This work utilizes on-column ligand synthesis and affinity capillary electrophoresis (ACE) to determine binding constants (Kb) of 9-flourenylmethyloxy carbonyl (Fmoc)-amino acid derivatives to the glycopeptide antibiotics ristocetin (Rist) and teicoplanin (Teic). In this technique, two separate plugs of sample are injected on to the capillary column and electrophoresed. The initial sample plug contains a d-Ala-d-Ala terminus peptide and either one or two non-interacting standard(s). The second plug contains a Fmoc-amino acid-N-hydroxysuccinimide (NHS) ester. The electrophoresis is then carried out with an increasing concentration of Rist or Teic in the running buffer. Upon electrophoresis the initial d-Ala-d-Ala peptide reacts with the Fmoc-amino acid yielding a new Fmoc-amino acid-d-Ala-d-Ala peptide derivative. Continued electrophoresis results in the binding of Rist or Teic to the Fmoc-amino acid-d-Ala-d-Ala peptide derivatives. Analysis of the change in the relative migration time ratio (RMTR) or electrophoretic mobility () of the Fmoc-amino acid-d-Ala-d-Ala peptide derivatives relative to the non-interacting standards, as a function of the concentration of Rist and Teic, yields a value for
Kb. These findings demonstrate the advantage of coupling on-column ligand synthesis to ACE for estimating binding parameters between antibiotics and ligands.Abbreviations Rist

Characterization and determination of a complex mixture of fatty acid methyl esters was performed for commercial fish oil using two-dimensional GC-MS data coupled with resolution techniques. Various principle component analysis methods such as significant factor analysis and fixed size moving window evolving factor analysis were used for the number of factors, zero concentration and selective regions. Then, the convoluted chromatograms were resolved into pure chromatograms and mass spectra using heuristic evolving latent projections (HELP) method. Fatty acids of C16:1omega7, C18:4omega3, C18:1omega11, C18:1omega9, C18:0, C20:2omega6, C20:1omega9, C22:1omega11, C22:1omega9 and C24:1omega9 were resolved an fied by using similarity searches between deconvoluted mass spectra and MS database, in different parts of total ion current chromatogram. Window target testing factor analysis is also applied for confirming the presence or absence of target analytes. The results of the present work show that combination of hyphenated chromatographic methods and resolution techniques provide a complementary method for accurate analysis of real multi-component systems such as fish oil. 相似文献

We report on surface-engineered microarrays that provide in situ cell sorting, localization, and immobilization of various subsets of human primary lymphocytes, followed by an on-chip bioassay for ionizing-radiation-induced cytogenetic damage. The microarray format eliminates the necessity of separating cell sub-populations by alternative means (such as fluorescence- or magnetic-activated cell sorting) prior to performing informational bioassays. To exemplify the potential of this on-chip cytometry approach, we have integrated the cytokinesis-block micronucleus cytome (CBMNcyt) assay with the microarray platform for analysis of the chromosome damage profile of specific subsets of human peripheral lymphocytes. Microarray results were compared with data obtained from the traditional CBMNcyt assay on heterogeneous lymphocyte populations, and with flow cytometry data. Our results suggest that cytogenetic damage caused by ionizing radiation is not uniformly distributed across all lymphocytes subsets, but rather concentrated in specific subsets. The salient features of our approach are that it requires very small volumes of reagents, allows sorting of lymphocyte subsets in situ, increases parallelism of cell assays and is amenable to high content microscopy analysis. The on-chip cytometry format opens new vistas for advanced cell-based assays, potentially bringing to light important information which remains hidden with conventional assays and hence engendering new discoveries in cell biology. 相似文献

In this study, a three-phase hollow fiber liquid-phase microextraction (HF-LPME) method combined with liquid chromatography–mass spectrometry was developed for direct determination of four non-steroidal anti-inflammatory drugs (ketoprofen, naproxen, diclofenac and ibuprofen) in sewage sludge. The drugs were extracted from non-spiked and spiked slurry samples with different amounts of sludge into an organic phase and then back-extracted into an aqueous phase held in the lumen of the hollow fiber. High enrichment factors ranging from 2761 to 3254 in pure water were achieved. In sludge samples, repeatability and inter-day precision were tested with relative standard deviation values between 10–18% and 7–15%, respectively. Average concentrations of 29 ± 9, 138 ± 2, 39 ± 5 and 122 ± 7 ng/g were determined in dried sludge from Källby sewage treatment plant (Sweden) for ketoprofen, naproxen, diclofenac and ibuprofen, respectively. 相似文献

In this article, a new ligandless dispersive liquid-liquid microextraction method has been developed for preconcentration of trace quantities of silver as a prior step to its determination by flame atomic absorption spectrometry. In the proposed approach, carbon tetrachloride and ethanol were used as extraction and dispersive solvents. Several factors that may be affected on the extraction process, like, extraction solvent, disperser solvent, the volume of extraction and disperser solvent, pH of the aqueous solution and extraction time were optimized. Under the optimal conditions, the calibration curve was linear in the range of 5.0 ng mL−1 to 2.0 μg mL−1 of silver with R2 = 0.9995 (n = 9) and detection limit based on three times the standard deviation of the blank (3Sb) was 1.2 ng mL−1 in original solution. The relative standard deviation for eight replicate determination of 0.5 μg mL−1 silver was ±1.5%. The high efficiency of dispersive liquid-liquid microextraction to carry out the determination of silver in complex matrices was demonstrated. The proposed method has been applied for determination of trace amount of silver in standard and water samples with satisfactory results. 相似文献

In this study, cellulose acetate (CA) ultrafiltration (UF) membranes were prepared using the phase inversion method. Effects of CA and polyethylene glycol (PEG) concentrations in the casting solution and coagulation bath temperature (CBT) on morphology of the synthesized membranes were investigated. Based on L9 orthogonal array of Taguchi experimental design 18 membranes were synthesized (with two replications) and pure water permeation flux through them were measured. It was found out that increasing PEG concentration in the casting solution and CBT, accelerate diffusional exchange rate of solvent 1-methyl-2-pyrrolidone (NMP) and nonsolvent (water) and consequently facilitate formation of macrovoids in the membrane structure. Increasing CA concentration, however, slows down the demixing process. This prevents instantaneous growth of nucleuses in the membrane structure. Hence, a large number of small nucleuses are created and distributed throughout the polymer film and denser membranes are synthesized. Rate of water flux through the synthesized membranes is directly dependent on the size and number of macrovoids in the membrane structure. Thus, maximum value of flux is obtained at the highest levels of PEG concentration and CBT (10 wt.% and 23 °C, respectively) and the lowest level of CA concentration (13.5 wt.%). Analysis of variance (ANOVA) showed that all parameters have significant effects on the response. In this communication, new experimental data on the solubility of n-hexane, cyclo-hexane and iso-octane in pure water are reported. The data have been measured using a static-analytic technique that takes advantage of a Rolsi™ sampling device in the temperature range of 298–353 K and at pressures up to 0.5 MPa. The experimental data measured in this work at 298 K have been compared with some selected data from the literature and good agreement is found. A group contribution plus association equation of state, namely the GCA-EoS, is used to model the phase equilibrium of water + hydrocarbon (C2 to n-C6, cy-C6, i-C4 and i-C8) system.
