Previous studies of the fractionated venom of the Brazilian armed spider Phoneutria nigriventer, obtained by gel filtration, have demonstrated the presence of a fraction PhM, a pool of small peptides (up to 2000 Da) that provoke contractions in smooth muscle of guinea pig ileum. Initial attempts to sequence these peptides were largely unsuccessful because of the low purification yield and the fact that the majority seemed to be blocked at their N-termini. In the present work, analysis of this venom fraction by mass spectrometry has revealed the existence of a highly complex mixture of peptides with molecular weights corresponding to those observed for the muscle-active peptides previously described (800-1800 Da). These peptides appear to be a family of isoforms with some particular features. The amino acid sequences of 15 isoforms have been determined by tandem mass spectrometry (MS/MS) using both electrospray ionization quadrupole time-of-flight mass spectrometry (ESI-Q/ToFMS) and matrix-assisted laser desorption/ionization tandem time-of-flight mass spectrometry (MALDI-ToF/ToFMS). These molecules contain post-translational modifications such as proteolysis and C-terminal amidation, which combine to generate additional isoforms. All the isoforms sequenced in this study possess an N-terminal pyroglutamic acid residue. A search for sequence similarities with other peptides in databanks revealed that these peptides are structurally related to the tachykinins, a family of neuro-hormone peptides. The data obtained in this study will be essential for the subsequent steps of this research, the synthesis of these peptides and pharmacological characterization of their biological activity.  相似文献

Morphological and structural variations of particles of Bioglass with two different grain sizes reacted in Tris-buffered solution were analyzed by means of N(2) adsorption/desorption at 77 K and HR-TEM/EDS. A remarkable increase in specific surface area (ssa) was observed after the first hour of dissolution. A plateau value corresponding to an increase of at least 2 orders of magnitude was reached after 2 days of dissolution. The ssa increase was faster for the smaller particle size sample, and the ratio between the ssa of the starting samples was not maintained during dissolution. Both micro- and mesopores were formed at different stages of the reaction for the two samples. Increasing ssa was also connected to the formation of a microcrystalline structure rich in Ca and P, as shown by TEM images. The segregation of both a SiO(2)-rich amorphous phase and a Ca/P-rich crystalline phase was observed after the first hour of dissolution. After 2 days of reaction, Ca/P-rich particles made of fine aciculate crystals were found either in close contact with SiO(2) particles or deposited on a small SiO(2)-rich core. A preliminary analysis of TEM data showed the formation, together with hydroxy carbonate apatite, of different types of calcium phosphates not detectable by powder X-ray diffraction.  相似文献

Methanol decomposition and oxidation on Pd(111) at millibar pressure were studied by in situ polarization-modulation infrared reflection absorption spectroscopy (PM-IRAS), on-line gas chromatography and pre- and postreaction X-ray photoelectron spectroscopy (XPS). Various dehydrogenation products such as methoxy CH3O, formaldehyde CH2O, formyl CHO, and CO could be spectroscopically identified. Methanol oxidation proceeds via dehydrogenation to formaldehyde CH2O, which either desorbs or is further dehydrogenated to CO, which is subsequently oxidized to CO2. Carbonaceous overlayers that are present during the reaction may favorably affect the selectivity toward CH2O. The reaction takes place on metallic Pd, and no indications of an involvement of Pd surface oxide were observed.  相似文献

We present a novel method of statistical analysis for the comparison of electrophoretic data. The method is based on the squared Euclidian distance of normalized signal data vectors of electrophoretic lanes. The differences in the electrophoretic patterns are evaluated by a statistical test based on Hubert's statistics which measures the significance of the signal grouping. We demonstrate the validity and applicability of the method in a large data set derived from automated fluorescent mRNA differential display analysis of the expression of acute-phase proteins during experimental Escherichia coli infection in mice. The current testing method is capable of finding theoretically similar natural groupings to be similar in a statistically significant way whereas theoretically dissimilar or random groupings can be recognized to be artifactual. We also show how the calculated pairwise signal distances can be utilized in methodological problem solving. These analytical methods can be applied to the study of other related problems of similarity analysis of electrophoretic patterns, and also provide useful tools for the development of automated recognition of differentially expressed mRNAs.  相似文献

