Two selected carboranethiol isomers were used to modify flat silver surfaces. Both isomers, 1,2-(HS)(2)-1,2-C(2)B(10)H(10) (a) and 9,12-(HS)(2)-1,2-C(2)B(10)H(10) (b), are relatively strong dipoles with two SH groups per molecule. They are both anchored to the surface via two SH groups per molecule. Topography and surface potential changes of the modified silver surfaces were studied using Scanning Kelvin Probe Force Microscopy (SKPFM). These measurements proved that both isomers are oppositely oriented on the surface. The former isomer increases, and the latter one decreases the surface potential of a modified silver film. The relative changes of the surface potential correlate well with the dipole moments of the isomers. Competitive chemisorption from a 1:1 mixture of both isomers shows that the isomer (a) is found in a significantly higher concentration on the surface than the isomer (b). This has been proved by both SKPFM and X-ray photoelectron spectroscopy (XPS) techniques. Additionally, contact angle measurements were carried out to characterise the modified surfaces, and these and XPS results show the presence of hydrophobic hydrocarbon contaminants.  相似文献

Heparan sulfate (HS) is a highly sulfated glycosaminoglycan with a variety of critical functions in cell signaling and regulation. HS oligosaccharides can mimic or interfere with HS functions in biological systems; however, their exploitation has been hindered by the complexity of their synthesis. Polyvalent displays of small specific HS structures on dendritic cores offer more accessible constructs with potential advantages as therapeutics, but the synthesis of single‐entity HS polyvalent compounds has not previously been described. Herein we report the synthesis of a novel targeted library of single‐entity glycomimetic clusters capped with varied HS saccharides. They have the ability to mimic longer natural HS saccharides in their inhibition of the Alzheimer’s disease (AD) protease BACE‐1. We have identified several single‐entity HS clusters with IC50 values in the low‐nanomolar range. These HS clusters are drug leads for AD and offer a novel framework for the manipulation of heparan sulfate–protein interactions in general.  相似文献

Adsorption of DNA molecules on mica, a highly negatively charged surface, mediated by divalent or trivalent cations is considered. By analyzing atomic force microscope (AFM) images of DNA molecules adsorbed on mica, phase diagrams of DNA molecules interacting with a mica surface are established in terms of concentrations of monovalent salt (NaCl) and divalent (MgCl2) or multivalent (spermidine, cobalt hexamine) salts. These diagrams show two transitions between nonadsorption and adsorption. The first one arises when the concentration of multivalent counterions is larger than a limit value, which is not sensitive to the monovalent salt concentration. The second transition is due to the binding competition between monovalent and multivalent counterions. In addition, we develop a model of polyelectrolyte adsorption on like-charged surfaces with multivalent counterions. This model shows that the correlations of the multivalent counterions at the interface between DNA and mica play a critical role. Furthermore, it appears that DNA adsorption takes place when the energy gain in counterion correlations overcomes an energy barrier. This barrier is induced by the entropy loss in confining DNA in a thin adsorbed layer, the entropy loss in the interpenetration of the clouds of mica and DNA counterions, and the electrostatic repulsion between DNA and mica. The analysis of the experimental results provides an estimation of this energy barrier. We then discuss some important issues, including DNA adsorption under physiological conditions.  相似文献

Optical fibers and molded optics have been formed by using the Ga5Sb10Ge25Se60 base glass with appropriate thermal treatment to generate microcrystals. The crystal growth during the fiber drawing and molding process induces additional scattering losses in the short wavelength region, but does not have a significant effect beyond 6 μm. The molded optics have very good dimensional precision and the form defect is less than 0.5 μm peak-to-valley, which is excellent particularly for optics designed for operating at 8-12 μm.  相似文献

This paper discusses the use of aperiodic (binary or ternary) sequences in combining pseudorandom number generators (RNG). We introduce a method for combining two or three RNGs using cut and project sequences. This combination method produces aperiodic number sequences having no lattice structure. We compare two surface treatments of biaxially-oriented polypropylene (BOPP), which are carried out in the same dielectric barrier discharge (DBD) apparatus, namely air corona, and N2 atmospheric pressure glow discharge (APGD). Changes in the surface energy and chemistry are investigated by contact angle measurements, by X-ray photoelectron spectroscopy (XPS) and by attenuated total reflectance infrared spectroscopy (ATR-FTIR). It is shown that N2 APGD treatment leads to a higher surface energy than air corona treatment, and to the formation of mostly amine, amide, and hydroxyl functional groups at the polypropylene surface. Finally, hydrophobic recovery of the treated film is studied; for both treatment types, the increased surface energy is found to decay in a similar manner with increasing storage time after treatment.  相似文献

Photodynamic-induced cytotoxicity by hypericin (HYP) was studied on three human melanoma cell lines: one pigmented cell line (G361) and two amelanotic cell lines (M18 and M6). No significant variation in the rate of uptake and in the maximum level of HYP incorporation for the different cells was observed. In the dark, no cytotoxicity was observed in the range0–10?6M HYP for the three cell lines. Amelanotic cells were found to be more sensitive than pigmented cells to irradiation of HYP with visible light (Λ > 590 nm). In addition, for the three cell lines HYP-induced photocytotoxicity was found to be drug-dose and light-dose dependent. Under the conditions used, thiobarbituric acid-reacting substances (TBARs) were significantly increased in amelanotic cells after irradiation (P < 0.0001). By contrast, the amount of TBARS remained unchanged in pigmented cells. Antioxidant defenses including enzymes and glutathione (GSH) were assayed before and after HYP photosensitization. Significantly increased total SOD activity was observed after photosensitization for amelanotic cells (P < 0.05), while glutathione peroxidase (GSHPx) and catalase (Cat) activities but also GSH levels were significantly decreased (P < 0.01). In pigmented cells a significantly increased Cat activity was found (P < 0.05), whereas GSHPx was unaffected after irradiation. It can be inferred that (a) HYP may be an effective PDT agent for melanoma and (b) there is a relationship between melanin content and sensitivity to HYP phototoxicity in human melanoma cells.  相似文献
