5,5′-Bis(2-phenylethynyl)-2,2′-bithiophene derivative 1 containing dendritic moieties linked through amide groups was synthesized. Compound 1 exhibits a rectangular columnar phase on slow cooling from the isotropic liquid. In contrast, a cubic phase is formed as a metastable liquid-crystalline phase by rapid cooling of the isotropic melt. Mechanical shearing induces a cubic–columnar phase transition, accompanied by the change of a photoluminescent color from yellow-green to green. The infrared spectra of 1 suggest that the mechanical stimuli can induce the partial dissociation of the hydrogen bond. This may lead to more disordering of the π-stacks of chromophores.  相似文献

The Strebel point is a Teichm ¨uller equivalence class in the Teichm ¨uller space that has a certain rigidity in the extremality of the maximal dilatation. In this paper,we give a sufficient condition in terms of the Schwarzian derivative for a Teichm ¨uller equivalence class of the universal Teichm ¨uller space under which the class is a Strebel point. As an application, we construct a Teichm ¨uller equivalence class that is a Strebel point and that is not an asymptotically conformal class.  相似文献

We describe the potential of microchip electrophoresis with a Hitachi SV1210, which can be used to evaluate the integrity of total RNA, for the analysis of mRNA expression. The ribonuclease (RNase) protection assay was performed by using microchip electrophoresis with cyanine 5 (Cy5) labeled 248-base antisense RNA probe (riboprobe) encoding adipose-type fatty acid binding protein (A-FABP) as the riboprobe. The fluorescence intensity corresponding to the protected RNA fragment increased in a dose-dependent manner with respect to the complementary strand RNA. Results were obtained in 120 s, and the same amount of Cy5-labeled antisense riboprobe as used in the conventional method can be used. Furthermore, 8 times more sensitive detection of mRNA by microchip electrophoresis could be obtained. An obvious increase in the mRNA expression of A-FABP, which is known as a differentiation marker of adipocytes, occurred during the adipocyte differentiation of 3T3-L1 cells. These results clearly indicate the potential of microchip electrophoresis for the analysis of mRNA expression in cells.  相似文献

To find candidates with high antimicrobial and low hemolytic activities, many gramicidin S (GS) analogs of various ring sizes have been designed and synthesized. However, syntheses of antimicrobially active analogues of GS having a disordered symmetry structure from C(2) have almost never been reported, because the stable, amphiphilic β-sheet structure of GS with C(2) symmetry is considered essential for its strong antibacterial activity. In the present studies, novel thirteen cycloundecapeptides 1-13 related to GS were synthesized and examined. Among them, cyclo(-Va1(1)-Orn(2)-Leu(3)-D-Phe(4)-X(5)-Pro(6)-Val(7)-Orn(8)-Leu(9)-D-Phe(10)-Pro(11)-) (X=Lys (10), Orn (11), Arg (12) and Lys(Lys) (13)) resulted in high antibiotic activity against both Gram-positive and Gram-negative microorganisms tested. In addition, 11 showed low toxicity against sheep blood cells compared with that of GS. Further, circular dichroism (CD) spectra of 10-13 had a curve similar to each other, suggesting that the conformations of these analogues in methanol are similar to each other. However, CD spectra of 10-13 were different from that of GS in the 190-210 nm region. These results suggest that the presences of one added amino acid residue at position 5 of 10-13 might be partially effective through a structural change in the biological activity of 10-13. In addition, the structural modifications at position 5 lower the undesirable hemolytic activity and enhance the desirable antibiotic activity.  相似文献

