Cyclisation and cross-linking strategies are important for the synthesis of cyclic and bicyclic peptides. These macrolactams are of great interest due to their increased biological activity compared to linear analogues. Herein, we describe the synthesis of a cyclic peptide containing an Hpi toxicophore, reminiscent of phakellistatins and omphalotins. The first intraannular cross-linking of such a peptide is then presented: using neat TFA to catalyse a Savige-Fontana tryptathionylation, the Hpi-containing peptide is converted to a bicyclic amatoxin analogue. As such, this methodology represents an efficient cyclisation method for cross-linking peptides and exposes a heretofore unrealised relationship between two different classes of peptide natural products. The amatoxins are a family of toxic bicyclic peptides that inhibit RNA polymerase II. Herein we discuss an improved synthesis of these compounds from easily obtainable amino acids by means of a solid-phase methodology. Interestingly, we obtained two products of the same mass following our final macrocyclisation, relating to a similar distribution of products described in some previous reports. One of these products was the desired amatoxin; Pro(2)-Ile(3)-S-deoxo-amaninamide 1 b. The other compound, after thorough investigation, was confirmed to be the epimer Pro(2)-D-allo-Ile(3)-S-deoxo-amaninamide 1 a, not an atropisomer structure as previously suggested in syntheses of related amanitin analogues. Crystallographic data of 1 a confirms the presence of a betaII-turn, rather than a betaI-turn common to the natural toxin and 1 b. This difference explains the large variation in CD spectra, although it seems to have relatively little effect on the bioactivity in vitro. These data provide new insights into the bicyclic amatoxin structure. 相似文献
Like-charged macroions in aqueous electrolyte solution can attract each other because of the presence of inter- and/or intramolecular correlations. Poisson-Boltzmann theory is able to predict attractive interactions if the spatially extended structure (which reflects the presence of intramolecular correlations) of the mobile ions in the electrolyte is accounted for. We demonstrate this for the case of divalent, mobile ions where each ion consists of two individual charges separated by a fixed distance. Variational theory applied to this symmetric 2:2 electrolyte of rodlike ions leads to an integro-differential equation, valid for arbitrary rod length. Numerical solutions reveal the existence of a critical rod length above which electrostatic attraction starts to emerge. This electrostatic attraction is distinct from nonelectrostatic depletion forces. Analysis of the orientational distribution functions suggests a bridging mechanism of the rodlike ions to hold the two macroions together. For sufficiently large rod length, we also observe "overcharging", that is, an over-compensation of the macroion charges by the diffuse layer of mobile rodlike ions. Our results emphasize the importance of the often rodlike internal structure that condensing agents such as polyamines, peptides, or polymer segments exhibit. Certain lipid monolayers at the air-water interface undergo a second-order transition from a tilted to an untilted liquid-crystalline state of their lipid hydrocarbon chains at sufficiently large lateral pressure. Recent experimental observations demonstrate that in the presence of divalent cations DNA adsorbs onto a zwitterionic lipid monolayer and decreases the tilt transition pressure. Lowering of the tilt transition pressure indicates that the DNA condenses the lipid monolayer laterally. To rationalize this finding we analyze a theoretical model that combines a phenomenological Landau approach with an extension of the Poisson-Boltzmann model to zwitterionic lipids. Based on numerical calculations of the mean-field electrostatic free energy of a zwitterionic lipid monolayer-DNA complex in the presence of divalent cations, we analyze the thermodynamic equilibrium of DNA adsorption. We find that adsorbed DNA induces a 10% reduction of the electrostatic contribution to the lateral pressure exerted by the monolayer. This result implies a small but notable decrease in the tilt transition pressure. Additional mechanisms due to ion-ion correlations and headgroup reorientations are likely to further enhance this decrease. 相似文献
