Further studies on its pharmacological actions will be interesting to develop lead molecules for better management of snakebite. 相似文献

Several high‐energy‐density strained polycyclic compounds nitromethyl‐l,3‐bishomocubane (NMBHC), nitromethylene‐1,3‐bishomocubane (NMyBHC), and bis(nitromethyl)‐1,3‐bishomocubane (DNTMBHC), which were synthesized for the first time from bishomocubanone, hold potential for application as standalone fuels in liquid bipropellant systems or as additives in liquid and solid propellant formulations. DFT analysis at the B3LYP/6‐31G(d) level of theory was employed to optimize the geometries of the compounds and to determine their densities, heats of formation, and various thermodynamic properties. The density specific impulse, determined by using equilibrium thermodynamics, demonstrated an improvement of 75 s for NMBHC and NMyBHC over standard hydrocarbons. The specific impulse with ammonium perchlorate showed an improvement of 25–30 s over hydroxy‐terminated polybutadiene. Thermogravimetric analysis revealed that NMBHC, NMyBHC, and DNTMBHC evaporated readily with activation energies of 58.8, 69.2, and 74.5 kJ mol?1, respectively. 相似文献

The effectiveness of 1H?pyrazole?3,5?dicarboxylic acid 5?benzyl ester 3?phenyl ester (PCBPE) as a preventer for deterioration of IS 513 Gr. D steel in 1 M HCl medium is evaluated via weight loss, electrochemical impedance, and polarization techniques. Kinetic and thermodynamic parameters assessed the feasibility of the adsorption process at diverse temperatures. The inhibition action on mild steel has been enhanced with increasing PCBPE concentration. It is found from the polarization studies that PCBPE behaves as mixed type inhibitor in HCl medium. The adsorption process of PCBPE on mild steel surface from acid environment is favoured Langmuir adsorption isotherm. The shielding efficiency of PCBPE has been enhanced at elevated concentrations, and it has been diminished at amplified temperatures. The Atomic Force Microscope (AFM), Scanning Electron Microscope (SEM), and Energy Dispersive Spectrum (EDS) were used to establish a surface characterization of metal specimens. A quantum chemical analysis of electron density distributions in the highest occupied molecular orbital (HOMO) and lowest unoccupied molecular orbital (LUMO) demonstrated how the inhibitor undergoes adsorption on mild steel in 1 M HCl. All experimental findings substantiate the corrosion mitigation performance of PCBPE on mild steel in acidic environments. 相似文献
