The identification and quantification of modified peptides are critical for the functional characterization of post-translational protein modifications (PTMs) to elucidate their biological function. Nowadays, quantitative mass spectrometry coupled with various bioinformatic pipelines has been successfully used for the determination of a wide range of PTMs. However, direct characterization of low abundant protein PTMs in bottom-up proteomic workflow remains challenging. Here, we present the synthesis and evaluation of tandem mass spectrometry tags (TMT) which are introduced via click-chemistry into peptides bearing alkyne handles. The fragmentation properties of the two mass tags were validated and used for screening in a model system and analysis of AMPylated proteins. The presented tags provide a valuable tool for diagnostic peak generation to increase confidence in the identification of modified peptides and potentially for direct peptide-PTM quantification from various experimental conditions. 相似文献

Conclusions The temperature transition of a solvated cross-linked polymer (the SGK-7 carboxyl cation-exchanger, which contains iron(III) ions) from the solid state to a state characterized by intense intramolecular motions was detected and studied by Mössbauer spectroscopy. Triggering of two types of motions was demonstrated: high-frequency vibrations of atoms with a higher amplitude (manifested by a sharp decrease in the probability of the Mössbauer effect) and slower (conformational) motions (manifested by a change in the shape of the spectrum). Potential dependences of the angle of contact between perfluorodecaline C10F18 and copper, copper(I) sulfide, and copper telluride Cu4Te3 in 0.1 M CH3COONa are measured. The data are in good agreement with the metal hardness measured in 1 M KOH. The uncharged-surface potentials (USP) for copper, copper(I) sulfide, and copper telluride equal 0.05–0.07, –0.02 to –0.04, and 0.05–0.07 V (NHE) in 0.1 M CH3COONa. Studying the effect of Br– ions shows that USP for copper and Cu2S shift in, respectively, the negative and positive directions with an increase in [Br–]. For copper telluride, USP shifts in the positive direction at low Br– concentrations, and at NaBr concentrations in excess of 0.04 M, in the negative direction. 相似文献
