Guanosine derivatives are important for diagnosis of oxidative DNA damage including 8-hydroxy-2'-deoxyguanosine (8-OHdG) as one of the most abundant products of DNA oxidation. This compound is commonly determined in urine, which makes 8-OHdG a good non-invasive marker of oxidation stress. In this study, we optimized and tested the isolation of 8-OHdG from biological matrix by using paramagnetic particles with an antibody-modified surface. 8-OHdG was determined using 1-naphthol generated by alkaline phosphatase conjugated with the secondary antibody. 1-Naphthol was determined by stopped flow injection analysis (SFIA) with electrochemical detector using a glassy carbon working electrode and by stationary electrochemical detection using linear sweep voltammetry. A special modular electrochemical SFIA system which needs only 10 μL of sample including working buffer for one analysis was completely designed and successfully verified. The recoveries in different matrices and analyte concentration were estimated. Detection limit (3 S/N) was estimated as 5 pg/mL of 8-OHdG. This method promises to be very easily modified to microfluidic systems as "lab on valve". The optimized method had sufficient selectivity and thus could be used for determination of 8-OHDG in human urine and therefore for estimation of oxidative DNA damage as a result of oxidation stress in prostate cancer patients.  相似文献

Avidin transgenic plants are a potential tool for providing resistance against various species of insect pests due to the sequestration of vitamin H (biotin) in the plant from the insect pests. In this project we compared three techniques for avidin determination in transgenic tobacco plants, a novel chip-based capillary electrophoretic method (Experion), classical polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulphate (SDS-PAGE) and a square wave voltammetric method using a carbon paste electrode. We determined that the automated chip-based capillary electrophoretic method is rapid, sensitive and the results obtained are well repeatable. The avidin content measured in transgenic tobacco leaves using chip-based capillary electrophoresis varied from 15 to 854 ng per mg of fresh mass depending on the individual plant.  相似文献

We study a very simple model of correlations and intermittency of identical final state pions in hadronic collisions. Final state pions are either products of resonance decays or they are “directly” produced. The “direct” production is simulated by an immediate decay of a resonance. For “direct pions” forming about a half of final state pions and for formation times of resonances less than 0.5fin/c we get density of sources which via Hanbury-Brown and Twiss interference leads to correlations of two identical pions consistent with recent data and shows intermittency patterns for the second factorial moment. The essential ingredient of the scheme is the combination of pions from resonance decays and direct pions. Due to life-times of resonances taken from experiment, pions from resonance decays are responsible for short-range correlations in the longitudinal momentum, whereas directly produced pions, due to their fast production, dominate in the region of longitudinal momentum difference of the order of 100 MeV/c. The combination of both can give an approximate scaling leading to intermittency.  相似文献

The present paper describes the synthesis, characterization, and utilization of multi-functional magnetic conjugates that integrate optical and magnetic properties in a single structure for use in many biomedical applications. Spontaneous interaction with eukaryotic cell membrane (HEK-239 cell culture) was determined using fluorescence microscopy, and fluorescence analyses. Both, differences in excitation, and emission wavelength were observed, caused by glutathione intake by cells, resulting in disintegration of core–shell structure of quantum dots, as well as adhesion of conjugate onto cell surface. When compared with quantum dots fluorescent properties, HEK-239 cells with incorporated nanoconjugate exhibited two excitation maxima (λ ex = 430 and 390 nm). Simultaneously, application of ideal λ ex for quantum dots (λ ex = 430 nm), resulted in two emission maxima (λ = 740 and 750 nm). This nanoconjugate fulfills the requirements of term theranostics, because it can be further functionalized with biomolecules as DNA, proteins, peptides or antibodies, and thus serves as a tool for therapy in combination with simultaneous treatment.  相似文献

The formation of quark-gluon plasma in heavy ion collisions should manifest itself by the increase of the ratio of medium to large mass dileptons, the former being produced predominantly in the QGP and the mixed phase whereas the latter are produced mostly by the Drell-Yan process. The increase of this ratio should be visible either when comparing collisions of lighter with heavier ions, e.g. O+O with Pb+Pb or when comparing low and large transverse energy events in heavy ion e.g. Pb+Pb collisions.  相似文献

We compared the suitability of avidin and streptavidin for avidin‐biotin technology in view of the sensitivity of the analysis using square‐wave voltammetry. We found out during our preliminary experiments that streptavidin gave approximately 100 times higher electrochemical response in comparison with avidin at the same experimental conditions and concentration. Thus, we used two approaches for streptavidin determination – analysis directly in electrochemical cell and analysis by adsorptive transfer technique (AdTS). Ten minutes long accumulation on carbon paste electrode surface was ascertained as optimal in both cases. Limits of detection were 0.3 aM (electrochemical cell) and 30 aM (AdTS).  相似文献

Prostate cancer with altered zinc(II) cell metabolism is the second most frequently diagnosed cancer in developed countries. The alterations of zinc(II) metabolism can influence metabolism of other metal ions and can also be associated with the expression and translation of metal-binding proteins including metallothioneins. The aim of this article was to optimize immunoseparation protocol based on paramagnetic beads conjugated with protein G for the isolation of metallothionein. Isolated metallothionein was determined by differential pulse voltammetry Brdicka reaction and SDS-PAGE. Optimal conditions: antigen-binding time - 60 min, temperature - 70°C, and buffer composition and pH - acetate buffer, pH 4.3, were determined. Under the optimized conditions, lysates from 22Rv1 prostate cancer cells treated with various concentrations of cadmium(II) and copper(II) ions were analyzed. We observed strong correlation in all experimental groups and all lysate types (r>0.83 at p<0.041) between metallothionein concentration related to viability and concentration of copper(II) ions and cadmium(II) ions in medium. Moreover, the results were compared with standard sample preparation as heat treatment and SDS-PAGE analysis.  相似文献
