The enzyme xanthine oxidase (XOD) has been recognized as a key enzyme causing oxidative injury to tissues by ischemia-reperfusion. For this reason, XOD inhibitor, which effectively suppresses this enzyme, plays an important role in the inhibition of many diseases related to reactive oxygen species (ROS). In order to screen XOD inhibitors rapidly and conveniently, a novel assay using flow injection analysis (FIA) was proposed in the present investigation. To optimize the practical FIA system, we studied the effect of the reagent concentrations and the flow condition on the enzymatic reaction, and then selected the optimum condition as follows: 200-mU/ml XOD concentration, 0.5-mM xanthine concentration, 0.5-ml/min flow rate, and 2-m mixing coil length. Under this condition, a typical XOD inhibitor quercetin was determined in the concentration range 0.1 - 1.5 mM at a sampling frequency of 10 samples/h. Using the optimized FIA method, we determined the XOD inhibitory activity of some food samples: onions, apples and teas, which are the high sources of flavonoids known as the potential XOD inhibitors. Among these samples, tea leaves showed the highest activity, the second was onions and the lowest was apples. Based on the result of the assay, not only quercetin, but also other components in investigated samples, contributed to the XOD inhibitory activity.  相似文献

Lithium made a Mn-Mg-O solid solution stable upon exposure of methane, decreased the ethane and ethylene formation rates, and increased the COx formation rate during the oxidative coupling of methane. Studies on gelation in the radical polymerization of diallyl dicarboxylates have been conducted by Simpson,9,11 Gordon,10 and Oiwa.13 However, the results obtained have not always been consistent and are still far from full elucidations. In this paper, the gel point in the polymerization of diallyl aromatic dicarboxylates, including diallyl phthalate (DAP), diallyl isophthalate (DAI), and diallyl terephthalate (DAT) is experimentally reexamined in detail and discussed according to Gordon's theory; the discrepancy between actual and theoretical gel point conversion was quite large and was enhanced in the order DAT > DAI > DAP. Moreover, from detailed inquiry into the primary chain length of the prepolymer it is suggested that the intramolecular chain transfer reaction plays an important role in the polymerization of diallyl ester accompanying the intramolecular cyclization reaction. The polydispersity coefficient (P w,0/P n,0) of the initial prepolymer of DAP is also estimated to be 2.0 from the extrapolation of P w/P n to zero conversion.  相似文献

Time-resolved optical transmission of annealing dye laser pulse (wavelength : 590 nm and pulse width : 250 nsec) itself has been measured in thin layers of Si+-implanted amorphized silicon-on-sapphire (SOS) with several energy densities in the range of 0.02 to 2.64 J/cm2. At the energy densities where the amorphized SOS is recrystallized, the transmission exhibits a sudden drop and subsequently remains at a detectable level. This laser-induced absorption is inconsistent with the presence of ordinary molten silicon but suggests an electronic structural-change from amorphous to crystalline state.  相似文献
