De novo site-specific 13C and 15N backbone and sidechain resonance assignments are presented for uniformly enriched E. coli thioredoxin, established using two-dimensional homo- and heteronuclear solid-state magic angle spinning NMR correlation spectroscopy. Backbone dihedral angles and secondary structure were derived from the statistical analysis of the secondary chemical shifts, and are in good agreement with solution values for the intact full-length thioredoxin, with the exception of a small number of residues located at the termini of the individual secondary structure elements. A large number of cross-peaks observed in the DARR spectra with long mixing times correspond to the pairs of carbon atoms separated by 4-6 angstroms, suggesting that DARR could be efficiently employed for observation of medium- and long-range correlations. The 108 amino acid residue E. coli thioredoxin is the largest uniformly enriched protein assigned to this degree of completeness by solid-state NMR spectroscopy to date. It is anticipated that with a combination of two-dimensional correlation experiments and high magnetic fields, resonance assignments and secondary structure can be generally derived for other noncrystalline proteins.  相似文献

A polymer bearing hydroxamic acid groups and having a high affinity for iron(III) was prepared through the following procedure. Acryloylalanine (III), prepared by the reaction of acryloyl chloride with alanine, was treated with N-hydroxysuccinimide in the presence of dicyclohexylcarbodiimide to give the N-hydroxysuccinimide ester (IV). The ester IV was polymerized by using AIBN in dioxane to give polymer V. Treatment of polymer V with methylhydroxylamine in DMF gave the hydroxamic acid polymer II. The water-soluble polymer II was purified by dialysis or by gel-permeation chromatography (GPC) on Sephadex G-25. Analytical GPC on Sephadex G-200 and Sepharose 4B indicate that the average molecular weight of the polymer is in the range of 5 × 105 to 1 × 106. The presence of hydroxamic acid groups is confirmed by the intense red-brown color produced by the addition of iron(III) to a 50% aqueous DMF solution of the polymer under acidic conditions. In pure water the polymer-iron complex precipitates as a tan solid. Iron-binding studies of the polymer reveal that the iron(III) trihydroxamic acid complex FeA3 forms at low concentrations of iron. At higher iron levels a lower order of stability is apparent, which can be accounted for by the conversion of FeA3 to FeA2+. In contrast, the FeA3 complex of the trihydroxamic acid deferoxamine-B is stable at all iron levels. These results are consistent with the polymer structure, which for steric reasons would favor a stable complex, FeA2+, between iron and two adjacent hydroxamic acid groups. An FeA3 complex would be expected to have a lower stability as a result of either bond angle strain and atomic compression, or a lower probability in bringing a third hydroxamic acid into position to form the octahedral complex.  相似文献

 We used a battery of different methods to study the association in aqueous sodium dehydrocholate (NaDHC) solutions. This salt associates by a stepwise mechanism. Below (9.6 ± 4.2) × 10−4 mol dm−3 there is a molecular solution with some strongly insoluble dehydrocholic acid produced by hydrolysis. Between (9.6 ± 4.2) × 10−4 and (5.2 ± 2.2) × 10−3 mol dm−3, an aggregate similar to acid soap (NaDHC.HDHC) appears and its amount and the aggregate's size increase with concentration. At =(2.20 ± 0.85) × 10−2 mol dm−3 the aggregates formed have properties usually associated with true micelles, such as solubilisation of water-insoluble dyes. These aggregates increase in size with concentration and change their shape at 8 × 10−2 mol dm−3, giving nonsymmetrical aggregates. The changes in the solution physicochemical properties at these concentrations may be misinterpreted and this explains the different values of the critical micelle concentration reported in the literature for substances with similar structure, such as bile salts. In this work, the coupling between flow analysis (FA)–vapor phase generation (VPG) and Fourier transform infrared spectrometry (FTIR) has been proposed as a novel and alternative strategy for the determination of nitrite. The analyte was transformed into the gaseous nitric oxide (NO) by on-line reaction with potassium iodide (KI) or ascorbic acid in acidic medium. The gaseous NO generated was transported by means of a N2 gas carrier stream inside the IR gas cell and the corresponding FTIR spectrum was acquired in a continuous mode. The absorbance at 1876 cm−1, corrected by a baseline established between 1879 and 1872 cm−1 at a nominal resolution of 2 cm−1, was selected as a measurement criterion. The effect of different spectroscopic and flow analysis experimental parameters, such as nominal resolution, number of scans, reducing agent and its concentration, acidic medium, reagents and sample flow rates, and the carrier gas flow rate on the analytical signal, and then in the figures of merit were initially evaluated by using a standard short path length (10 cm) IR gas cell. The optimization of the system was carried out by the univariate method. The main aims of this study were: (i) to investigate the on-line generation of gaseous nitric oxide in a continuous flow system, and (ii) the use of Fourier transform infrared spectrometry as an alternative and selective detector for the determination of nitrite. The proposed method was initially tested and applied for the determination of nitrite in samples with very high concentration of nitrite, such as frankfurters.  相似文献

