Ion-extraction by a crown ether with a photoresponsive carboxylate cap.

A new photoresponsive crown ether (I) with an azobenzene-4-oxyacetate function was synthesized. The photo-isomerized cis-(1) extracted K⁺ ion more selectively than trans-(I) and the thermal cis-trans isomerization was suppressed by added metal ions.     

Redox-switched crown ethers monocrown-biscrown interconversion coupled with redox of a thiol-disulfide couple

To give a redox-switch function to crown ethers, 4′-mercaptomono-benzo-15-crown-5 (CrSH) and its oxidized disulfide form (CrSSCr) were synthesized. The solvent extraction proved that the ion affinity of CrSH is generally greater than that of CrSSCr.     

Asymmetric weak decay of polarized hypernuclei

We measured the asymmetric weak decay of the polarized _Λ⁵He hypernuclei. The polarization was derived by the observed asymmetry of the mesonic decay pions for the first time. The asymmetry parameter of the nonmesonic decay has been evaluated using the obtained polarization and the asymmetry of the decay protons. The positive asymmetry parameter contradicts theoretical prediction based on meson exchange model. The present data indicates the limitation of the model in ΛN weak interaction, in particular short range phenomena such the nonmesonic decay of hypernuclei.     

Directional solidification of InxGa1−xAs

We have investigated a constitutional supercooling and segregation phenomena in In_xGa_1−xAs crystals unidirectionally solidified in a vertical system. The constitutional supercooling generates characteristic fluctuations of composition along the growth direction and this can be explained by a free nucleation ahead from the growth interface. The macroscopic compositional profiles of the grown crystals suggest that a transport of solute is mainly dominated by the diffusion. Such a growth mode is partly attributed to the difference in density between InAs and GaAs.     

Assay of Angiotensin I-converting Enzyme-inhibiting Activity Based on the Detection of 3-Hydroxybutyrate with Water-soluble Tetrazolium Salt.

A newly synthesized substrate, 3-hydroxybutyrylglycyl-glycyl-glycine (3HB-GGG), was applied to the assay of ACE-inhibiting activity to overcome the smaller selectivity and sensitivity of the conventional method. In this study, an ACE-inhibiting assay was improved by the use of a water-soluble tetrazolium salt, 4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulfonate sodium salt (WST-1), for the detection of 3-hydroxybutyrate, derived from 3HB-GGG. The optimized conditions were as follows: 0.333 mM NAD(+), 0.333 mM WST-1, 0.1 mM EDTA, 0.633 U ml(-1) diaphorase, and 0.700 U ml(-1) 3-hydroxybutyrate dehydrogenase. The developed assay was efficiently applicable to evaluate the ACE-inhibiting activity of practical ACE inhibitors.     

Practical Antibody Recruiting by Metabolic Labeling with Caged Glycans

We propose a de novo glycan display approach that combines metabolic labeling and a glycan-caging strategy as a facile editing method for cell-surface glycans. This method enables the introduction of antigen glycans onto cancer cells to induce immune responses through antibody recruiting. The caging strategy prevents the capture of α-rhamnose (an antigen glycan) by endogenous antibodies during the introduction of the glycan to the targeted cell surface, and subsequent uncaging successfully induces immune responses. Therefore, this study proposes a practical method for editing the cell-surface glycocalyx under promiscuous conditions, such as those in vivo, which paves the way for the development of glycan function analysis and regulation.     

Promotion effects of optical antipodes on the formation of helical fibrils: chiral perfluorinated gelators

A chiral gelator, RR- or SS-N,N'-diperfluorooctanoyl-1,2-diaminocyclohexane, gelated racemic 2-butanol. The gel was most stable at the racemic mixture, its stability lowered with the increase in the optical purity of the gelator. Notably, characteristic helically coiled fibrils were formed in the narrow region of enantiomer excess (ee = 0.2-0.4). Promotion effects of the antipodal enantiomers are proposed.     

Synthesis and optical resolution of a new flavin with axial chirality and redox-dependent racemization

A new flavin (3-methyl-10-(2′-phenyl)phenylisoalloxazine) with restricted rotation about the C(1′)?N(10) single bond was synthesized and optically resoluted for the first time. (+)- Isomer (100% enantiomeric purity) did not racemize at ambient conditions, whereas it racemized invariably when it was reduced (by four different methods). The kinetic studies proved that the novel racemization mode is attributed to conversion of the “planar” oxidized form to the “bent” reduced form.
