Isomerization of aldehyde-2,4-dinitrophenylhydrazone derivatives and validation of high-performance liquid chromatographic analysis

The most widely used method for qualitative and quantitative analysis of carbonyl compounds is the 2,4-dinitrophenylhydrazine method through the formation of 2,4-dinitrophenylhydrazone derivatives. However, this method may cause an analytical error because 2,4-dinitrophenylhydrazones have both E- and Z-stereoisomers. Purified aldehyde-2,4-dinitrophenylhydrazone demonstrated only the E-isomer. However under UV irradiation and the addition of acid, both E- and Z-isomers were seen. The spectral patterns of Z-isomers were different from those of E-isomers and the absorption maximum wavelengths were shifted towards shorter wavelengths by 5-8 nm. An equilibrium Z/E isomer ratio was observed in 0.02-0.2% (v/v) phosphoric acid solutions. In the case of acetaldehyde- and propanal-2,4-dinitrophenylhydrazones, the equilibrium Z/E isomer ratios were 0.32 and 0.14, respectively. However, when irradiated with ultraviolet light at 364 nm, the isomer ratios were increased beyond this constant ratio and reached 0.55 and 0.33, respectively. Zero-order rates for decreases of aldehyde derivatives were observed under UV irradiation (364 nm), however the decreases of concentration were not observed in phosphoric acid solutions. The best method for the determination of aldehyde-2,4-dinitrophenylhydrazones by HPLC is to add phosphoric acid to both the sample and the standard solution, to form a 0.02-1% acid solution.     

Measurement of the product ion ratio N4+/N3+ in the radiolysis of nitrogen

The ratio of N₄⁺ to N₃⁺ formed in the radiolysis of gaseous nitrogen has been measured to be 4.7 ± 0.4 using a time-resolved atmospheric pressure ionization mass spectrometer. The limit of error has been evaluated from the ion mass discrimination of the apparatus.     

Studies of the polymerization of diallyl compounds. XXII. Polymerization of diallyl oxalate in the evolution of carbon dioxide at elevated temperatures

The polymerization of diallyl oxalate was conducted in the presence of radical initiators at a high temperature range of 80–180°C; a large decrease in degree of polymerization, an increase in residual unsaturation of the resulting polymer, and the evolution of carbon dioxide were observed with the elevation of temperature. These findings were reasonably interpreted by considering the dismutation of the uncyclized growing radical to yield the allyl radical, carbon dioxide, and polymer carrying a terminal double bond. The kinetics of the polymerization of diallyl oxalate in the evolution of carbon dioxide at elevated temperatures were also discussed in detail.     

The synthesis of hexahydrooxoepithiopyridinedicarboximides by the reaction of thioamides with N-substituted maleimides

The synthesis of hexahydrooxoepithiopyridinedicarboxyimide (5: X₂ = N-Ph) by the reaction of thioamides 1 with N-substituted maleimide ( 2a ) was examined. The reaction of primary thioamides, such as thiobenzamide and p-toluthioamide with N-phenylmaleimide gives compounds 5 together with corresponding 4-hydroxy-1,3-thiazoles 4 . However, a similar reaction of secondary thioamides, such as N-methylthioacetamide, thiobenzanilide, with N-phenylmaleimide did not provide compounds 5 without addition of acid. The reaction pathway and the configuration of 5 were also investigated.     

Stereoselective synthesis of (Z)-2-fluoro-1-alkenyl(phenyl)iodonium tetrafluoroborates

[reaction: see text] (Z)-2-Fluoro-1-alkenyl(phenyl)iodonium salts were stereoselectively prepared by the reaction of alkynyl(phenyl)iodonium salts with aqueous HF in good yields. The method is applicable to the synthesis of fluoroalkenyliodonium salts having functional groups such as ketone, ester, and chloride. (Z)-2-Fluoro-1-alkene, (Z)-2-fluoro-2-alkenoate, and (Z)-beta-fluoroenyne could be stereoselectively prepared from the fluoroalkenyliodonium salt.     

