Rhodium-catalyzed cyclization of 1,6-enynes triggered by addition of arylboronic acids

1,6-Enynes reacted with arylboronic acids in the presence of a catalytic amount of a rhodium(I) complex under mild conditions to give (Z)-1-(1-arylethylidene)-2-vinylcyclopentanes. The regioselective addition of an arylrhodium(I) species across the carbon-carbon triple bond triggered the cyclization process. Intramolecular carborhodation onto the pendent alkene in a 5-exo mode furnished a five-membered ring. Finally, the rhodium(I) methoxide generated by beta-methoxy elimination reacted with the arylboronic acid to promote the next catalytic cycle.     

A study on LH- and FSH-RIA kits by immunoradiometric assay

An evaluation of LH- and FSH-RIA BEAD kits based on IRMA was carried out. The results obtained with the methods characterized by the use of monoclonal antibodies, i.e., one is linked to solid phase, and another is isotopically labeled, were compared with those determined by the Daiichi LH- and FSH-kits. Intra- and inter-assay precision, recovery, linearity, and specificity of both methods were favorable without exceptions. The cross reactivity of the LH kit to 5,000 mIU/ml hCG revealed within the range of less than 3 mIU/ml. Significant correlations were observed between the results derived from conventional Daiichi LH- and FSH-kits. The results from the conventional kits exhibited 30 to 40% of those from the Daiichi kits, considered to be mainly due to the difference in standard calibrations used. Among the individuals within the normal menstrual cycle, the serum LH- and FSH-levels determined by the present kits gave a typical pattern with a peak in the preovulatory phase. On the other hand, the LH- and FSH-values of individuals in normal pregnancy revealed strikingly decreased in comparison with those of non-pregnant women.     

Improvement of some pharmaceutical properties of nocloprost by β- and γ-Cyclodextrin Complexation

Inclusion complexation of nocloprost, a potent antiulcer prostaglandin derivative, with -, -, and -cyclodextrins (CyDs) in aqueous solutions has been studied by the solubility method and¹³C-NMR spectroscopy. The steric requirement of host-guest interaction was reflected in the magnitude of the stability constants and the thermodynamic parameters of the inclusion complexes. Solid complexes of nocloprost with - and -CyDs in a molar ratio of 1 : 2 were obtained on the basis of aBs-type phase solubility diagram. The X-ray diffraction data suggested that nocloprost is included in the cylindrical channel formed by coaxial alignment of -CyD molecules to give a channel type structure. Release and thermal behavior of the solid complexes was examined and compared with nocloprost itself. The results indicated that the -CyD complex may have great utility among the three CyDs, being a rapid dissolving form of nocloprost with improved thermal stability.     

A novel synthesis of (+)-biotin from L-cysteine

(+)-Biotin (1) was synthesized in 25% overall yield over 11 steps from L-cysteine. The contiguous asymmetric centers at C-3a and C-6a were formed through a novel and highly stereoselective Lewis base-catalyzed cyanosilylation of alpha-amino aldehyde 3 to provide anti-O-TMS-cyanohydrin 4 with high stereoselectivity and in high yield (anti/syn = 92:8, 96%). Treatment of 4 with a di-Grignard reagent, 1,4-bis(bromomagnesio)butane, followed by carbon dioxide, efficiently installed the 4-carboxybutyl chain at C-4 to give keto acid 5. The final cyclization to bicyclic compound 7b, a precursor to 1, was realized by a palladium-catalyzed intramolecular allylic amination of cis-allylic carbonate 6b that was elaborated from 5.     

