Prion diseases are incurable neurodegenerative diseases that affect both humans and animals. The infectious agent is a pathogenic
form of the prion protein that accumulates in brain as amyloids. Currently, there is neither cure nor reliable preclinical
diagnostics on the market available. The growing number of reports shows that passive immunisation is one of the most promising
strategies for prion disease therapy, where antibodies against prions may prevent and even cure the infection. Since antibodies
are large molecules and, thus, might not be suitable for the therapy, different antibody fragments are a good alternative.
Therefore, we have designed and prepared single-chain antibody fragments (scFvs) derived from the PrPSc-specific murine monoclonal antibody V5B2. Using a new expression vector pMD204, we produced scFvs in two opposing chain orientations
in the periplasm of Escherichia coli. Both recombinant antibody fragments retained the specificity of the parent antibody and one of these exhibited binding properties
comparable to the corresponding murine Fab fragments with the affinity in nM range. Our monovalent antibody fragments are
of special interest in view of possible therapeutic reagents for prion diseases as well as for development of a new generation

Partial least squares regression (PLS) calibration models based on Fourier transform infrared (FTIR-ATR) and Raman spectra (FT-Raman) were applied to the rapid and accurate simultaneous determination of the main properties of diesel fuels. Training sets were composed of over ninety commercial diesel fuel samples. The methods use baseline-uncorrected, raw FTIR-ATR and FT-Raman spectra. Two spectral regions were studied: full spectral region and “fingerprint” region. The models were validated using the cross-validation process. Based on the correlation coefficient and root mean square error of cross validation (RMSECV) values the both developed calibration models, PLS/FTIR-ATR and PLS/FT-Raman, were very accurate and comparable with standard testing methods. The following diesel fuel properties may be confidently estimated: cetane number, cetane index, density, viscosity, distillation temperatures at 10% (T10), 50% (T50) and 90% (T90) recovery, as well as the contents of total aromatics and polycyclic aromatic hydrocarbons. As compared to the “fingerprint” spectral region, the PLS/FTIR-ATR model using full spectral region displayed slightly better performances with the most of the correlation coefficient values above 0.98. 相似文献

The effects of solvent and temperature on the complexation of adamantyl mannoside with β-cyclodextrin and 6-O-monotosyl-6-deoxy-β-cyclodextrin were explored experimentally and by means of molecular dynamics simulations. Efficient binding was observed only in hydrogen-bonded solvents, which indicated solvophobically driven complexation. The stability of the inclusion complex was considerably higher in aqueous media. A pronounced temperature dependence of ΔrH○ and ΔrS○, resulting in perfect enthalpy–entropy compensation, was observed in water. The complexation thermodynamics was in line with classical rationale for the hydrophobic effect at lower temperatures and the nonclassical explanation at higher temperatures. This finding linked cyclodextrin complexation thermodynamics with insights regarding the effect of temperature on the hydration water structure. The complexation enthalpies and entropies were weakly dependent on temperature in organic media. The signs of ΔrH○ and ΔrS○ were in accordance with the nonclassical hydrophobic (solvophobic) effect. The structures of the optimized product corresponded to those deduced spectroscopically, and the calculated and experimentally obtained values of ΔrG○ were in very good agreement. This investigation clearly demonstrated that solvophobically driven formation of cyclodextrin complexes could be anticipated in structured solvents in general. However, unlike in water, adamantane and the host cavity behaved solely as structure breakers in the organic media explored so far. 相似文献

A study on the synthesis and mechanistical aspects of formation of 3-methyl-5-oxo-3-pyrazolin-1-carboxamide (MOPC) starting from S-methylisothiosemicarbazide hydrogen iodide and methyl acetoacetate was performed. In the alkaline aqueous solution, the intermediate methyl acetoacetate S-methylisothiosemicarbazone undergoes substitution of CH3S? anion by hydroxide anion, cyclization, carbanion formation, and elimination of methanol, thus yielding corresponding Na-enolate salt of pyrazol-5-one derivative. The structure of the compound obtained after protonation of the formed enolate salt was determined by means of spectroscopic techniques and single-crystal X-ray diffraction analysis. The mechanism of conversion of methyl acetoacetate S-methylisothiosemicarbazone into MOPC was investigated by means of the B3LYP functional, and it was found that the reaction is thermodynamically controlled. 相似文献