We report the interaction between B-type tri-lacunary heteropolyoxotungstate anions and actinyl(V) cations in aqueous solution, yielding a greater understanding of the stability of the O≡An≡O(1+) linear dioxo actinide moiety. Previously we reported that B-α-[BiW(9)O(33)](9-) and B-α-[SbW(9)O(33)](9-) will react with NpO(2)(1+) to yield [(Np(3)W(4)O(15))(H(2)O)(3)(MW(9)O(33))(3)](18-) (M = Bi, or Sb). Single crystal structural characterisation of salts of these complexes revealed a core in which three Np(V) atoms interact with a central W(VI) atom through bridging oxo groups. These bridging oxygen atoms come from one of the two axial oxygens in O≡Np≡O(1+) and represent a highly unusual interaction for a discrete molecular species. In this study visible/near infra-red spectroscopy indicates that [(Np(3)W(4)O(15))(H(2)O)(3)(BiW(9)O(33))(3)](18-) could be readily stabilized in solution at near neutral pH for several months, with (NH(4))(14)Na(4)[(Np(3)W(4)O(15))(H(2)O)(39)BiW(9)O(33))(3)]·62H(2)O crystallising from solution in high yield. At lower pH and [BiW(9)O(33)](9-) : NpO(2)(1+) ratios additional Np(V) species could be observed in solution. Stabilization of [(Np(3)W(4)O(15))(H(2)O)(3)(SbW(9)O(33))(3)](18-) in solution proved more challenging, with several distinctive Np(V) near infra-red transitions observed in solution. Slow complexation kinetics and reduction to Np(IV) was also observed. High [SbW(9)O(33)](9-) : NpO(2)(1+) molar ratios and careful control of solution pH was required to prepare solutions in which [(Np(3)W(4)O(15))(H(2)O)(3)(SbW(9)O(33))(3)](18-) was the only neptunium containing species. In stark contrast to the NpO(2)(1+) chemistry, [BiW(9)O(33)](9-) readily oxidizes PuO(2)(1+) to PuO(2)(2+) yielding further evidence of the decreased stability of Pu(V)vs. Np(V). Np L(II)-edge XAFS measurement revealed very good agreement with single crystal diffraction data for the Np structural environment for [(Np(3)W(4)O(15))(H(2)O)(3)(MW(9)O(33))(3)](18-) (M = Bi, or Sb) in the solid state. There was also good agreement between coordination shells for [(Np(3)W(4)O(15))(H(2)O)(3)(BiW(9)O(33))(3)](18-) in the solid state and in solution, yielding further confirmation of the high stability of this particular cluster. 相似文献

In vivo endocannabinoid (EC) microdialysis has only seldom been performed, mostly in rodent brain tissue. Low solubility in aqueous media, adsorption to surfaces, and instability with co-present human serum albumin (HSA) are the major obstacles in EC microdialysis. The addition of hydroxypropyl-ß-cyclodextrine (HPCD) to the perfusion fluid has been previously described to facilitate lipid microdialysis, but the general biophysical properties of HPCD, especially with respect to peripheral EC microdialysis, have not been described before. We report on the characterization of EC microdialysis using an in vitro system using Ringer’s solution with 10% HPCD as the perfusion fluid and with fatty acid-free HSA as the matrix fluid. The endocannabinoids anandamide (AEA) and 2-arachidonoyl glycerol (2AG) were measured using LC-MS/MS. AEA was stable in the perfusion and matrix fluids, whereas 2AG was only stable in the perfusion fluid. In the matrix fluid, 2AG underwent rapid isomerization to 1-arachidonoyl glycerol. A relative recovery of 3.5% for AEA was found with 10% HPCD in the perfusion fluid and a flow rate of 1 μL/min. For 2AG, a similar relative recovery of 3.5% was estimated. Since 2AG was found unstable in the matrix fluid, a reliable calculation of the relative recovery rates was not possible. Delivery and recovery experiments revealed unequal inward and outward EC transport across the microdialysis membrane. Contrary to usual microdialysis findings, we observed increasing recovery rates for AEA with increasing flow rates. Long equilibration times of several hours were necessary to obtain constant relative recovery rates. In a proof-of-concept study in humans, we collected AEA from subcutaneous abdominal adipose tissue employing the described methodology. Our study suggests that the microdialysis technique is not suitable for the exact quantification of tissue EC concentrations, but it allows for their rough estimation.
High-purity syntheses are reported for a series of first, second, and third row transition metal and actinide hexahalide compounds with equivalent, noncoordinating countercations: (Ph(4)P)(2)TiF(6) (1) and (Ph(4)P)(2)MCl(6) (M = Ti, Zr, Hf, Th, U, Np, Pu; 2-8). While a reaction between MCl(4) (M = Zr, Hf, U) and 2 equiv of Ph(4)PCl provided 3, 4, and 6, syntheses for 1, 2, 5, 7, and 8 required multistep procedures. For example, a cation exchange reaction with Ph(4)PCl and (NH(4))(2)TiF(6) produced 1, which was used in a subsequent anion exchange reaction with Me(3)SiCl to synthesize 2. For 5, 7, and 8, synthetic routes starting with aqueous actinide precursors were developed that circumvented any need for anhydrous Th, Np, or Pu starting materials. The solid-state geometries, bond distances and angles for isolated ThCl(6)(2-), NpCl(6)(2-), and PuCl(6)(2-) anions with noncoordinating counter cations were determined for the first time in the X-ray crystal structures of 5, 7, and 8. Solution phase and solid-state diffuse reflectance spectra were also used to characterize 7 and 8. Transition metal MCl(6)(2-) anions showed the anticipated increase in M-Cl bond distances when changing from M = Ti to Zr, and then a decrease from Zr to Hf. The M-Cl bond distances also decreased from M = Th to U, Np, and Pu. Ionic radii can be used to predict average M-Cl bond distances with reasonable accuracy, which supports a principally ionic model of bonding for each of the (Ph(4)P)(2)MCl(6) complexes. 相似文献
