The extract from a laboratory culture of an Indonesian isolate of the cyanobacterium Phormidium sp. displayed inhibitory activity in a Ras-Raf protein interaction assay. Assay-guided fractionation led to the isolation of both active and inactive materials of novel structure. The major inactive metabolite, phormidolide, was nevertheless highly toxic to brine shrimp (LC(50) = 1.5 microM), and hence, its structure was elucidated using various spectroscopic methods, primarily NMR. A series of partial structures were developed from standard experiments and then assembled using GHMBC, 2D INADEQUATE, and ACCORD-ADEQUATE data obtained on a (13)C-enriched sample. The relative stereochemistry at phormidolide's 11 chiral centers was established using the J-based configuration analysis method in concert with the G-BIRD(R)-HSQMBC NMR experiment. Absolute stereochemistry was determined on a bis-acetonide derivative using the variable temperature Mosher ester method. The robust number of NMR restraints provided from determination of most homonuclear and heteronuclear coupling constants in phormidolide, along with an abundance of NOE information, allowed construction of a refined lowest energy three-dimensional structure in Macromodel. Phormidolide is one of only a few macrolide-type natural products to be reported from marine cyanobacteria.  相似文献

Replacing the 3- and 3′′-protons of the ligand 2,6-di(pyrazol-1-yl)pyridine L by mesityl groups changes the electronic ground state of [Cu(L)2]2+ complexes from {d}1 to {d}1. This is the best example so far for a “homoleptic” Jahn–Teller-compressed six-coordinate CuII complex.  相似文献

A rapid, sensitive and selective method is described for the determination of chromium(VI) in presence of 100-fold amounts of chromium(III) by flame atomic absorption spectrometry (FAAS) in conjunction with coprecipitative preconcentration of its ethyl xanthate complex onto naphthalene. The solid mixture consisting of the chromium(VI) complex together with naphthalene is dissolved in 8.0 ml of dimethyl formamide (DMF) and chromium(VI) content was established by FAAS. Calibration graphs were rectilinear over the chromium(VI) concentration in the range 0-200 μg l−1. Five replicate determinations of 20 μg of chromium(VI) present in 1.0 l of sample solution gave a relative standard deviation of 3.1%. The detection limit corresponding to three times the standard deviation of the blank was found to be 0.5 μg l−1. The developed procedure has been successfully utilized for the estimation of chromium(VI), chromium(total) (after oxidation with bromate) and chromium(III) (by subtracting chromium(VI) content from chromium(total) value contents of several tannery industries.  相似文献

The influence of the pH on the complexation equilibria between (S)- or (R)-alprenolol and the cellulase Cel7A was investigated by isothermal titration calorimetry. The results obtained agree with those of previous, similar studies of the same equilibria in which the protein was immobilized on silica particles, packed in a chromatographic column. The association constant and the complexation enthalpy and entropy of the (S)-enantiomer increase with increasing pH. For (R)-alprenolol, the binding is endothermic at all pH values. Thus, for both enantiomers in the pH range 5.5-6.8, the binding is an entropically driven process.  相似文献

This study investigated the speciation and bioavailability of selenium in yeast-based intervention agents from multiple manufacturers from several time points. Sources of selenized yeast included Nutrition 21 (San Diego, CA), which supplied the Nutritional Prevention of Cancer (NPC) Trial from 1981-1996; Cypress Systems (Fresno, CA; 1997-1999); and Pharma Nord (Vejle, Denmark; 1999-2000), which supplied the Prevention of Cancer by Intervention by Selenium (PRECISE) Trial pilot studies. The low-molecular-selenium species were liberated from the samples by proteolytic hydrolysis followed by separation by ion exchange liquid chromatography and detection by inductively coupled plasma-mass spectrometry. The results for the NPC tablets showed that selenomethionine, together with 3 unidentified selenium compounds, were predominant in the sample hydrolysates. The relative amounts of the 4 selenium species varied (p < 0.05) among several of the 7 tablet batches used during the course of the NPC Trial. In comparison, 5 batches of more recently produced selenized yeasts, which were used as a source of selenium in the PRECISE and other trials, contained less of the unknown compounds and more selenomethionine at 54-60% of the total selenium in the yeasts. One batch of yeast, however (from 1985), which originated from the same producer as the yeast used in the NPC tablets, contained only 27% of selenium in the sample as selenomethionine. Human subjects receiving 200 microg selenium/day in the UK PRECISE Pilot Trial showed a higher concentration (p < 0.01) and higher increase from baseline in plasma selenium than did the same dosage used in the NPC Trial. Differences in intake, speciation, or bioavailability of selenium from the yeast-based supplements in the population groups studied may explain this. Furthermore, the selenium concentration in whole blood from the Danish PRECISE Pilot Trial was higher (p < 0.001) than that obtained with synthetic L-selenomethionine in a comparable group of Danes, both groups having been treated with 300 microg selenium/day.  相似文献