Effects of various thiol molecules added on morphology of dendrimer-gold nanocomposites

Interactions between poly(amidoamine) dendrimer (PAMAM)-gold nanocomposites and alkanethiols and between the former nanocomposites and thiol-modified poly(amidoamine) dendrons in ethyl acetate were investigated by adding alkanethiols, such as 1-propanethiol and 1,3-propanedithiol, and thiol-modified poly(amidoamine) dendrons, generations 0.5 and 2.5 (G0.5-SH and G2.5-SH). The PAMAM dendrimers with surface methyl ester groups used were generations 1.5 and 5.5 (G1.5 and G5.5). The mean particle sizes of PAMAM-gold nanocomposites were about 2.1 for G1.5 and 2.4 nm for G5.5. In both nanocomposite systems where 1-propanethiol and 1,3-propanedithiol were added, the mean particle size was about 4 nm, twice that of the systems where these thiols were not added. Increasing the addition of 1,3-propanedithiol made the average particle size smaller for both nanocomposites systems. To compare with alkanethiol, thiol-modified poly(amidoamine) dendron with a highly branched structure on one side was synthesized. Using G2.5-SH as a protective agent, dendron-gold nanocomposites with mean diameters of 3 to 4 nm were obtained. The difference in particle size was seen only when the combination of PAMAM-gold nanocomposites and thiol-modified dendron was less sterically dense, modified dendron (G0.5-SH). The mechanisms for morphology changes in the dendrimer-gold nanocomposites by the addition of these thiols are discussed.     

Significance of anti-HBc IgM in acute hepatitis and hepatitis B associated chronic liver disease

To determine the diagnostic value of anti-HBc IgM in acute viral hepatitis or chronic liver disease B, Anti-HBc IgM was measured by a RIA and an ELISA in 32 patients with acute hepatitis (4 with type A, 15 with type B and 13 with type non A non B), 18 patients with chronic hepatitis and 19 patients with liver cirrhosis. In acute hepatitis B, anti-HBc IgM (both RIA and ELISA) was positive in 14(93%) of 15 patients and its cut-off index value was very high. However, anti-HBc IgM was always negative in one patient with typical course of type B. In 1 of 4 patients with acute hepatitis A and 2 of 13 with non A non B, anti-HBc IgM (RIA and/or ELISA) was positive. These 3 patients were positive for anti-HBs at the onset of disease, so we could not made the diagnosis of acute hepatitis B. Anti-HBc IgM was positive in 21(51%) of 37 patients with HBsAg-positive chronic liver disease by RIA and in 11 (30%) by ELISA, and its cut-off index value was relatively low. These results suggest that when adequate cut-off index value is established, anti-HBc analysis is useful for differentiating recent and current infections from remote infections.     

Dianion of sulfinylacetone as a synthetic equivalent of beta-enolate of propionic acid: a novel synthesis of carboxylic acids from alkyl halides with three-carbon elongation

The reaction of the dianion of phenylsulfinylacetone with alkyl halides afforded beta-keto sulfoxides, which were first chlorinated with hexachloroethane and then treated successively with KH and t-BuLi to give carboxylic acids in three-steps in moderate overall yields from the alkyl halides. This procedure affords a good method for a synthesis of carboxylic acids from alkyl halides with three-carbon elongation.     

Ligand Substitution Reaction of Bis(ethylenediamine)glycinatocobalt(III) Complex with Ethylenediamine Catalyzed by the Photo-excited Tris(2,2′-bipyridine)ruthenium(II) Complex

Substitution reaction with ethylenediamine of coordinated glycinate ligand in bis(ethylenediamine)-glycinatocobalt(III) complex has been studied in the presence of photo-excited tris(2,2′-bipyridine)ruthenium(II) complex in alkaline aqueous solution (buffered around pH 12) containing 1.0M chloride ion at 25°C. VIS absorption and CD spectra were used for the racemate and the optically active isomers of the Co(III) complexes, respectively. The reaction was catalyzed by the excited Ru(II) complex to give tris(ethylenediamine)cobalt(III) complex. Mechanism of the ligand-substitution reaction and role of the excited Ru(II) complex were discussed.     

Deuterium relaxation time ofα-d-tryptophan included in cyclodextrin host molecules

Deuterium relaxation times ofd- andl--d-tryptophan included in -cyclodextrin derivatives were directly measured by deuterium NMR spectroscopy. The results showed that the molecular motion of the tryptophan molecule was strongly restricted even in the cavity of unmodified -cyclodextrin and the additional recognition groupings — ammonium and carboxylate — on -cyclodextrin did not affect the molecular motion of tryptophan, though the association constants were significantly enhanced.Deceased on March 22, 1987.