Recently, increasing evidence suggests that the antihypertensive drug nifedipine acts as a protective agent for endothelial cells, and that the activity is unrelated to its calcium channel blocking. Nifedipine is unstable under light and reportedly decomposes to a stable nitrosonifedipine (NO-NIF). NO-NIF has no antihypertensive effect, and it has been recognized as a contaminant of nifedipine. The present study for the first time demonstrated that NO-NIF changed to a NO-NIF radical in a time-dependent manner when it interacted with human umbilical vein endothelial cells (HUVECs). The electron paramagnetic resonance (EPR) signal of NO-NIF radicals in HUVECs showed an asymmetric pattern suggesting that the radicals were located in the membrane. The NO-NIF radicals had radical scavenging activity for 1,1-diphenyl-2-picrylhydrazyl, whereas neither NO-NIF nor nifedipine did. In addition, the NO-NIF radical more effectively quenched lipid peroxides than NO-NIF or nifedipine. Furthermore, NO-NIF attenuated the superoxide-derived free radicals in HUVECs stimulated with LY83583, and suppressed iron-nitrilotriacetic acid (Fe-NTA)-induced cytotoxicity in rat pheochromocytoma (PC12) cells. Our findings suggest that NO-NIF is a candidate for a new class of antioxidative drugs that protect cells against oxidative stress.  相似文献

By using three different hybrid exchange-correlation functionals containing 20%, 35%, and 50% of exact Hartree-Fock (HF) exchange of the density functional theory and its time-dependent extension plus the Hartree-Fock and the configuration interaction of single excitation methods, equilibrium geometries, and their 30 vibrational-normal-mode frequencies of the ground S(0)((1)A(1)) and the first excited S(1)((1)B(2)) states of fluorobenzene (FB) were calculated. The dispersed fluorescence spectrum and internal conversion (IC) rate of the A(1)B(2)→X(1)A(1) transition were simulated by Franck-Condon (FC) calculations within the displaced harmonic oscillator approximation plus anharmonic and distorted corrections. The simulated spectral profile is primarily described by the Franck-Condon progression from the ring-breathing modes ν(9) and ν(10) which belong to totally symmetry modes. Anharmonic corrections simultaneously improve the intensity order of 9(1)(0) and 10(1)(0) bands and diminish 1(1)(0) transition that is fairly strong in harmonic simulations. It is concluded that the amount of Hartree-Fock exchange does impact the geometries and vibrational frequencies of FB molecule, but not the relative intensities of the transitions. It is anharmonic corrections that make the relative intensities of the transitions in good agreement with experimental results. Distorted corrections could assign most of the dominant overtones of out-of-plane nontotally symmetry modes, and the results agree well with the experimental assignments. Furthermore, it was found that the internal conversion rate is dominated by three promoting modes that are computed with lowing symmetry to C(1). By choosing dephasing width as 10 cm(-1) that is consistent with spectral simulation, we obtained the lifetimes of the A(1)B(2)→X(1)A(1) de-excitation as 11 and 19 ns, respectively, from TD(B3LYP) and HF∕CIS calculations in comparison with the experimental value 14.75 ns.  相似文献

We have recently proposed a measure of the thermal stability of a protein: the water-entropy gain at 25?°C upon folding normalized by the number of residues, which is calculated using a hybrid of the angle-dependent integral equation theory combined with the multipolar water model and the morphometric approach. A protein with a larger value of the measure is thermally more stable. Here we extend the study to analyses on the effects of heme on the thermal stability of four cytochromes c (PA c(551), PH c(552), HT c(552), and AA c(555)) whose denaturation temperatures are considerably different from one another despite that they share significantly high sequence homology and similar three-dimensional folds. The major conclusions are as follows. For all the four cytochromes c, the thermal stability is largely enhanced by the heme binding in terms of the water entropy. For the holo states, the measure is the largest for AA c(555). However, AA c(555) has the lowest packing efficiency of heme and the apo polypeptide with hololike structure, which is unfavorable for the water entropy. The highest stability of AA c(555) is ascribed primarily to the highest efficiency of side-chain packing of the apo polypeptide itself. We argue for all the four cytochromes c that due to covalent heme linkages, the number of accessible conformations of the denatured state is decreased by the steric hindrance of heme, and the conformational-entropy loss upon folding becomes smaller, leading to an enhancement of the thermal stability. As for the apo state modeled as the native structure whose heme is removed, AA c(555) has a much larger value of the measure than the other three. Overall, the theoretical results are quite consistent with the experimental observations (e.g., at 25?°C the α-helix content of the apo state of AA c(555) is almost equal to that of the holo state while almost all helices are collapsed in the apo states of PA c(551), PH c(552), and HT c(552)).  相似文献

