Crystallization of Arthrobacter sp. strain 1C N-(1-D-carboxyethyl)-L-norvaline dehydrogenase and its complex with NAD+

The novel NAD+-linked opine dehydrogenase from a soil isolate Arthrobacter sp. strain 1C belongs to an enzyme superfamily whose members exhibit quite diverse substrate specificites. Crystals of this opine dehydrogenase, obtained in the presence or absence of co-factor and substrates, have been shown to diffract to beyond 1.8 ? resolution. X-ray precession photographs have established that the crystals belong to space group P21212, with cell parameters a = 104.9, b = 80.0, c = 45.5 ? and a single subunit in the asymmetric unit. The elucidation of the three-dimensional structure of this enzyme will provide a structural framework for this novel class of dehydrogenases to enable a comparison to be made with other enzyme families and also as the basis for mutagenesis experiments directed towards the production of natural and synthetic opine-type compounds containing two chiral centres.     

Mass spectrometric studies of alkyl-, phenyl- and chloropyrimidines

The mass spectra of alkyl- and chloropyrimidines are recorded. A generalised fragmentation mechanism for the derivatives of pyrimidine is proposed. The principle of α-cleavage for heterocyclic compounds as proposed in the previous paper is shown to be valid for pyrimidine derivatives. Electronic structures of some molecular and intermediate ions are calculated by the openshell CNDO/2 and INDO methods.     

Synthesis of polystyrene latex with amphoteric surfactant and its characterization

An emulsion polymerization of styrene in the presence of an amphoteric surfactant; N,N-dimethyl-n-laurylbetaine (LNB) was carried out at pH 7.0. The polymerization rate and the concentration of the latex particle were proportional to the LNB concentration of 0.6 power. The latex particle sizes became smaller with increasing concentrations of LNB. The molecular weights of polystyrene latices increased with the concentration of LNB. The zeta-potentials of latex particles were significantly dependent on the pH, and showed the existence of an isoelectric point.     

Semidifferential electroanalysis with a solid working electrode

Semidifferential electroanalysis is described for hexacyanoferrate(III), dichromate, copper(II), p-aminophenol, p-benzoquinone, m-dinitrobenzene, guanine, guanosine, adenine, and adenosine at a stationary solid working electrode. Nearly symmetrical, peaked curves are obtained for the electrode processes of all the samples investigated. The predicted dependence of peak height and peak potential on concentration, electrode area, and potential scan rate are confirmed-experimentally for the glassy carbon disk electrode. It is demonstrated that the technique with the solid working electrode provides higher sensitivity and better resolution than ordinary linear sweep voltammetry. The sensitivity is somewhat worse than in differential pulse voltammetry, but the technique has the advantage of Speed.     

Two-dimensional mapping by high-performance liquid chromatography of pyridylamino oligosaccharides from various glycosphingolipids.

A method to map sugars two-dimensionally for the analysis of the structures of oligosaccharides from glycosphingolipids is described. Nine neutral and ten acidic oligosaccharides were obtained from glycosphingolipids by endoglycoceramidase digestion and labelled with 2-aminopyridine. The pyridylamino oligosaccharides were clearly separated by high-performance liquid chromatography on commercially available C18-silica and amide-silica column. All compounds tested were mapped without any overlapping. The separation of the pyridylamino oligosaccharides on the C18-silica column depended on the numbers and positions of sialic acid and N-acetylhexosamine residues; on the amide-silica column, the separation depended on the total number of sugar residues.