A column-switching high-performance liquid chromatographic method with UV detection for the determination of cocaine (COC) and benzoylecgonine (BZE) in human blood plasma samples is described. The method uses an alkyl-diol-silica ADS-C18 extraction precolumn. A 50- micro L plasma sample was introduced to the ADS precolumn in order to separate the analytes from proteins and endogenous compounds. The fraction containing COC and BZE was back-flushed and transferred to an Alltech mixed-mode C(18)/cation-exchange analytical column for final separation. The validation of the method revealed quantitative recoveries from 95.0 to 99.0% for COC at three different concentrations (0.5, 1.0 and 2.0 micro g mL(-1)), and from 96.0 to 99.0% for BZE at the same concentration levels with coefficients of variation <4.00% (n=5). The detection limit (signal to noise ratio (S/N)>3) was 0.03 micro g mL(-1) for all the compounds with an injection volume of 50 micro L. However, it was possible to enhance the sensitivity further by injecting larger plasma volumes, up to 200 micro L, at the same optimal conditions. The overlap of sample preparation, analysis and reconditioning of the extraction column, increase the overall sample throughput to 5 samples h(-1). The developed method has been applied to human blood plasma samples from subjects suspected of cocaine abuse.  相似文献

A new solid solution TlFe0.22Al0.78As2O7 has been synthesized by a solid-state reaction. The structure of the title compound has been determined from a single-crystal X-ray diffraction and refined to final values of the reliability factors: R(F2)=0.030 and wR(F2)=0.081 for 1343 independent reflections with I>2σ(I). It crystallizes in the triclinic space group P-1, with a=6.296(2) Å, b=6.397(2) Å, c=8.242(2) Å, α=96.74(2)°, β=103.78(2)°, γ=102.99(3)°, V=309.0(2) Å3 and Z=2. The structure can be described as a three-dimensional framework containing (Fe/Al)O6 octahedra connected through As2O7 groups. The metallic units and diarsenate groups share oxygen corners to form a three-dimensional framework with interconnected tunnels parallel to the a, b and c directions, where Tl+ cations are located. The ionic conductivity measurements are performed on pellets of the polycrystalline powder. At 683 K, The conductivity value is 5.23×10−6 S cm−1 and the ionic jump activation energy is 0.656 eV. The bond valence analysis reveals that the ionic conductivity is ensured by Tl+ along the [001] direction.  相似文献

Bismuth as BiCl4 and BH4 ware successively retained in a column (150 mm × 4 mm, length × i.d.) packed with Amberlite IRA-410 (strong anion-exchange resin). This was followed by passage of an injected slug of hydrochloric acid resulting in bismuthine generation (BiH3). BiH3 was stripped from the eluent solution by the addition of a nitrogen flow and the bulk phases were separated in a gas–liquid separator. Finally, bismutine was atomized in a quartz tube for the subsequent detection of bismuth by atomic absorption spectrometry. Different halide complexes of bismuth (namely, BiBr4, BiI4 and BiCl4) were tested for its pre-concentration, being the chloride complexes which produced the best results. Therefore, a concentration of 0.3 mol l−1 of HCl was added to the samples and calibration solutions. A linear response was obtained between the detection limit (3σ) of 0.225 and 80 μg l−1. The R.S.D.% (n = 10) for a solution containing 50 μg l−1 of Bi was 0.85%. The tolerance of the system to interferences was evaluated by investigating the effect of the following ions: Cu2+, Co2+, Ni2+, Fe3+, Cd2+, Pb2+, Hg2+, Zn2+, and Mg2+. The most severe depression was caused by Hg2+, which at 60 mg l−1 caused a 5% depression on the signal. For the other cations, concentrations between 1000 and 10,000 mg l−1 could be tolerated. The system was applied to the determination of Bi in urine of patients under therapy with bismuth subcitrate. The recovery of spikes of 5 and 50 μg l−1 of Bi added to the samples prior to digestion with HNO3 and H2O2 was in satisfactory ranges from 95.0 to 101.0%. The concentrations of bismuth found in six selected samples using this procedure were in good agreement with those obtained by an alternative technique (ETAAS). Finally, the concentration of Bi determined in urine before and after 3 days of treatment were 1.94 ± 1.26 and 9.02 ± 5.82 μg l−1, respectively.  相似文献

Frutalin, a galactose-specific lectin used to detect specific tumour markers, is a protein with low expression level in breadfruit. In the present study, fed-batch fermentation in a stirred tank bioreactor was used as a strategy to enhance protein production by a recombinant Pichia pastoris KM71H. By using this process, the production of recombinant frutalin was 4-fold higher than the value obtained in shaker flasks batch assays. Supplementation of the fermentation medium with trace elements (Pichia trace minerals, PTM) was also evaluated in order to stimulate production of the recombinant protein. The addition of PTM to the minimum medium afforded a recombinant protein production of 13.4 mg L?1, which was 2.5-fold higher than that achieved from the culture medium without PTM supplementation. These results are significant as the development of strategies to improve the production of recombinant frutalin may broaden its application in cancer diagnosis.  相似文献
