Photooxidation of cyclopentadiene has been carried out in methanol using white light of LED lamp, rose bengal as photo initiator, and compressed air at 0?°C. Under conditions of [thiourea] ? [cyclopentadiene], the consumption of thiourea follows a pseudo-first-order reaction kinetics with half life of 75?±?10?min; corr. coeff. r?=?0.989. Slow addition of the monomer and maintaining excess thiourea concentration in reaction mass improves the yield. cis-3,5-Dihydroxy-1-cyclopentene is acetylated without isolation to obtain cis-3,5-Diacetoxy-1-cyclopentene of high purity (>99%) with overall isolated yield of 30%. Desymmetrization of the diacetate to (1R,4S)-4-hydroxycyclopent-2-en-1-yl acetate has been carried out via enzymatic transesterification with methanol in methyl tert-butyl ether (MTBE) at 5?°C using Novozym-435®. The enantiomerically pure monoacetate (e.e. >99%) was obtained in 95% isolated yield. The recovered enzyme was reused for more than 10 times without loss in yield and selectivity. The entire protocol does not require purification of final product by chromatography.  相似文献

Considering the importance of benzothiazepine pharmacophore, an attempt was carried out to synthesize novel 1,5-benzothiazepine derivatives using polyethylene glycol-400 (PEG-400)-mediated pathways. Initially, different chalcones were synthesized and then subjected to a cyclization step with benzothiazepine in the presence of bleaching clay and PEG-400. PEG-400-mediated synthesis resulted in a yield of more than 95% in less than an hour of reaction time. Synthesized compounds 2a–2j were investigated for their in vitro cytotoxic activity. Moreover, the same compounds were subjected to systematic in silico screening for the identification of target proteins such as human adenosine kinase, glycogen synthase kinase-3β, and human mitogen-activated protein kinase 1. The compounds showed promising results in cytotoxicity assays; among the tested compounds, 2c showed the most potent cytotoxic activity in the liver cancer cell line Hep G-2, with an IC50 of 3.29 ± 0.15 µM, whereas the standard drug IC50 was 4.68 ± 0.17 µM. In the prostate cancer cell line DU-145, the compounds displayed IC50 ranges of 15.42 ± 0.16 to 41.34 ± 0.12 µM, while the standard drug had an IC50 of 21.96 ± 0.15 µM. In terms of structural insights, the halogenated phenyl substitution on the second position of benzothiazepine was found to significantly improve the biological activity. This characteristic feature is supported by the binding patterns on the selected target proteins in docking simulations. In this study, 1,5-benzothiazepines have been identified as potential anticancer agents which can be further exploited for the development of more potent derivatives.  相似文献

The outstanding capability of two-dimensional gel electrophoresis in separating all types of proteins basically depends on the efficiency of sample preparation. Sample preparation is one of the most critical steps in two-dimensional gel electrophoresis. Unfortunately, due to severe solubility, resolution of protein on gel is usually hampered, and thus, analysis remains a difficult task. However, technically several problems are generally encountered during protein extraction and isoelectric focusing. In the present investigation, we emphasized on evaluation and comparison of six different protein solubilization methods intended for resolving and analyzing silkworm hemolymph proteins by two-dimensional gel electrophoresis. Our findings revealed that the buffer composition of 8 M urea, 4 % 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate, 40 mM Tris base, 65 mM dithiothreitol, and 0.2 % ampholyte can effectively solubilize and yields maximum protein spots.  相似文献

Simple, sensitive, rapid and cost effective extraction spectrophotometric methods are described for the assay of mebeverine hydrochloride (MBH) in bulk samples and pharmaceutical formulations. These two methods (Bromophenol blue and Erichrome Black‐T) are based on the formation of chloroform soluble ion‐pair complexes of MBH with Bromophenol blue (BPB) and with Erichrome Black‐T (EBT), to form yellow and pink colored chromogen in a Glycine‐HCl buffer of pH 2.4 (BPB) and in a KCl‐HCl buffer of pH 1.4 (EBT) with absorbance maximum at 416 nm and at 524 nm for BPB and EBT respectively. The calibration graph is found to linear over 0.2–20 μg/mL (BPB) and 0.2–20 μg/mL (EBT), with molar absorptivity values of 1.8295 × 104 1 moL?1 cm?1 and 1.5896 × 104 1 moL?1 cm?1, respectively. The LOD (Limit of Detection) were found to be 0.090 μg/mL and 0.084 μg/mL and LOQ (Limit of Quantification) were 0.2997 μg/mL and 0.2730 μg/mL for the BPB and EBT method, respectively. The results of analysis for the two methods have been validated statistically and by recovery studies. The results are compared with those obtained with reported method. The proposed methods are simple, sensitive, accurate and suitable for quality control applications.  相似文献

Asymmetric dihydroxylation of vinyl furanosides 1-6 by use of OsO4, AD-mix-α® and β® is described yielding the corresponding hexofuranose sugars. Vinyl furanosides 2 and 3, with an ester group at C-3, and vinyl manno furanoside 5 on asymmetric dihydroxylation with AD-mix α® exhibited high R diastereoselectivity at C-5. Reversal in diastereoselectivity at C-5 was observed for the 3-deoxy vinyl furanoside 6 giving furanosaccharide 6S with the S configuration at C-5.  相似文献

Data envelopment analysis (DEA) allows us to evaluate the relative efficiency of each of a set of decision-making units (DMUs). However, the methodology does not permit us to identify specific sources of inefficiency because DEA views the DMU as a “black box” that consumes a mix of inputs and produces a mix of outputs. Sugarcane (Saccharum spp.) is a dynamic C4 polyploidy grass used as a major source of sucrose and an alternative for ethanol, food, and energy. Despite growing scientific interest, various sucrose metabolism regulatory aspects have been limited. Biochemical and gene expression studies were conducted on developmental stages, 240–420 days of planting (DAP) in mature leaves of three high and three low sucrose sugarcane cultivars. Sucrose synthase (SS) and sucrose phosphate synthase (SPS) activities were found to be remarkably higher at 240–360 DAP but decrease at 420 DAP. Twofold increases of SS activity was estimated at 240–360 DAP while SPS activity trend was found to be lower than the SS activity. In comparing SS and SPS activities with the brix of respective DAP, results show that these activities are significant and positively correlated with ‘r’ values of 0.69 and 0.68 for SS and SPS, respectively. However, the soluble acid invertase (SAI) and neutral invertase (NI) activities were found to decrease significantly with the maturity of cultivars, negatively correlating with brix at ‘r’ values 0.83 and 0.89 for SAI and NI, respectively. The antioxidant enzyme activity was modulated similar to the invertases activity. Of the six genes, ESAS 11 and 23 associated with sucrose accumulation and ESTS 34 and 41 associated with sugar transport in sugarcane were differentially expressed among the selected high and low sugarcane cultivars. Hence, these findings reinforce the selection of diverse sugarcane cultivars for gene expression studies targeting to quantitative traits and candidate marker determination.  相似文献
