Focus of this work was the development and characterization of a new immobilized enzyme reactor (IMER) containing human recombinant butyrylcholinesterase (rBChE) for the on-line kinetic characterization of specific, pseudo-irreversible and brain-targeted BChE inhibitors as potential drug candidates for Alzheimer's disease (AD). Specifically, a rBChE-IMER containing 0.99 U of covalently bound target enzyme was purposely developed and inserted into a HPLC system connected to a UV-vis detector. Selected reversible cholinesterase inhibitors, (-)-phenserine and (-)-cymserine analogues, were then kinetically characterized by rBChE-IMER, and by classical in solution assays and their carbamoylation and decarbamoylation constants were determined. The results support the elucidation of the potency, inhibition duration, mode of action and specific structure/activity relations of these agents and allow cross-validation of the two assay techniques.  相似文献

A new restricted access materials combined to molecularly imprinted polymers (RAM-MIP) with hydrophilic external layer were prepared and their applicability in p-acetaminophenol (AMP) selective recognition and release was evaluated. Glycidilmethacrylate (GMA) was used as pro-hydrophilic co-monomer, and GMA epoxide ring opening with perchloric acid was performed for the hydrophilic modification of polymeric surface. Morphological and hydrophilic properties by scanning electron microscopy and water content measurement were determined and recognition and selectivity properties of RAM-MIP were compared to the unmodified MIP. With RAM-MIP, both hydrophobic non-specific interactions between template or its analogues and polymeric matrices, and bovine serum albumin absorption were drastically reduced.  相似文献

Copper and germanium complexes of β-substituted nitrocorroles were reacted with 4-amino-4H-1,2,4-triazole to give the corresponding β-amino-β-nitro derivatives, in moderate to good yields. This is the first successful example of a vicarious nucleophilic substitution performed on corrole derivatives, because the same reaction carried out on silver complexes afforded the corresponding 6-azahemiporphycenes by way of corrole ring expansion. The first step of this work is related to the modification of a synthetic protocol for preparation of the β-substituted nitro corroles. The nitration reaction was carried out on a copper corrole using NaNO(2) as the primary source of NO(2)(-) coupled with AgNO(2) used as oxidant. By variation of the molar ratio of the reagents it was possible to direct the product distribution toward mono- and dinitro derivatives. The reaction between mono- and dinitro derivatives of (TtBuCorrCu) with 4-amino-4H-1,2,4-triazole gave good results, leading to the isolation of 2-(NH(2))-3-(NO(2))-TtBuCorrCu and 2,18-(NH(2))(2)-3,17-(NO(2))(2)-TtBuCorrCu in moderate yields. To elucidate factors that influence the reaction, and to highlight the different behavior observed for different metal complex substrates, the electrochemistry of three copper complexes, TtBuPCorrCu, (NO(2))TtBuPCorrCu, and (NO(2))(2)TtBuPCorrCu, was studied by cyclic voltammetry and thin-layer UV-visible spectroelectrochemistry. The nitro groups on (NO(2))(x)TtBuPCorrCu are highly electron-withdrawing, which leads not only to a substantial positive shift of all redox potentials but also to a unique redox behavior and UV-vis spectrum of the singly reduced product as compared to the parent compound, TtBuPCorrCu. Finally, the amination reaction was carried out on a Ge(IV) nitrocorrolate, giving in good yield the 2-amino-3-nitroderivative, which was structurally characterized by single crystal X-ray crystallography.  相似文献

Organic functionalization of carbon nanotube sidewalls is a tool of primary importance in material science and nanotechnology, equally from a fundamental and an applicative point of view. 1 , 2 Here, an efficient and versatile approach for the organic/organometallic functionalization of single‐walled carbon nanotubes (SWCNTs) capable of imparting multimodality to these fundamental nanostructures, is described. Our strategy takes advantage of well‐established Cu‐mediated acetylene‐azide coupling (CuAAC) reactions applied to phenylazido‐functionalized SWCNTs for their convenient homo‐/heterodecoration with a number of organic/organometallic frameworks, or mixtures thereof, bearing terminal acetylene pendant arms. Phenylazido‐decorated SWCNTs were prepared by chemoselective arylation of the CNT sidewalls with diazonium salts under mild conditions, and subsequently used for the copper‐mediated cycloaddition protocol in the presence of terminal acetylenes. The latter reaction was performed in one step by using either single acetylene derivatives or equimolar mixtures of terminal alkynes bearing either similar functional groups (masked with orthogonally cleavable protecting groups) or easily distinguishable functionalities (on the basis of complementary analytical/spectroscopic techniques). All materials and intermediates were characterized with respect to their most relevant aspects/properties by TEM microscopy, thermogravimetric analysis coupled with MS analysis of volatiles (TG‐MS), elemental analysis, cyclic voltammetry (CV), Raman and UV/Vis spectroscopy. The functional loading and related chemical grafting of both primary amino‐ and ferrocene‐decorated SWCNTs were spectroscopically (UV/Vis, Kaiser test) and electrochemically (CV) determined, respectively.  相似文献