We report on theoretical Auger electron kinetic energy distribution originated from sequential two-step Auger decays of molecular double core-hole (DCH) state, using CH(4), NH(3), and H(2)CO molecules as representative examples. For CH(4) and NH(3) molecules, the DCH state has an empty 1s inner-shell orbital and its Auger spectrum has two well-separated components. One is originated from the 1st Auger transition from the DCH state to the triply ionized states with one core hole and two valence holes (CVV states) and the other is originated from the 2nd Auger transition from the CVV states to quadruply valence ionized (VVVV) states. Our result on the NH(3) Auger spectrum is consistent with the experimental spectrum of the DCH Auger decay observed recently [J. H. D. Eland, M. Tashiro, P. Linusson, M. Ehara, K. Ueda, and R. Feifel, Phys. Rev. Lett. 105, 213005 (2010)]. In contrast to CH(4) and NH(3) molecules, H(2)CO has four different DCH states with C1s(-2), O1s(-2), and C1s(-1)O1s(-1) (singlet and triplet) configurations, and its Auger spectrum has more complicated structure compared to the Auger spectra of CH(4) and NH(3) molecules. In the H(2)CO Auger spectra, the C1s(-1)O1s(-1) DCH → CVV Auger spectrum and the CVV → VVVV Auger spectrum overlap each other, which suggests that isolation of these Auger components may be difficult in experiment. The C1s(-2) and O1s(-2) DCH → CVV Auger components are separated from the other components in the H(2)CO Auger spectra and can be observed in experiment. Two-dimensional Auger spectrum, representing a probability of finding two Auger electrons at specific pair of energies, may be obtained by four-electron coincidence detection technique in experiment. Our calculation shows that this two-dimensional spectrum is useful in understanding contributions of CVV and VVVV states to the Auger decay of molecular DCH states.  相似文献

A rapid and diverse synthesis of biologically important 6-substituted-5,6-dihydro-4-hydroxy-2-pyrones through a double Reformatsky reaction of aldehydes to δ-hydroxy-β-ketoesters followed by lactonization is described. Due to the high functional group tolerance and reaction site discrimination between aldehyde, nitrile, and ester groups in the substrate, the protocol can provide the dihydropyrones with bromo, nitro, carboxylic acid, and β-ketoester groups, which are suitable for the further derivatizations. Furthermore, the protocol has been successfully applied to the rapid total synthesis of naturally occurring Yangonin.  相似文献

Gas-flow ionization chambers for radioisotope (RI) monitoring systems at RI institutes throughout Japan are commonly used to measure RIs which leak from the RI institutes. Before the Japan’s 2011 Tohoku earthquake [11 March 2011, moment magnitude (M w) 9.0], ionization current data measured with a gas-flow ionization chamber at the RI institute of Fukushima Medical University were found to change. The question we must raise is whether the variation ionization current can be considered to the variation of outdoor radon concentration. The conversion factors (from ionization current to radon concentration in air) of the gas-flow ionization chamber can be obtained by measuring four levels of radon concentration (outdoor air, indoor air, high level and radon-free gas) with an AlphaGUARD monitor and the chamber itself. The two gas-flow ionization chambers consist of the air intake and terminal exhaust duct of the RI institute. It was found that the radon concentration in the exhaust air was the same as that in the air intake. This study provided evidence that variations of outdoor radon concentration could be determined using gas-flow ionization chambers for RI monitoring systems.  相似文献
