Isolation and Stereostructures of Dolastatin G and Nordolastatin G, Cytotoxic 35-Membered Cyclodepsipeptides from the Japanese Sea Hare Dolabella auricularia

A bioassay-directed fractionation of the cytotoxic constituents of the Japanese sea hare Dolabella auricularia resulted in the isolation of two 35-membered depsipeptides dolastatin G (1) and nordolastatin G (2), which showed cytotoxicity against HeLa S(3) cells with IC(50) values of 1.0 and 5.3 &mgr;g/mL, respectively. The gross structures of these substances were established by spectroscopic analysis including 2D NMR techniques. The absolute stereostructure of 1 was determined by chiral HPLC analysis of amino acid components obtained from acid hydrolysis of 1 and by the enantioselective syntheses of two degradation products arising from polyketide portions. Nordolastatin G (2) is a congener that has the same absolute stereochemistry as that of 1.

Syntheses of polyurethanes derived from oximes and their photodegradation

Polyoxime-urethanes have been synthesized from dioximes and diisocyanates, and their structures have been ascertained by elemental analysis and infrared spectroscopy. They can be represented by the general formula: These polymer films were photolyzed by using a high-pressure 100-W mercury lamp, and the rate of decomposition of the N? O bond was determined by use of ultraviolet and infrared spectra, or comparing the photochemical behavior with that of corresponding model compounds. Upon irradiating these polymers in the presence or absence of sensitizers or a hydrogen donor, there were obtained photolyzed polymers of low molecular weight, of which the molecular weights were estimated by observing the viscosity changes. To deduce the mechanism of photodegradation of polyoxime-urethane, photolysis of benzophenone oxime-phenyl urethane was investigated as a model compound.     

Gold electrodes from recordable CDs for mercury quantification by flow injection analysis

The development of a new methodology for the construction of very efficient flow cells for mercury detection by potentiometric stripping analysis, employing the thin gold layer of recordable CDs as working electrode is reported. This new source of electrodes (CDtrodes) show very attractive performance, similar to that obtained with commercial gold electrodes, with superior versatility. The low cost of this new source of "gold electrodes" allows a frequent replacement of the electrode, avoiding cumbersome clean-up treatments. Various experimental parameters have been optimized to yield low detection limits (0.25 ng/mL of mercury for 5 min deposition at 0.3 V) and good precision (standard deviation of 1.9% was obtained for 15 repetitive measurements using 10 ng/mL of mercury). Standard curves were found to be linear over the range of 0.5-100 microg L(-1) of mercury. The flow cells developed were used for the quantification of mercury in oceanic and tap water.     

Neighboring effect of thiol group in acyl transfer reaction on a polymer with a lipoic acid structure

Reductive S-monoacylation of lipoamide with some carboxylic acids and tri-n-butylphosphine was carried out in acetonitrile to obtain the corresponding S-monoacylated compounds in excellent yield. This method was applied to the preparation of S-monoacylated dihydrolipoyl polymer with a neighboring thiol group. The acyl transfer from the polymer to cyclohexylamine was enhanced as much as 64 times by adding silver perchlorate.

The proposed structures of patellamides B and C, cytotoxic lipophilic cyclic peptides from a marine tunicate, have been revised on the basis of the spectral data of their partial hydrolysates, and their revised structures have been synthetically confirmed by the use of diphenyl phosphorazidate(DPPA) and diethyl phosphorocyanidate(DEPC).     

Synthesis of a novel silicon-bridged [2.2]metacyclophan-9-ene and its photolytic transannular reaction

A novel [2.2]metacyclophane in which two benzene rings are linked together with a carbon-carbon double bond and a disilane unit was prepared. Photolysis of the cyclophane in the presence of oxygen afforded the 4,5-dihydro-4,5-disilapyrene derivative via a transannular dehydrogenation reaction.     

Conversion of endohedral <Superscript>133</Superscript>Xe-fullerene to endohedral <Superscript>133</Superscript>Xe-fullerenol to be used in nuclear medicine

Summary Hydrophilic endohedral ¹³³Xe-fullerenols, [¹³³Xe@C₆₀(OH)_xand ¹³³Xe@C₇₀(OH)_x], were synthesized from hydrophobic endohedral ¹³³Xe-fullerenes. The yield of endohedral ¹³³Xe-fullerenols extracted in water was about 40% and 23% for C₆₀and C₇₀, respectively. The products stored in 0.9% NaCl solution at 20 °C were stable enough to be used in nuclear medicine.
