Angiotensin II (Ang II) is an octapeptidic hormone, which plays an important role in the mechanisms of blood pressure control. In this work, extensive molecular dynamics (MD) simulations have been carried out on this peptide, both in aqueous and in dimethyl sulfoxide (DMSO) environments. Experimentally proposed models for the structure of angiotensin II in both environments are not consensual and the results obtained have provided some further insight about the structural properties of this hormone. In these simulations, the N-terminus of Ang II in the aqueous environment has been associated with a considerable larger flexibility than the correspondent C-terminus, but this was not found in the case of the DMSO environment. This is consistent with the assumption that the biological activity of Ang II is associated with its C-terminal residues embedded in a hydrophobic environment of its AT1 receptor. Other features detected in DMSO environment were an H(His6 imidazole)-O(Phe8 carboxylate) hydrogen bond and a salt-bridge structure involving the Asp1 and Arg2 side chains. An additional important conformational feature is the spatial proximity between Tyr4 and His6 in both water and DMSO environments. This molecular feature may trigger the interest for the synthetic chemists to apply rational design for the synthesis of novel AT1 antagonists.  相似文献

We report here new chemical evidence for the generation of radical molecular ions of compounds with a conjugated pi-system (polyene) in ESI and HR-MALDI mass spectrometry. The oxidation potential of the neutral polyenes was calculated by cyclic-voltammetry and the results compared with those previously published for other complex conjugated compounds that have also been shown to form M*+ in ESI-MS. This study clearly demonstrates the correlation between the oxidation potential and the formation of the M*+ for the polyenes studied.  相似文献

A heat-stable enzyme was isolated from the cellulase complex of a thermophilic strain of the micromyceteThielavia terrestris. The purified enzyme exhibited both endoglucanase and xylanase activities and had a mol mass of 69,000 Daltons and an isoelectric point of 6.4. When the cells were grown at 48°C, the initial activity of the purified enzyme using carboxymethylcellulose as a substrate was 150 nkat/mg and the Michaelis constant was 6.6 g/L. The heat stability of the enzyme was high, losing only 20% of the initial activity after a 6-h incubation at 65 °C. When cultures were grown on microcrystalline cellulose and xylose was added after 48 h of growth, endoglucanase and xylanase activities were more than doubled. Similar increases in these activities were observed by growing the cultures on straw.  相似文献

The most promising method capable of providing accurate measurements of the first and second normal-stress differences in shear flows at shear rates typical of polymer processing is the so-called hole-pressure method, but its use has not been as widespread as would be expected, namely due to the experimental difficulties associated with performing such experiments accurately. In this work, we use a small-scale modular slit die to assess the practical utility of the method for in-line monitoring of polymer melt flow. We provide a quantitative analysis of intrinsic error sources and use state-of-the-art data acquisition tools to minimize errors associated with pressure transducers. Our results demonstrate that the method can be used to accurately measure the viscosity and first normal-stress difference in melts but probably not the second normal-stress difference because the intrinsic errors are too high, even when the influence of all the potential error sources is minimized or eliminated.  相似文献

It is well-known that when a single-degree-of-freedom (sdof) system is excited by a continuous motion of the foundation, the force transmissibility, relating the force transmitted to the foundation to the applied force, equals the displacement transmissibility. Recent developments in the generalization of the transmissibility to multiple-degree-of-freedom (mdof) systems have shown that similar simple and direct relations between both types of transmissibility do not appear naturally from the definitions, as happens in the sdof case.  相似文献

In this study, the thermo-oxidative stability of two new phosphorylated derivatives of cardol, a compound from the cashew (Anacardium occidentale L.) industry waste CNSL (cashew nutshell liquid), were evaluated. The antioxidant capacity of these new molecules upon two mineral oils, NH10 and NH20 were also studied by thermogravimetric analysis (TG/DTG), observing the onset and offset temperatures variation. The results showed that both MP and DP Cardol increased considerably the onset and offset temperatures of NH10 and NH20 oils.  相似文献