The feasibility of constructing polymer/clay nanocomposites with polypeptides as the matrix material is shown. Cationic poly‐L‐lysine · HBr (PLL) was reinforced by sodium montmorillonite clay. The PLL/clay nanocomposites were made via the solution‐intercalation film‐casting technique. X‐ray diffraction and transmission electron microscopy data indicated that montmorillonite layers intercalated with PLL chains coexist with exfoliated layers over a wide range of relative PLL/clay compositions. Differential scanning calorimetry suggests that the presence of clay suppresses crystal formation in PLL relative to the neat polypeptide and slightly decreases the PLL melting temperature. Despite lower crystallinity, dynamic mechanical analysis revealed a significant increase in the storage modulus of PLL with an increase in clay loading producing storage modulus magnitudes on par with traditional engineering thermoplastics. © 2002 Wiley Periodicals, Inc. Bismuth and Sb were evaluated as internal standards (IS) to minimize matrix effects on the direct and simultaneous determination of As, Cu, and Pb in cachaça by graphite furnace atomic absorption spectrometry using W-coated platform plus Pd-Mg(NO3)2 as modifier. For 20 μL injected sample, calibration within the 0.5-10 μg L−1 As, 100-1000 μg L−1 Cu and 0.5-30 μg L−1 Pb intervals were established using the ratios As absorbance to Sb absorbance, Cu absorbance to Bi absorbance and Pb absorbance to Bi absorbance versus analytes concentration, respectively. Typical linear correlations of 0.998, 0.999 and 0.999 were, respectively, obtained. The proposed method was applied for direct determination of As, Cu and Pb in 10 commercial cachaça samples and results were in agreement with those obtained by inductively coupled plasma mass spectrometry at 95% confidence level. The found characteristic masses were 30 pg As, 274 pg Cu and 39 pg Pb. The useful lifetime of the graphite tube was around 760 firings. Recoveries of As, Cu and Pb added to cachaça samples varied, respectively, from 98% to 109%, 97% to 108% and 98% to 104% with internal standards and from 48% to 54%, 53% to 92% and 62% to 97% without internal standards. The limits of detection were 0.13 μg L−1 As, 22 μg L−1 Cu and 0.05 μg L−1 Pb. The relative standard deviations (n = 12) for a spiked sample containing 20 μg L−1 As, Pb and 500 μg L−1 Cu were 1.6%, 1.0%, and 1.8% with IS and 4.3%, 5.2%, and 5.5% without IS.  相似文献

The cycloaddition reaction of azomethine ylides, generated through decarboxylation, with (E)-3-arylidene-4-chromanones as dipolarophiles has been investigated. A high degree of regioselectivity has been observed in the synthesis of a new class of functionalized dispiroheterocyclic compounds bearing chromanone and acenaphthenequinone framework. The structures were established by spectroscopic techniques as well as single crystal X-ray analysis.  相似文献

The thermal conductivity and heat capacity of high-purity single crystals of yttrium titanate, Y2Ti2O7, have been determined over the temperature range 2 K?T?300 K. The experimental heat capacity is in very good agreement with an analysis based on three acoustic modes per unit cell (with the Debye characteristic temperature, θD, of ca. 970 K) and an assignment of the remaining 63 optic modes, as well as a correction for CpCv. From the integrated heat capacity data, the enthalpy and entropy relative to absolute zero, are, respectively, H(T=298.15 K)−H0=34.69 kJ mol−1 and S(T=298.15 K)−S0=211.2 J K−1 mol−1. The thermal conductivity shows a peak at ca. θD/50, characteristic of a highly purified crystal in which the phonon mean free path is about 10 μm in the defect/boundary low-temperature limit. The room-temperature thermal conductivity of Y2Ti2O7 is 2.8 W m−1 K−1, close to the calculated theoretical thermal conductivity, κmin, for fully coupled phonons at high temperatures.  相似文献

IR, Raman and surface-enhanced Raman scattering (SERS) spectra of L-proline were recorded and analyzed. The molecular plane assumes a tilted orientation with respect to the metal surface.
