In a re-analysis of nucleon form factors in perturbative quantum chromodynamics we examine the
-dependence of the nucleon quark distribution amplitude by means of some model distribution amplitudes. We point out some problems that arise in the determination of the
-evolution of the distribution amplitude. Our suggestions to cure these problems are discussed and resulting nucleon quark distribution amplitudes are presented. А protocol for the efficient and selective recovery of human ferritin heavy chain (FTH1) expressed intracellularly in Hansenula polymorpha was developed. It was based on electropermeabilisation and an increase in the cell wall porosity by pulsed electric field (PEF) treatment and subsequent incubation with a low concentration of a lytic enzyme. Irreversible plasma membrane permeabilisation was induced by applying rectangular electric pulses in the flow mode. The electrical treatment itself did not cause the release of the recombinant protein but induced the sensitisation of H. polymorpha cells to the lytic enzyme. Consequently, the subsequent incubation of the permeabilised cells with lyticase led to the recovery of approximately 90% of the recombinant protein, with a purification factor of 1.8. A similar efficiency was obtained by using the industrial lytic enzyme Glucanex. The released FTH1 appears in the form of an oligomer with a molecular mass of approximately 480 kDa, which is able to bind iron. The possibility for scaling the proposed protocol is discussed. 相似文献

Transition‐metal carbene complexes and their reactivities are a key topic of chemistry. They are an integral part of researches in catalysis, organic synthesis, coordination chemistry, and numerous other areas. In this context, we report the synthesis of a low‐valent bis(η5:η1‐(di‐p‐tolyl)‐pentafulvene)niobium chloride. Owing to the π‐η5:σ‐η1 coordination mode of the pentafulvenes and the resulting high nucleophilic character of the exocyclic carbon atom of the ligand, the bis(η5:η1‐pentafulvene)niobium complex is able to achieve the umpolung of a coordinated vinyl unit and the resulting formation of the first η5:η1 cyclic niobium Schrock carbene complex. This new synthetic route is, in comparison to classical α‐hydrogen elimination reactions or thermolysis of diazo compounds, completely unprecedented. The reactivity of the cyclic carbene function and the remaining fulvene ligand is demonstrated by double N?H bond activation of primary amines to niobium imido complexes. 相似文献

The compartmentalization of chemical reactions is an essential principle of life that provides a major source of innovation for the development of novel approaches in biocatalysis. To implement spatially controlled biotransformations, rapid manufacturing methods are needed for the production of biocatalysts that can be applied in flow systems. Whereas three‐dimensional (3D) printing techniques offer high‐throughput manufacturing capability, they are usually not compatible with the delicate nature of enzymes, which call for physiological processing parameters. We herein demonstrate the utility of thermostable enzymes in the generation of biocatalytic agarose‐based inks for a simple temperature‐controlled 3D printing process. As examples we utilized an esterase and an alcohol dehydrogenase from thermophilic organisms as well as a decarboxylase that was thermostabilized by directed protein evolution. We used the resulting 3D‐printed parts for a continuous, two‐step sequential biotransformation in a fluidic setup. 相似文献

The perturbation expansion for a general class of many-fermion systems with a nonnested, nonspherical Fermi surface is renormalized to all orders. In the limit as the infrared cutoff is removed, the counterterms converge to a finite limit which is differentiable in the band structure. The map from the renormalized to the bare band structure is shown to be locally injective. A new classification of graphs as overlapping or nonoverlapping is given, and improved power counting bounds are derived from it. They imply that the only subgraphs that can generater factorials in ther th order of the renormalized perturbation series are indeed the ladder graphs and thus give a precise sense to the statement that ladders are the most divergent diagrams. Our results apply directly to the Hubbard model at any filling except for half-filling. Let f be a 1-periodic, smooth function and $\hat f_k(k\in {\rm Z})$ its Fourier coefficients. The reconstruction of f from a given sequence of perturbed values of $\hat f_k$ is an ill-posed problem. Applying Tihonov’s regularization method, this problem can be solved in a stable manner. The efficiency of this method is closely related to the optimal choice of the smoothing parameter. In this paper we compute an optimal smoothing parameter by the method of generalized cross-validation. The related algorithm is tested in some numerical examples. Multiple internal reflection spectroscopy (MIRS) was applied to analyse atomic bonds at deep (500 m) interfaces of directly bonded Si/Si wafer pairs. It is shown that under the conditions used the polarized spectra contain information only about the interface layer a few nanometers thick. Examples are given of analysing the Si-H- and SiO-H vibration modes and bands of undissociated water in interfaces of bonded hydrophobic and hydrophilic wafer pairs, respectively, after annealing at temperatures between 200 and 1100 °C. Knowledge bases for real-world domains such as mechanical engineering require expressive and efficient representation and processing tools. We pursue a declarative-compilative approach to knowledge engineering.While Horn logic (as implemented in PROLOG) is well-suited for representing relational clauses, other kinds of declarative knowledge call for hybrid extensions: functional dependencies and higher-order knowledge should be modeled directly. Forward (bottom-up) reasoning should be integrated with backward (top-down) reasoning. Constraint propagation should be used wherever possible instead of search-intensive resolution. Taxonomic knowledge should be classified into an intuitive subsumption hierarchy.Our LISP-based tools provide directtranslators of these declarative representations into abstract machines such as an extended Warren Abstract Machine (WAM) and specialized inference engines that are interfaced to each other. More importantly, we provide source-to-sourcetransformers between various knowledge types, both for user convenience and machine efficiency.These formalisms with their translators and transformers have been developed as part of CoLab, acompilationlaboratory for studying what we call, respectively, vertical and horizontal compilation of knowledge, as well as for exploring the synergeticcolaboration of the knowledge representation formalisms.A case study in the realm of mechanical engineering has been an important driving force behind the development of CoLab.
