Hyperphosphorylation of Tau, a protein that stabilizes microtubules, leads to the breakdown of the microtubular structure and ultimately to the formation of neurofibrillar tangles within neurons. Here, we report monitoring of Tau phosphorylations electrochemically, using Tau protein films chemically linked to gold surfaces and 5'-γ-ferrocenyl (Fc) adenosine triphosphate (Fc-ATP) as a co-substrate. Fc-phosphorylation reactions of Tau are explored using the three protein kinases, glycogen synthase kinase (GSK-3β), sarcoma (Src)-related kinase, and protein kinase A (PKA), which catalyze Fc-phosphorylation of different residues and regions within Tau. The kinetic parameters of the biochemical process (K(M) and V(max)) were determined.  相似文献

The Breath‐Figure technique was employed to imprint honeycomb structures in the polymer films via the condensation of water vapor on the surface of an evaporating polymer solution. Generally, the condensed water droplets can be stabilized by an end‐functional polymer or by particles added to the polymer solution. In this study, we carried out a systematic experiment on the effect of different stabilizers on the porous honeycomb structure under identical physical conditions. The end‐functional polymer produced a large area of regular spherical bubbles, whereas adding particles to the polymer solution leads to smaller arrays of the flattened bottom bubbles. The separation length between pores was larger for polymer/particle sample than that of the end‐functional polymer films. The vibrational properties of mesoporous silica (SBA-15) were investigated by deep ultraviolet (UV) Raman and infrared spectroscopies with and without the presence of platinum nanoparticles in the mesopores that were incorporated by sonication. Raman and IR spectral line assignments were made by comparison to amorphous silicas. This procedure permitted identification of vibrations of longitudinal (LO) and transverse (TO) optical lattice modes, the presence of Si-OH, and vibrational modes associated with the presence of three-, four-, and six-membered siloxane rings. Hydraulic pressing of the mesoporous silica with pressure in the range 3-7 tons cm(-2) destroys the X-ray diffraction pattern and strongly decreases the Raman peak (D2) associated with three-membered rings at the surface. In the presence of platinum nanoparticles in the silica mesopores, a peak attributed to a Pt-O stretching vibration appears at between 530 and 580 cm(-1) in the UV-Raman spectrum, which can be used to monitor the presence of the platinum particles and their interaction with the support. The D2 feature in the UV-Raman spectra also decreases with increasing Pt loading, which is attributed to interactions of the Pt nanoparticles with the silica surface.  相似文献

The structure of the polymeric substrate plays an important role in the nucleation of calcium carbonate crystals. In this study a synthetic water-soluble poly(acrylamide-co-2-acrylamido-2-methyl-1-propane sodium sufonate-co-n-vinyl pyrrolidone) was found to be a substrate favoring the nucleation of polymorphs of calcium carbonate crystals under specific experimental conditions. Morphological characterization of the polymorphs was done using atomic force microscopy, scanning electron microscopy, energy dispersive spectroscopy, FTIR analysis, and X-ray diffraction. If calcium carbonate is precipitated in the presence of terpolymer, a remarkable increase in nucleation density (number of crystals per unit area) was observed. Stacked crystals of rhombohedral morphology that formed may be due to the presence of sodium sulfonate groups on the terpolymer. However, in the presence of poly-L-aspartic acid, almost all crystals are hollow and have needlelike or plate like morphology was formed. This change in calcium carbonate morphology can be explained by the variation of the polymer conformation, if poly- L-aspartic acid is present.  相似文献

In an updated version of the polymerization‐filling technique, the surface of particulate fillers has been activated by metallocenes. Homogeneously filled polyolefins have accordingly been produced by the in situ copolymerization of ethylene and α‐olefins, e. g., 1‐octene and 1,9‐decadiene. Different fillers have been successfully used, including inorganic particles with either an acidic surface (kaolin, glass beads, silica) or a basic surface (magnesium hydroxide, wollastonite), graphite and nickel particles. Hydrogen has proved to be efficient in reducing the molecular weight of the produced polymers.  相似文献

In optimal control of robots, engineering practice is to calculate off‐line an optimal open‐loop control, based on nominal values of the model parameters. Due to the deviation between the nominal model parameters and the possible realisations of the model parameters, the performance of the system usually does not correspond to the chosen open‐loop control. Hence, expensive on‐line control corrections are necessary to improve system performance. By incorporating available a priori and statistical information about the unknown model parameters into the control process, these expenses are reduced to a large extent.  相似文献