High-temperature treatment steps in fabrication process of dye sensitized solar cell (DSSC) significantly contribute to the manufacturing costs and limit the use of temperature sensitive substrates. Therefore our aim was to develop a simple method for the preparation of water-based TiO2 paste. The paste is based on peroxotitanic acid (PTA) sol–gel matrix and commercial TiO2 nanoparticles (P25). Two fabrication processes to decompose/transform the PTA matrix in the printed TiO2 layer are explored and combined: annealing at temperatures up to 250 °C and/or oxygen plasma treatment. The results show that the PTA matrix in the paste converts to anatase phase and to some extent also attaches to the TiO2 nanoparticles P25 acting as an interconnecting network within TiO2 layer. The transformation of the PTA matrix occurs around 250 °C, but in the presence of TiO2 nanoparticles P25 it starts already at 120 °C. In addition the results reveal that the crystallization is achievable also solely with the oxygen plasma treatment. The efficiency of the TiO2 layers, exposed to different post-deposition treatments, is evaluated in DSSCs. The results show that oxygen plasma treatment of the TiO2 layers could efficiently replace temperature curing at 250 °C. Within this study the DSSCs with the efficiency up to 4.2 % measured under standard test conditions (1,000 W/m2, AM1.5, 25 °C) were realized. 相似文献

Waxes and wax emulsions are one of the most important solutions for non-biocidal wood protection. Wax treated wood is designed for outdoor use, therefore it is of considerable importance to elucidate the influence of weathering on the photodegradation processes. It is presumed, that wax treatment will reduce water uptake and thus reduce or slow down photodegradation processes. In order to test this hypothesis, three types of wax emulsions at two different concentrations were vacuum impregnated into Norway spruce wood specimens: an emulsion of montan wax (LGE), an emulsion of polyethylene (WE1) and an emulsion of oxidized polyethylene (WE6) wax. The samples were exposed to artificial accelerated weathering (AAW) for 500 cycles. Before and after AAW colour, the contact angle of water and moisture content were determined. Chemical and morphological changes at exposed surfaces were also investigated with FTIR spectroscopy and SEM. In parallel, the uptake of wax emulsions and water into the axial surfaces of samples was determined with a tensiometer. The moisture content measurements showed that the best hydrophobic effect was achieved with LGE treated wood, where the lowest colour and FTIR changes were observed as well. These results were supported by tensiometer measurements as well. Presented data clearly showed that high loadings of waxes reduce or at least slow down weathering. Among tested waxes, montan wax was found the most effective. 相似文献

The first single‐diamond cubic phase in a liquid crystal is reported. This skeletal structure with the space group is formed by self‐assembly of bolaamphiphiles with swallow‐tailed lateral chains. It consists of bundles of π‐conjugated p‐terphenyl rods fused into an infinite network by hydrogen‐bonded spheres at tetrahedral four‐way junctions. We also present a quantitative model relating molecular architecture to the space‐filling requirements of six possible bicontinuous cubic phases, that is, the single‐ and double‐network versions of gyroid, diamond, and “plumber′s nightmare”. 相似文献

In airway myocytes, like in many cells, Ca(2+) signaling is controlled by inositol 1,4,5-trisphosphate (InsP(3)) via InsP(3) receptors (InsP(3)R) located in the sarco-endoplasmic reticulum. Three types of InsP(3)R exist, labeled Types 1, 2, and 3, which differ in their gating kinetics. We analyze a possible impact of the different gating kinetics of Type 1 and Type 3 InsP(3)R on the time course of cytosolic Ca(2+) concentration in tracheal smooth muscle cells upon agonist stimulation. Previous experimental data in rat tracheal myocytes showed that upon gradually increased stimulation with acetylcholine (ACh), a contractile agonist that acts via InsP(3) production, signal spikes, several spikes with declining maxima, and sustained oscillations appear. Our model reproduces the time courses of cytosolic Ca(2+) measured in tracheal myocytes. Moreover, by postulating slight variations in the model parameters which determine the total number of receptors expressed and the ratio between Type 1 and Type 3 InsP(3)R, it offers an explanation to the experimental observation of qualitatively different responses of cells within a presumably homogeneous tissue. 相似文献

We report three self‐assembled iron complexes that comprised an anti‐parallel open form (o‐ L anti), a parallel open form (o‐ L syn), and a closed form (c‐ L ) of diarylethene conformers. Under kinetic control, FeII2(o‐ L anti)3 was isolated, which exhibited a dinuclear structure with diamagnetic properties. Under light‐irradiation control, FeII2(c‐ L )3 was prepared and exhibited paramagnetism and spin‐crossover behaviour. Under thermodynamic control and in the presence of indispensable [FeIII(Tp*)(CN)3]?, FeII2(o‐ L anti)3 and FeII2(c‐ L )3 transformed into tetranuclear FeIII2FeII2(o‐ L syn)2, which exhibited complete spin‐crossover behaviour at T1/2=353 K. 相似文献