1,3-Dipolar cycloaddition reaction of phenanthridinium N-imines and N-benzoylimine with acetylenic compounds

N-Aminophenanthridinium salt reacted with dimethyl acetylenedicarboxylate in the presence of potassium carbonate to give a 3,3a-dihydropyrazolo[1,5-f]phenanthridine but with mono-substituted acetylenes (ethyl propiolate and benzoylacetylene) it produced aromatized pyrazolo-[1,5-f]phenanthridines (XII and XIII). The reaction of the N-benzoylimine with di-substituted acetylenes (dimethyl acetylenedicarboxylate and methyl phenylpropiolate) and mono-substituted acetylenes (ethyl propiolate and benzoylacetylene) yielded 1,3a-dihydropyrazolo[1,5-f]phenanthridines and the aromatized products (XII and XIII), respectively.     

Synthesis of 1-alkylselenocyclobutene via intermediate allenyl selenoketene

Reactions of 2-alkynyl arylethynyl selenide 1 with alkyl iodides 3 in the presence of lithium aluminium hydride via allenyl selenoketene 2 afforded cyclobutene 4. Allenyl group of the intermediate allenyl selenoketene 2 was monitored by React IR.     

Profiling and comprehensive expression analysis of ABC transporter solute-binding proteins of Bacillus subtilis membrane based on a proteomic approach

We analyzed ABC transporter solute-binding proteins (SBPs) of the Bacillus subtilis membrane using a proteomic approach. We prepared a washed cell membrane fraction that was insoluble in 134 mM nondetergent sulfobetaine and then extracted proteins using mixtures of detergents in a stepwise manner. The membrane proteins were resolved by three two-dimensional polyacrylamide gel electrophoresis (2-D PAGE) or two one-dimensional (1-D) PAGE procedures, electroblotted, and digested in the presence of 5% or 80% acetonitrile. Thereafter, matrix-assisted laser desorption/ionization-time of flight-mass spectrometry (MALDI-TOF MS) identified 637 proteins corresponding to 15.9% of the total cellular proteins. We predicted that among these, 256 were membrane proteins, 101 were lipoproteins or secretory proteins and 280 were soluble proteins containing peripheral proteins that function in both the cytoplasm and the cell membrane such as SecA and FtsY. Among the 637 proteins, we identified 30 SBPs among 38 importers predicted by a bioinformatic search of the genome. We confirmed expression of the genes for the 30 SBPs using DNA microarray analysis. We compared the 2-D gel separation profiles of submembrane fractions solubilized by 1% n-dodecyl-beta-D-maltoside from cells cultured on Luria Bertani (LB), S7, and S7 medium without glutamate as well as DNA microarray data on LB and S7. The results suggested that YcdH, YtmK and YurO are binding proteins for Mn(++), glutamate and glucose, respectively, and that YqiX and YxeM are binding proteins for amino acids (tryptophan in S7 medium).     

Thermally stable rigid π-allylnickel compounds, (π-CHRCR′CR2)Ni(PPhMe2)C6Cl5

The reaction of C₆Cl₅Ni(PPhMe₂)₂Cl with CHRCR′CH₂MgX (X = Br or Cl) yields π-allylnickel compounds, (π-CHRCR′CH₂)Ni(PPhMe₂)C₆Cl₅ (Ia, R = R′ = H; Ib, R = H, R′ = CH₃; Ic, R = CH₃, R′ = H), which are stable in the solid state below ca. 150°C and are fairly stable in solution in the absence of oxygen. The π-allyl group was found by PMR spectroscopy to be rigid even in the presence of an excess of PPhMe₂, P(OEt)₃ or P(OMe)₃.     

SbF5/PAF—a novel fluorinating reagent in preparing fluorine compounds

A novel fluorination reagent and catalyst, SbF₅/PAF (porous aluminum fluoride), was prepared by impregnating SbCl₅ into PAF and then treating with anhydrous hydrogen fluoride. The prepared reagent had an excellent catalytic activity in halogen-exchange, and also improved the properties of SbF₅, such as hydroscopicity, corrosion, and toxicity. SbF₅/PAF was successfully used in organic synthesis as a fluorinating reagent, and a fixed bed catalyst for F/Cl exchange.