Biopolymeric microparticles were prepared by rapid expansion of high pressure CO2-chitosan (Chi) solution in sodium bis-(2-ethyl hexyl) sulfosuccinate (AOT) solution. At pressures higher than 2 MPa, ultrafine particles were formed while under this value, wires were obtained. The formation of Chi/AOT complex was confirmed by Fourier Transform Infrared (FTIR) Spectroscopy, whereas scanning electron microscopy was used to characterize the morphology, size and shape of the particles. The FTIR spectrum proved the interaction between the sulfonate groups of AOT and the amino groups of Chi. Microparticles are quasi-spherical in wet conditions and irregular after freeze drying, presenting a rough surface with many pores. Lyophilized hydrophobic microparticles were used to remove phenol and o-cresol from aqueous solution, and the adsorption process showed a maximum efficiency in the 7–8 pH range. The uptake of phenol and o-cresol increased with the amount of particles and decreased with increasing the pollutant concentration. The adsorption occurred rapidly in the first 60–120 minutes, and leveled off thereafter.   相似文献

This work evaluates the biodegradation of N 4-acetylsulfapyridine (AcSPY) and N 4-acetylsulfamethazine (AcSMZ), metabolites of two of the most commonly used sulfonamides (SAs) in human and veterinary medicine, respectively. Aerobic transformation in effluent wastewater was simulated using aerated fixed-bed bioreactors. No visible changes in concentration were observed in the AcSMZ reactor after 90 days, whereas AcSPY was fully degraded after 32 days of experiment. It was also demonstrated that AcSPY transformed back to its parent compound sulfapyridine (SPY). The environmental presence of these two metabolites in wastewater effluent had been previously investigated and confirmed, together with three more SA acetylated metabolites and their corresponding parent compounds, in 18 different wastewater treatment plants in Hesse (Germany). Sulfamethoxazole (SMX) and SPY were the two SAs detected most frequently (90% and 89% of the samples, respectively) and in the highest concentrations (682 ng L−1 for SMX and 532 ng L−1 for SPY). To conclude, hazard quotients were calculated whenever toxicity data were available. None of the SAs studied posed an environmental risk.  相似文献

Here we present an ultra-performance liquid chromatography–mass spectrometry (UPLC–MS) method for extracellular measurements of known and unexpected metabolites in parallel. The method was developed by testing 86 metabolites, including amino acids, organic acids, sugars, purines, pyrimidines, vitamins, and nucleosides, that can be resolved by combining chromatographic and m/z dimensions. Subsequently, a targeted quantitative method was developed for 80 metabolites. The presented method combines a UPLC approach using hydrophilic interaction liquid chromatography (HILIC) and MS detection achieved by a hybrid quadrupole–time of flight (Q–ToF) mass spectrometer. The optimal setup was achieved by evaluating reproducibility and repeatability of the analytical platforms using pooled quality control samples to minimize the drift in instrumental performance over time. Then, the method was validated by analyzing extracellular metabolites from acute lymphoblastic leukemia cell lines (MOLT-4 and CCRF-CEM) treated with direct (A-769662) and indirect (AICAR) AMP activated kinase (AMPK) activators, monitoring uptake and secretion of the targeted compound over time. This analysis pointed towards a perturbed purine and pyrimidine catabolism upon AICAR treatment. Our data suggest that the method presented can be used for qualitative and quantitative analysis of extracellular metabolites and it is suitable for routine applications such as in vitro drug screening.  相似文献

This paper presents two procedures for the determination of four selective serotonin reuptake inhibitors (citalopram, paroxetine, fluoxetine, and sertraline) and one metabolite (norfluoxetine) in sewage sludge utilizing three-phase hollow fiber liquid-phase microextraction (HF-LPME). First, direct HF-LPME was used for extraction, clean-up, and preconcentration. The pharmaceuticals were extracted from slurry samples into an organic phase and then back-extracted into an aqueous phase in the lumen of the hollow fiber. Second, a procedure combining pressurized hot water extraction and HF-LPME for clean-up and preconcentration was developed for the same analytes and matrix. The extracts were subsequently analyzed by liquid chromatography-mass spectrometry. For direct HF-LPME, limits of detection were between 1 and 12 ng g(-1) (dry weight) and the relative standard deviation (RSD) values were 3-12%. For the second method, limits of detection were approximately 6 ng g(-1) for all the compounds and RSD values were 8-12%. The methods were validated by comparison of results for the same samples. Sewage sludge from a Swedish wastewater treatment plant was analyzed by both methods; average concentrations were similar for citalopram, paroxetine, and fluoxetine with values of approximately 530, 40, and 200 ng g(-1) , respectively.  相似文献

A QuEChERS method has been developed for the determination of 14 organochlorine pesticides in 14 soils from different Portuguese regions with wide range composition. The extracts were analysed by GC-ECD (where GC-ECD is gas chromatography-electron-capture detector) and confirmed by GC-MS/MS (where MS/MS is tandem mass spectrometry). The organic matter content is a key factor in the process efficiency. An optimization was carried out according to soils organic carbon level, divided in two groups: HS (organic carbon >2.3%) and LS (organic carbon <2.3%). The method was validated through linearity, recovery, precision and accuracy studies. The quantification was carried out using a matrix-matched calibration to minimize the existence of the matrix effect. Acceptable recoveries were obtained (70-120%) with a relative standard deviation of ≤16% for the three levels of contamination. The ranges of the limits of detection and of the limits of quantification in soils HS were from 3.42 to 23.77 μg kg(-1) and from 11.41 to 79.23 μg kg(-1), respectively. For LS soils, the limits of detection ranged from 6.11 to 14.78 μg kg(-1) and the limits of quantification from 20.37 to 49.27 μg kg(-1) . In the 14 collected soil samples only one showed a residue of dieldrin (45.36 μg kg(-1) ) above the limit of quantification. This methodology combines the advantages of QuEChERS, GC-ECD detection and GC-MS/MS confirmation producing a very rapid, sensitive and reliable procedure which can be applied in routine analytical laboratories.  相似文献
