Chemical compatibility of two drugs, namely, etamsylate and fluconazole was studied with lactose as excipient, employing differential scanning calorimetry (DSC) and X-ray diffraction (XRD) techniques. The DSC patterns recorded for the mixtures of both the drugs with the common excipient (lactose) indicated that fluconazole as well as etamsylate were incompatible with lactose at high temperatures. X-ray diffraction patterns recorded for pure drugs and lactose and the mixtures of individual drugs with lactose prepared at room temperature by intimate grinding of the components revealed incompatibility of both the drugs with lactose also at room temperature. The ring-closing metathesis reactions (RCM) of six standard diene substrates leading to five-, six-, or seven-membered carbo- or heterocycles were investigated under controlled microwave irradiation. RCM protocols were performed with standard Grubbs type II and a cationic ruthenium allenylidene catalyst in neat and ionic liquid-doped methylene chloride under sealed vessel conditions. Very rapid conversions (15 s) were achieved utilizing 0.5 mol % Grubbs II catalyst under microwave conditions. Careful comparison studies indicate that the observed rate enhancements are not the result of a nonthermal microwave effect.  相似文献

Morphological and thermodynamic transitions in drugs as well as their amorphous and crystalline content in the solid state have been distinguished by thermal analytical techniques, which include dielectric analysis (DEA), differential scanning calorimetry (DSC), and macro-photomicrography. These techniques were used successfully to establish a structure versus property relationship with the United States Pharmacopeia standard set of active pharmaceutical ingredient (API) drugs. A distinguishing method is the DSC determination of the amorphous and crystalline content which is based on the fusion properties of the specific drug and its recrystallization. The DSC technique to determine the crystalline and amorphous content is based on a series of heat and cool cycles to evaluate the drugs ability to recrystallize. To enhance the amorphous portion, the API is heated above its melting temperature and cooled with liquid nitrogen to ?120 °C (153 K). Alternatively a sample is program heated and cooled by DSC at a rate of 10 °C min?1. DEA measures the crystalline solid and amorphous liquid API electrical ionic conductivity. The DEA ionic conductivity is repeatable and differentiates the solid crystalline drug with a low conductivity level (10?2 pS cm?1) and a high conductivity level associated with the amorphous liquid (10pS cm?1). The DSC sets the analytical transition temperature range from melting to recrystallization. However, analysis of the DEA ionic conductivity cycle establishes the quantitative amorphous and crystalline content in the solid state at frequencies of 0.10–1.00 Hz and to greater than 30 °C below the melting transition as the peak melting temperature. This describes the “activation energy method.” An Arrhenius plot, log ionic conductivity versus reciprocal temperature (K?1), of the pre-melt DEA transition yields frequency dependent activation energy (E a, J mol?1) for the complex charging in the solid state. The amorphous content is inversely proportional to the E a where the E a for the crystalline form is higher and lower for the amorphous form with a standard deviation of ±2%. There was a good agreement between the DSC crystalline melting, recrystallization, and the solid state DEA conductivity method with relevant microscopic evaluation. An alternate technique to determine amorphous and crystalline content has been established for the drugs of interest based on an obvious amorphous and crystalline state identified by macro-photomicrography and compared to the conductivity variations. This second “empirical method” correlates well with the “activation energy” method.  相似文献

Over the last thirty years, the world has seen HIV circulate the globe, affecting 33 million people to date and killing 2 million people a year. The disease has affected developed and developing countries alike, and in the U.S., remains one of the top ten leading causes of death. Many regions of the world are highly impacted by this disease, including sub-Saharan Africa, South and South-East Asia, and Eastern Europe. Fortunately, multilateral, global efforts, along with successful developments in diagnostic tools and anti-retroviral drugs (ARVs) have successfully curbed the spread of HIV over the last ten years. In spite of this fact, access to HIV treatment and preventive healthcare is varying and limited in developing countries. A lack of healthcare infrastructure, financial support, and healthcare workers are some logistical factors that are responsible. HIV stigmatization, discrimination, and inadequate education pose additional social challenges that are hindering countries from advancing in HIV prevention. This review focuses on current technological tools that are used for HIV diagnosis and ongoing research that is aimed at addressing the conditions in low-resource settings. Recent developments in microfluidic applications and mobile health technologies are promising approaches to building a compact, portable, and robust device that can provide information-rich, real-time diagnoses. We also discuss the role that governments, healthcare workers, and even researchers can play in order to increase the acceptance of newly introduced devices and treatments in rural communities.  相似文献

The synthesis of a generation 5 (G5) poly(amidoamine) (PAMAM) dendrimer platform having cyclooctyne ligands that were subsequently be used for a copper-free Huisgen 1,3-dipolar cycloaddition (click reaction) with azido modified methotrexate is described. The G5 PAMAM dendrimer was first partially (70%) acetylated and then coupled with 20 cyclooctyne ligands through amide bonds. The remaining primary amine groups on the dendrimer surface were neutralized by acetylation. The platform was then ‘clicked’ with different numbers (5, 10, and 17) of γ-azido functionalized methotrexate. The copper-free click reactions were stoichiometric with excellent yields.  相似文献

Summary N-(orthovanillidene)L-alanine (OVAlaH2), N-(salicylidene)L-valine (SValH2), N-(orthovanillidene)L-valine (OVValH2), N-(orthovanillidene)L-tyrosine (OVTyrH3) and N-(salicylidene)L-glutamic acid (SGluH3) react with LaIII, PrIII, NdIII and SmIII to yield new chiral complexes. This paper discusses the development of a stability-indicating reversed-phase LC method for analysis of cholecalciferol as the bulk drug and in formulations. The mobile phase was acetonitrile–methanol–water 50:50:2 (v/v). The calibration plot for the drug was linear in the range 0.4–10 μg mL−1. The method was accurate and precise with limits of detection and quantitation of 64 and 215 ng, respectively. Mean recovery was 100.71%. The method was used for analysis of cholecalciferol in pharmaceutical formulations in the presence of its degradation products and commonly used excipients.

The different approaches that have been taken in the development of analytical methods for the determination of nitrate within the field are reviewed. The emphasis has been placed on providing a critical appraisal of the chemistry that underpins current commercial systems and the need to remove the dependence on heavy metal and concentrated acid components. The search for more environmentally acceptable and user friendly systems has long been pursued and the present communication seeks to explore the recent development in portable testing technologies and how they might evolve in the future. In particular, the role of electrochemical techniques in the latter are investigated and their potential application compared and contrasted with the more traditional wet chemical detection strategies.  相似文献

Cubic copper ferrite thin films, obtained by rf sputtering on quartz and subsequent post-annealing and quenching, show a large coercivity of about 300–600 Oe. Stress measurements using X-ray diffraction show high value of stress of about 400–1000 MPa. Both the stress and coercivity are found to increase with the decrease of the thickness of the films. There appears to be a contribution of the stress to the coercivity of the films, in the in-plane M–H loops.  相似文献
