Rapid oligosaccharide synthesis using a fluorous protective group

The Bfp-OH, a novel fluorous protecting reagent, was able to be easily prepared. The Bfp group was readily introduced to a carbohydrate, removed in high yield, and recyclable after cleavage. The use of the Bfp group made it possible to synthesize a pentasaccharide by minimal column chromatography purification. Each synthetic intermediate was able to be easily purified only by simple fluorous-organic solvent extraction and monitored by TLC, NMR, and MS.     

Conformation of poly(<Emphasis Type="SmallCaps">l</Emphasis>-glutamic acid) in bipolar amphiphiles with alkyl ammonium cations of different hydrophobicity

The conformation of the sodium salt of poly(l-glutamic acid) (P(Glu)) in solutions of the cationic bipolar amphiphile 1, 20-isosanediylbis(alkylammonium chloride) (C ₂₀(RA) ₂) with different alkyl head groups as a function of amphiphile concentration was investigated using circular dichroism (CD). RA included methylammonium (MA), ethylammonium (EA), propylammonium (PA), butylammonium (BA), and pentylammonium (PeA) cationic groups. The CD spectrum of each C ₂₀(RA) ₂ had a double minima corresponding to the a-helix of P(Glu), which was replaced by CD spectra with a single minimum at wavelengths larger than 222 nm as concentration increased. These changes in the CD spectrum were ascribed to the conformational change from random coil to a-helix and to aggregates of helices. In C ₂₀(EA) ₂ solutions, a step-like change in the CD intensity was observed at 222 nm as a function of the ratio of C ₂₀(RA) ₂ to P(Glu). At the step, the CD spectrum of the complete a-helix was observed. At 10–35 °C, an a-helix was induced in P(Glu) in the order: C ₂₀(EA) ₂>C ₂₀(MA) ₂>C ₂₀(PA) ₂>C ₂₀(BA) ₂>C ₂₀(PeA) ₂. This order was ascribed to the best fit of ethylammonium to the P(Glu) side chain.     

Interaction of cubosomes with plasma components resulting in the destabilization of cubosomes in plasma

Cubosomes are novel dispersed nanoparticles with bicontinuous cubic phases of monoolein in their interior. We investigated their disintegration process in plasma by in vitro and in vivo studies. Cubosomes were incubated with whole plasma or plasma components such as HDL, LDL, and albumin. The lypolysis study indicated lipolytic activity of whole plasma towards cubosomes. Gel filtration chromatography revealed that HDL, LDL and albumin interacted with cubosomes. HDL affected cubosomes’ integrity and gave rise to smaller particles which contained the components of both cubosomes and HDL. Upon incubation with LDL, cubosomes fused with LDL. Albumin was shown to take up monoolein out of the particles. Cubosomes were disintegrated by whole plasma as a result of the interaction with plasma components. It was concluded that in vivo observation of a long circulation time of a hydrophobic substance in cubosomes was due to the sustained behavior of cubosome remnant particles.     

Observation and analysis of single DNA nano-kinetics by pin-fiber video scope

The nano-kinetic movement of a single DNA molecule was observed and analyzed by a newly developed video-microscope system with an optical fiber, called a pin-fiber video scope. A single lambda-DNA molecule was put in focus using fiber-illumination, and the stretching and shrinking motion was measured. The molecule's kinetics were analyzed by numerical calculations and are discussed. A photocleavage phenomenon of DNA molecules was also visualized by the pin-fiber video scope. The new video-microscope system has the potential to observe and analyze the nano-kinetics of a single molecule.     

Uniaxial alignment of the smallest diamagnetic susceptibility axis using time-dependent magnetic fields

A diamagnetic particle with magnetic susceptibilities chi3 < chi2 = chi1 < 0 was subjected to a rotating magnetic field to obtain an alignment of the chi3 axis (the smallest susceptibility axis) in the direction perpendicular to the plane of the rotating magnetic field. A polymer short fiber, whose fiber axis coincides with the chi3 axis, was suspended in a fluid with the same density, and then a rotating magnetic field generated by a rotation of a pair of permanent magnets was applied. The fiber axis, rotating following the applied field, finally ended up with an alignment perpendicular to the plane of the rotating magnetic field. The experimental data on the time course of the alignment was in good agreement with the numerical calculation based on the equation of rotation.     

Sparsely substituted chlorins as core constructs in chlorophyll analogue chemistry. I. Synthesis

Five routes to stable chlorins bearing 0 or 1 meso substituents have been investigated, among which reaction of a 9-bromo-1-formyldipyrromethane and 2,3,4,5-tetrahydro-1,3,3-trimethyldipyrrin proved most effective. Application of this route afforded metallochlorins [Cu(II), Zn(II), and Pd(II)] including the chlorin lacking any β-pyrrole and meso substituents.     

Three new quassinoids,ailantinol E,F, and G,from Ailanthus altissima

Three new quassinoids, ailantinol E (1), ailantinol F (2), and ailantinol G (3), and related compounds were isolated from Ailanthus altissima grown in Taiwan. Their structures were elucidated from spectral evidence. Each new quassinoid was evaluated for its antitumor promoting effects against Epstein-Barr virus early antigen activation introduced by 12-O-tetradecanoylphorbol-13-acetate in Raji cells. The new quassinoids were found to show potent activity without showing any cytotoxicity. The screening for inhibitors against nitric oxide donor action was also conducted using the new quassinoids and some standard samples.     

Three-component coupling of acylphosphonates and two carbonyl compounds promoted by low-valent samariums: one-Pot synthesis of beta-hydroxyphosphonates

Three-component coupling of acylphosphonates and two carbonyl compounds leading to beta-hydroxyphosphonates has been achieved with low-valent samariums. Thus, acylphosphonates reacted with aldehydes in the presence of semicatalytic amounts of samarium metal or SmI(2) to give acyloxyphosphonates in good yields. The second coupling reaction of the acyloxyphosphonates with aldehydes or ketones promoted by SmI(2) afforded beta-hydroxyphosphonates instead of olefins. Moreover, these two reactions could be carried out in one pot.