The inhibition of the model enzyme, haloalkane dehalogenase from Sphingomonas paucimobilis, was investigated by a combination of electrophoretically mediated microanalysis with a partial filling technique, followed by indirect or direct detection. In this setup, part of the capillary is filled with a buffer suitable for the enzymatic reaction (20 mM glycine buffer, pH 8.6) whereas the rest of the capillary is filled with the background electrolyte optimal for separation of substrates and products. Two different background electrolytes and corresponding detection approaches were used to show the versatility of the developed method. The inhibition effect of 1,2-dichloroethane on the dehalogenation of brominated substrate 1-bromobutane was studied by means of 10 mM chromate - 0.1 mM cetyltrimethylammonium bromide (pH 9.2) in combination with indirect detection or 20 mM beta-alanine - hydrochloric acid (pH 3.5) in combination with direct detection. The method was used to estimate the inhibition constant K(I) (0.44 mM by indirect detection and 0.63 mM by of direct detection) and to determine the inhibition type. Compared to spectrophotometric and other discontinuous assays, the method is rapid, can be automated, and requires only small amount of reagents that is especially important in the case of enzymes and inhibitors.  相似文献

The application of the combination of experimental design (ED) and artificial neural networks (ANNs) for the quantification of overlapped peaks in capillary zone electrophoresis is described. When the total separation cannot be achieved by separation techniques, the use of ED-ANN can be a suitable approach. The unstability of EOF causes peak shift that has to be corrected in order to apply ED-ANN methods. In this work, normalization procedure of electropherograms with consequent application of ANNs for quantification purpose was developed. Both, spectra and electropherograms can be used as multivariate data. In general, both kinds of data were found to be suitable for unresolved peaks quantification by ED-ANN approach.  相似文献

An on-line preconcentration procedure for the determination of manganese using flow-injection approach with flame atomic absorption spectrometry as a detection method is described. The proposed method is based on the complexation between Mn(II) and 5,10,15,20-tetrakis(4-carboxyphenyl)porphyrin (TCPP). Two approaches were investigated for enrichment of manganese; the formation of Mn-TCPP complex in a solution followed by its retention on a sorbent and the sorption of manganese ions onto the TCPP-modified resin. The best results was obtained for the first approach when 10−5 M reagent was on-line mixed with an aqueous sample solution and passed through the microcolumn packed with anion-exchange resin Amberlite IRA-904 for 5 min. The sorbed complexes were then eluted with 0.5 ml of 2 M HNO3. A good precision (2.2-3.1% R.S.D. for 50 μg l−1 manganese) and the enrichment factor of 30 were obtained with the detection limit of 12 μg l−1 for 5 min loading time. The interference of anions and cations has been studied to optimize the conditions and the method was applied for determination of manganese in natural water samples. The results obtained by FI-FAAS and ETAAS (as a reference method) were not statistically different for a significance level of 0.05.  相似文献

We developed a robust nanosecond photonic crystal switching material by using poly(N-isopropylacrylamide) (PNIPAM) nanogel colloidal particles that self-assemble into crystalline colloidal arrays (CCAs). The CCA was polymerized into a loose-knit hydrogel which permits the individual embedded nanogel PNIPAM particles to coherently and synchronously undergo their thermally induced volume phase transitions. A laser T-jump from 30 to 35 degrees C actuates the nanogel particle shrinkage; the resulting increased diffraction decreases light transmission within 900 ns. Additional transmission decreases occur with characteristic times of 19 and 130 ns. Individual NIPAM sphere volume switching occurs in the approximately 100 ns time regime. These nanogel nanosecond phenomena may be useful in the design of fast photonic crystal switches and optical limiting materials. Smaller nanogels will show even faster volume phase transitions.  相似文献

Degradation of poly(ester-urethanes), poly(ether-urethanes) and poly(acrylic-urethanes), as a base for automotive paintings in interior applications, has been studied by DSC. The samples were clearcoat and black-pigmented paints, unstabilized and stabilized with HALS Tinuvin 292, UV absorber Tinuvin 1130 and antioxidant Hostanox O3, exposed to weathering in Xenotest and in Arizona desert. From the dependences of oxidation onset temperature on the heating rate, the kinetic parameters enabling to calculate the oxidation induction time for a chosen temperature have been obtained. From the values of oxidation induction time, the protection factors of the additives and the residual stability of the polymer after an ageing stress has been evaluated. It has been shown that the equivalence between the two methods of weathering depends on the polymer composition. A new criterion for the evaluation of synergism/antagonism of additives in the stabilizing mixture has been proposed.  相似文献
