An electrochemical dsDNA nanobiosensor was fabricated using amino‐functionalized multi walled carbon nanotubes modified glassy carbon electrode (NH2fMWCNTs/GCE) for the sensitive detection of DNA bases and electrochemical monitoring of drug‐DNA interaction. The influence of functional groups on MWCNT was studied by MWCNT functionalized with NH2 (NH2fMWCNTs) and COOH (COOHfMWCNT) groups based on the signal of DNA bases. The modified electrodes were characterized by scanning electron microscopy. One layer of calf thymus double stranded deoxyribonucleic acid (ct‐dsDNA) was immobilized onto the NH2fMWCNTs/GCE (dsDNA/NH2fMWCNTs/GCE). The dsDNA/NH2fMWCNTs/GCE were used to investigate the interaction between the dsDNA and the anticancer drug gemcitabine by differential pulse voltammetry in acetate buffer of pH 4.70. For the confirmation of interaction, the lowering in intensity of the current signals of guanine and adenine was considered as an indicator. Electrochemical impedance spectroscopy studies were performed for the comparison of the modified surfaces. In order to define and visualize the interaction mechanism between gemcitabine and dsDNA/NH2fMWCNTs/GCE at the molecular level, in silico methods including docking and molecular dynamics simulations were employed.  相似文献

Determination of 241Am/243Am ratios is required for vanous purposes including assay of Am by isotope dilution techniques. Alpha-spectrometry on electrodeposited sources is a preferred technique for this determination. However, there is an inherent problem of tail contribution which necessitates the use of suitable algorithms to account for the same. Recently, in the frame of a Coordinated Research Program (CRP) of the International Atomic Energy Agency (IAEA), WinALPHA software has been developed which is a combination of an asymmetrical Gaussian for the main part of the peak and a low energy function. Therefore, it was of interest to compare the use of this algorithm with the routinely used method, in our laboratory, based on geometric progression (G. P.) decrease. Since, there are no reference materials available commercially for 241Am/243Am ratios, synthetic mixtures covening a wide range (0.3 to 2.0) of 241Am/243Am α-activity ratios were used and un-ignited electrodeposited sources were prepared for α-spectrometry. The α-spectra obtained using PIPS detector, were evaluated using the two algonthms The 241Am/243Am α-activity ratios obtained were also compared with those determined by thermal ionization mass spectrometry (TIMS). An agreement of about 1% was obtained in the 241Am/243Am ratios determined by the two methods and also by using the two algorithms for α-spectrum evaluation.  相似文献

Neutron reflectivity (NR) is used to probe the solid, liquid, vapor interface of a porous superhydrophobic (SH) surface submerged in water. A low-temperature, low-pressure technique was used to prepare a rough, highly porous organosilica aerogel-like film. UV/ozone treatments were used to control the surface coverage of hydrophobic organic ligands on the silica framework, allowing the contact angle with water to be continuously varied over the range of 160 degrees (superhydrophobic) to <10 degrees (hydrophilic). NR shows that the superhydrophobic nature of the surface prevents infiltration of water into the porous film. Atomic force microscopy and density functional theory simulations are used in combination to interpret the NR results and help establish the location, width, and nature of the SH film-water interface.  相似文献

Ultraviolet (UV) radiation from sunlight is a major etiologic factor for skin cancer, the most prevalent cancer in the United States, as well as premature skin aging. In particular, UVB radiation causes formation of specific DNA damage photoproducts between pyrimidine bases. These DNA damage photoproducts are repaired by a process called nucleotide excision repair, also known as UV‐induced DNA repair. When left unrepaired, UVB‐induced DNA damage leads to accumulation of mutations, predisposing people to carcinogenesis as well as to premature aging. Genetic loss of nucleotide excision repair leads to severe disorders, namely, xeroderma pigmentosum (XP), trichothiodystrophy (TTD) and Cockayne syndrome (CS), which are associated with predisposition to skin carcinogenesis at a young age as well as developmental and neurological conditions. Regulation of nucleotide excision repair is an attractive avenue to preventing or reversing these detrimental consequences of impaired nucleotide excision repair. Here, we review recent studies on molecular mechanisms regulating nucleotide excision repair by extracellular cues and intracellular signaling pathways, with a special focus on the molecular regulation of individual repair factors.  相似文献

Phytochemical investigation of the plant Clerodendrum phlomidis Linn. F. (Lamiaceae) has now led to the isolation of two new flavonoid glycosides (1, 2) together with six known compounds identified as pectolinaringenin (3), pectolinaringenin-7-O-β-d-glucopyranoside (4), 24β-ethylcholesta-5,22E,25-triene-3β-ol (5), 24β-ethylcholesta-5,22E,25-triene-3β-O-β-D-glucopyranoside (6), (2S,3S,4R,10E)-2-[(2′R)-2′-hydroxytetracosanoylamino]-10-octadecene-1,3,4-triol (7) and andrographolide (8) mainly by spectroscopic analysis. Compounds 4 and 68 are reported for the first time from C. phlomidis.  相似文献

Here, the large amplitude free flexural vibrations of isotropic/laminated orthotropic rings are investigated, using a shear flexible curved beam element based on field consistency principle. A laminated refined beam theory is introduced for developing the element, which satisfies the interface transverse shear stress and displacement continuity, and has a vanishing shear stress on the inner and outer surfaces of the beam. The formulation includes in-plane and rotary inertia effects, and the non-linearity due to the finite deformation of the ring. The governing equations obtained using Lagrange's equations of motion are solved through the direct integration technique. Amplitude-frequency relationships evaluated from the dynamic response history are examined. Detailed numerical results are presented considering various parameters such as radius-to-thickness ratio, circumferential wave number and ovality for isotropic and laminated orthotropic rings. The nature and degree of the participation of various modes in non-linear asymmetric vibration of oval ring brought out through the present study are useful for accurate modelling of the closed non-circular structures.  相似文献

Two strategies for the optimization of centric scan SPRITE (single point ramped imaging with T1 enhancement) magnetic resonance imaging techniques are presented. Point spread functions (PSF) for the centric scan SPRITE methodologies are numerically simulated, and the blurring manifested in a centric scan SPRITE image through PSF convolution is characterized. Optimal choices of imaging parameters and k-space sampling scheme are predicted to obtain maximum signal-to-noise ratio (SNR) while maintaining acceptable image resolution. The point spread function simulation predictions are verified experimentally. The acquisition of multiple FID points following each RF excitation is described and the use of the Chirp z-Transform algorithm for the scaling of field of view (FOV) of the reconstructed images is illustrated. Effective recombination of the rescaled images for SNR improvement and T*2 mapping is demonstrated.  相似文献

A simple sensitive spectrophotometric method has been developed for the determination of atrazine in herbicide formulations and real samples. The method was based on the reaction of atrazine with pyridine to form a quaternary halide which in the presence of alkali forms a carbinol base. The heterocyclic ring of the carbinol base breaks and forms the glutaconic dialdehyde. The glutaconic dialdehyde group was coupled with sulfanilic acid to form a yellow coloured product having λ max 450 nm or coupled with aniline to form a orange red coloured product having λ max 480 nm. The Beer's law was obeyed over the range from 0.1 to 25 µg mL?1 and molar absorptivity 1.5 × 104 L mol?1 cm?1 for sulfanilic acid, and from 0.08 to 12 µg mL?1 and molar absorptivity 1.3 × 104 L mol?1 cm?1 for aniline were observed. The reaction conditions and other analytical parameters were optimised. The proposed method has been successfully applied for the analysis of commercial formulations and real samples.  相似文献
